生长抑素对Hela细胞中Claudin-3和Claudin-4基因表达的调节作用

摘    要:目的:探讨生长抑素对Hela细胞的生长调控作用以及对claudin-3和claudin-4基因的表达调控。方法:通过Hela细胞株培养,并以浓度为10-6、10-8、10-10和10-12 M的生长抑素(SST)作用于Hela细胞,未经药物处理的细胞设为对照组。在处理后采用流式细胞仪检测Hela细胞的凋亡。并采用实时荧光定量PCR和Western blot分别检测claudin-3和claudin-4 m RNA和蛋白质表达量。结果:SST加入Hela细胞孵育12 h小时后,10-10 M、10-8 M和10-6 M浓度的SST对Hela细胞有显著性的诱导凋亡作用。不同浓度的SST作用于Hela细胞12 h后,claudin-3和claudin-4的m RNA和蛋白表达量都出现不同水平的增加。结论:在Hela细胞中SST可以促进claudin-3和claudin-4的基因表达,从而对宫颈癌的发展和扩散有抑制作用。

Abstract:Objective:To investigate the regulation of somatostatin on Hela cell growth as well as claudin-3 and claudin-4 gene expression.Methods:Hela cells were treated with 10-6, 10-8, 10-10 and 10-12 M somatostatin (SST), respectively. Untreated cells were used as control. Flow cytometry was used to detect the apoptosis of Hela cells. Realtime-PCR and Western blot were performed to assay the effect of SST on mRNA and protein expression of gene claudin-3 and claudin-4.Results:After treatment with SST by differential concentrations for 1 2 h, it was found that 10-10 M, 10-8 M and 10-6 M SST could induce a significant apoptosis in the Hela cells. Under different concentrations of SST, the mRNA and protein expression of claudin-3 and claudin-4 were up-regulated.Conclusion:In Hela cells, SST could induce claudin-3 and claudin-4 gene expression and inhibit development and diffuse of cervical cancer cells.
