摘    要:目的:研究五鹤续断(WHDA)注射液对D-半乳糖所致小鼠衰老模型的抗衰老作用及其机制。方法:昆明种小鼠(雌雄各半)48只,随机分为对照组、模型组、阳性WHDA组、7.2g/kgWnDA组、3.6g/kgWHDA组及1.8g/kgWHDA组,每组8只。腹腔注射D-半乳糖造模,Morris水迷宫测量小鼠学习和记忆能力。检测各组小鼠皮肤羟脯氨酸、脑组织MDA、LP、SOD及GSH-Px含量,采用双抗体夹心ELISA法检测小鼠血清中IL-2及IL-6含量。结果:Morris水迷宫实验结果表明WHDA各组潜伏期与模型组相比较明显减少(P〈0.05),模型组小鼠穿越平台的次数比其它各组小鼠少(P〈0.05)。脑组织氧化性指标(SOD、MAD、LP及GSH.Px)检测,对照组及WHDA各组MAD与LP含量低于模型组(P〈0.05)。对照组及WHDA各组SOD与GSH-Px的活性明显高于模型组(P〈0.05)。注射过D-半乳糖小鼠皮肤羟脯氨酸含量都明显低于对照组(P〈0.05),WHDA组小鼠皮肤中羟脯氨酸的含量明显高于模型组(P〈0.05)。WHDA组小鼠血清中IL-2、IL-6含量显著高于对照组和模型组(P〈0.05),模型组小鼠血清中的IL-2、IL-6比对照组要低(P〈0.05)。结论:WHDA能改善D.半乳糖所致的衰老小鼠的学习记忆能力,消除其体内自由基,增强机体免疫能力,具有较好的抗衰老作用。

Abstract:Objective: To study the effect of Wu-He Dipsacus asper ( WHDA ), Traditional Chinese Medicine, injection on mice aging model induced by D-galactose. Methods: Forty-eight Kunming mice (24 male and 24 female) were randomly divided into control group, mod el group, positive control group, 7.2 g/kg WHDA group, 3.6 g/kg WHDA group and 1.8 g/kg WHDA goup with eight in each group. The model was induced through injecting D-galactose into peritoneal cavity and Morris water maze was used to detect the learning and cognitive abil- ity of mice. The skin hydroxyproline, brain tissue malondialdehyde(MDA ), lipofuscin(LP), superoxide dismutase(SOD ) and glutathione per oxidase(GSH-Px) levels of mice were detected; the IL-2 and IL-6 levels in serum of mice were detected by using double antibody sandwich ELISA method, Results: Each WHDA group was significantly reduced in latency period compared with the model group during Morris water maze test ( P 〈 0.05) and the number of mice in model group through the platform was less than other mice in each group ( P 〈 0.05). The levels of MAD and LP of the control group and each WHDA group were less than model group in the detection of heart, brain tissue oxidation index (SOD, MAD, LP and GSH-Px, P 〈0.05). The activity of SOD and GSH-Px in the control group and each WHDA group was signifi- cantly higher than that in the model group ( P 〈 0.05). The skin hydroxyproline content of mice which had been injected with D-galactose was significantly lower than that in the control group ( P 〈 0.05) and the skin hydroxyproline content of mice of WHDA group was significantly higher than that in the model group ( P 〈 0.05). The IL-2, IL-6 levels in serum of mice in WHDA group were significantly higher than those in the control group and the model group ( P 〈0.05) and the IL-2, IL-6 levels in serum of mice in the model group were lower than those in the control group ( P 〈 0.05). Conclusion: The effective constituents of WHDA have a variety of biological activity which can have a good ef fect on anti-aging by different ways, improving teaming and memory function, eliminating free radicals antioxidant, and enhancing the body immunity and other aspects.
