鱼类精子携带的外源基因导入

将鲤鱼（或泥鳅）精子在保存液内与人生长激素（ｈＧＨ）重组质粒ＤＮＡ保温，再与鲤（或泥鳅）卵受精。由此发育的鱼苗经ＤＮＡ分子杂交和ＰＣＲ检测证明，３３．３％（或３７．０％）的个体带ｈＧＨ基因，其拷贝数在４—１５０／细胞之间。在一定条件下，精子可作为携带外源基因的载体，通过受精作用产生转基因鱼。

INTRODUCTION OF FOREIGN GENE CARRIED BY SPERMS

Mature eggs from carp (Cyprinus carpio L.)and loach (Misgurnus anguillicaudatusCantor)were inseminated with the same species sperms which had been incubated withrecombinant plasmid DNA containing human GH gene in a solution containing 2.9%glucose,1%sodium citrate,0.2% sodium bicarbonate,0.03% potassium chloride and30mg/ml bovine albumin.Individuals of 33.3% carp fry and 37.0% loach fry showed tocarry the foreign gene with 4-150 copies per cell by dot blot hybridization and PCR analysis.The studies indicated that sperm could be a vector to carry and introduce foreign gene intofish eggs in the course of fertilization.
