Abstract:Compared with traditional chemical flocculants,microbial flocculants are safe,non-toxic,no secondary pollution,and have the potential development.Phanerochaeate chrysosporium can produce microbial flocculants,but there is a lack of optimization of nutrient conditions for producing flocculants.Using Kaolin and single factor method,the effects of carbon source,nitrogen source,carbon nitrogen ratio and inoculation amount on the yield and flocculation efficiency of the flocculant produced by Phanerochaete chrysosporium were investigated.Finally,the nutrient conditions of the flocculant produced by the bacteria were optimized.The results showed that the maximum amount of polysaccharide(281.3 mg/L)was produced when glucose was used as carbon source and the maximum biomass(4.35 g/L)was produced when starch was used as carbon source,and the flocculation efficiencies of flocculants produced by glucose and starch as carbon sources were 65%and 43%,respectively.When ammonium nitrate was used as nitrogen source,the amount of polysaccharide produced was the highest(163.61 mg/L),and the flocculant flocculation efficiency was 82.5%.When the C/N ratio was 60~100,it was beneficial to the production of flocculant,and when it was 90,the flocculation efficiency reached the maximum(80.5%).There was no significant difference in biomass when the inoculation amount was 0.25%~1%,when the inoculation amount was 1%,the amount of polysaccharide produced was 155.57 mg/L,and the flocculation efficiency reached the maximum(87.14%).To sum up,the best nutritional conditions were as follows:glucose as carbon source,ammonium nitrate as nitrogen source,C/N ratio of 90 and inoculation amount of 1%,which laid a foundation for large-scale production of flocculant.
