| 摘 要： | TNF是一种具有多种生物学作用的细胞因子，高纯度TNF的获得是进一步研究其作用机理和探讨防治方法的基础．对克隆后工程菌表达的纯化进行研究，经盐析。透析、离子交换等步骤，成功地制备了高纯度的rhTNF．经SDS－PAGE测定，分子量17kD，纯度＞95％，比活性为3．44×107U／mg，与国外同类产品相似．

| Abstract: | TNF is a cytokine with many biological activities. The highly-purified TNF is necessary for studying the effect of TNF and in searching for preventive methods against its action. Thehighly expressed recombinant organism and the purification of TNF were studied. They successfully produced the highly purified rhTNF following steps of salting-out, dialysis, and ion-exchangechromatography. The apparent molecular weight is 17 kD and its purity is more than 95% bySDS-PAGE with specific activity of 3.44×107 U/mg which is similar to rhTNF obtained fromSigma Chemical Company. |
