PF127修饰的石墨烯复合材料的制备及其载药系统研究

摘    要:实验制备负载阿霉素的PF127修饰的还原态石墨烯复合材料,并对其性能进行研究和评价。其方法是首先制备由弗朗尼克F127非共价功能化修饰的还原氧化石墨烯材料,记为PF127/GN,然后将该复合物对药物阿霉素进行负载,记为PF127/GN/DOX,并与未经阿霉素负载的纳米药物载体(PF127/GN)进行对比分析。观察其药物释放行为,细胞内递送过程和细胞毒性。研究发现,PF127/GN粒子大小为80μm左右,且粒子分布较均匀,厚度约增加至9.646μm;利用阿霉素对纳米载体进行负载之后,负载效果随药物浓度的增加而不断提高,负载在氧化石墨烯表面的阿霉素的释放行为可通过改变体系的pH值进行调节。负载了阿霉素的纳米药物载体能够发挥良好的释放作用,具有相对较强的生物毒性。因此经过阿霉素负载的PF127修饰的还原态石墨烯复合材料性能良好,具有广阔的应用前景。

Preparation and study of drug delivery system of pluronic PF127 modified graphene nanosheet composite materials

Abstract:The PF127 /GN nanohybrid was made to be capable of effectively encapsulating doxorubicin( DOX),and modified with graphene nanosheet as PF127 /GN. The drug release behavior and the delivery process in the cell and cell toxicity were assayed. Result showed that the particle size of PF127 /GN was about 80 μm,the distribution was relatively uniform,and the thickness increased to about 9. 646 μm. The load effect increased with the increase of drug concentration after doxorubicine equipped nanomedicine,the release of doxorubicine could improve by adjusting the pH values of the external environment. The modified pluronic PF127 graphene nanosheet is stable dispersion under physiological conditions and has good biocompatibility.
