分泌抗加压素单克隆抗体的杂交瘤细胞株的建立(之一)

基于深入研究加压素在体内的作用的需要,特制备其单克隆抗体。以赖氨酸加压素为抗原免疫小鼠,取其脾细胞与小鼠骨髓瘤细胞融合,筛选出两株特异性高的杂交瘤细胞,产生的抗体对赖氨酸加压素有强的结合反应,对精氨酸压素有一定的交叉反应,对催产素基本无反应。鉴定证明,此抗体属IgG1亚类,为纯的、均一性的单克隆抗体。

HYBRID CELL LINES SECRETING MONOCLONAL ANTIBODIES AGAINST VASOPRESSIN

Vasopressin ( VP ) , a 9 -peptide hormone, is synthesized in the suprao-ptic and paraventrieular nuclei of the hypothalaraas and is transported through the hypothalamus-pituitary tract to the posterior pituitary where it is released into the blood. Beside its antidiuretic and pressor action, VP has a wide distribution in the central nervous system and may play an important role in brain function. Based on the importance of research of the inaction of VP, we have started the work on the preparation of hybrid cell liaes secreting monoclonal antibodies against VP.Synthetic 8 -lysine VP was conjugated to bovine thyroglobulin by car-bodimide and used as an antigen. After initial immunisation with coapled antigen in complete Freund's adjuvant,the BALB/c mice were Tboostered with the same antigen without adjuvant directly into the spleen 4 days before fusion. The spleen cells of the immunized mice and the SP2/0 myeloma cells were fused by using PEG-1000; Two specific hybrid cell lines secreting McAbs against VP ( AVM1 and AVM2 ) were obtained. By solid phase ELISA, it was demonstrated that the McAbs had high reactivity with 8-ly-sine VP and some cross-reactivity with 8-arginine VP, but not with oxyto-cin. The double immumodiffusion test showed tEat these McAbs all belonged to the mouse IgG1 subclass. Biological experiments with these McAbs are being carried out.
