土壤中PHA高产菌株的分离及鉴定

摘    要:从成都某炼油厂油罐和排污口附近土壤中分离到1株PHA(聚羟基烷酸)的高产菌株,最高含量为19.25%。根据形态特征观察、生理生化特征鉴定及16S rDNA序列比对,结果表明该菌株为蜡状芽胞杆菌。并对其发酵条件进行初步探索,确定培养的最适温度为37℃,最适pH值为7.0,为下一步构建基因工程菌和优化发酵条件提供了理论基础。

Abstract:A polyhydroxyalkanoate(PHA) high-yielding strain was isolated and purified from soil samples collected from neighboring site of oil storage tanks and the sewage outfall of a refinery in Chengdu,and the highest content of PHA was 19.25%(W/DW).Based on the morphological observation,physiological and biochemical characteristics and the result of sequences comparison and verification of 16S rDNA,the strain was identified as Bacillus cereus.Preliminary experiment of its fermentation conditions was confirmed that the optimal culture temperature and pH were 37 ℃,and 7.0.These had provided a theoretical basis for further study on the construction of building a recombinant engineering strain that could produce PHA and the optimization of the fermentation conditions.
