亚硝酸盐对培养心肌细胞周期的影响

摘    要:本文用流式细胞仪测定了NaNO2对体外培养的Wistar乳鼠心肌细胞周期的影响。结果表明,10^-6mol/L NaNO2引起S期细胞明显减少(P〈0.05);10^-8mol/L NaNO2对其 显著(P〉0.05)。应用「3H」TdR掺入法测定了NaNO2对心肌细胞增殖的作用。实验发现,10^-6mol/L NaNO2明显抑制细胞增殖,而10^-9-10^7mol/L的NaNO2则促进细胞增殖

Abstract:In our previous work we have demonstrated that sodium nitrite caused acute myocardial necrosis in rats in vivo. The direct effect of sodium nitrite on cell cycle in cultured neonatal rat heart cells and its mechanism were investigated with the following results-. (1) large doses of sodium nitrite (10-6mol/L) could decrease the percentage of S phase of myocytes (P<0.05) (2) The effects of sodium nitrite on 3H]TdR incorporation into DNA of myocytes were; small doses (10-9- 10-7mol/L) was stimulatory, while large dose (10-6mol/L) was inhibitive. (3) Sodium nitrite (10-8-10-6mol/L) could increase the melondialdehyde contents of myocytes. These results suggested that the changes of myocytes induced by sodium nitrite were relative to lipid peroxidation.
