马铃薯StKUP12的表达及参与钾营养的功能研究

摘    要:旨在初步探究StKUP12的钾转运功能,为该基因在钾营养方面的进一步研究奠定基础。通过同源克隆得到StKUP12,对该基因进行生物信息学分析、表达分析以及钾营养缺陷型酵母互补试验。结果显示,StKUP12全长2 547 bp,编码848 个氨基酸,蛋白分子量93.9 kD,理论等电点6.59,预测含有K^+转运结构域和磷酸化位点。StKUP12蛋白与潘那利番茄(Solanum pennellii)同源性较高,达98.00%;与巴西橡胶树(Heava brasilliersis)同源性较低,为77.74%。表达分析显示,该基因在根、茎、叶、顶芽和芽眼中均有表达,同时受低钾、高盐、ABA和PEG胁迫的调控。酵母互补试验表明该转运体具有吸收外界K^+的功能。实验结果表明StKUP12属于钾离子转运体,具有高亲和性的钾离子吸收功能。

Abstract:The aim of this study is to explore the potassium transport function of StKUP12 and to lay a foundation for further research of StKUP12 involved in potassium nutrition.StKUP12 was cloned by homologous cloning,and analyzed by bioinformatics,expression level and complementary test of potassium deficient yeast.Results showed that the gene contained 2 547 bp,encoding 848 amino acids.Its protein molecular weight was 93.9 kD and theoretical isoelectric point was 6.59.It was predicted that there were K^+ transport domain and phosphorylation site.The StKUP12 protein had 98.00% homology with Solanum pennellii,and had 74.55% homology with Vitis vinfera.qRT-PCR analysis showed that the gene expressed in root,stem,leaf,apical shoot and bud eye,and this gene expression was regulated by low potassium,high salt,ABA and PEG.Yeast complementation test showed that the transporter absorbed K^+ from the medium.These results prove that StKUP12 belongs to potassium transporter and has potassium absorption function with high affinity.
