Histamine H2 receptors and cyclic AMP in brain |

Abstract: | The intravenous injection of histamine to 2–3 day old chicks resulted in a rapid and marked increase in the cyclic AMP content of the cerebral hemispheres that had been removed and frozen within 0.5s using a freeze-blowing technique. This response was not antagonized by pretreatment of the chicks with the histamine H1-receptor antagonists mepyramine and diphenhydramine but was blocked by the H2-receptor antagonists burimamide and metiamide. Parallel experiments on slices of chick cerebral hemispheres demonstrated that the H1 antagonists only produced a weak and non-competitive antagonism of the effects of histamine on cyclic AMP production. On the other hand the H2 antagonists at low concentrations competitively blocked the histamine response. It is suggested that increased cyclic AMP formation in chick cerebral hemispheres can be mediated through stimulation of histamine H2-receptors. |
