牛樟芝多糖对HepG2细胞酒精性氧化损伤的保护作用

摘    要:研究了不同剂量(100、200和400μg/mL)的牛樟芝粗多糖(CP)和醇提物后的水提物(WEE)对酒精诱导的HepG2细胞氧化损伤的保护作用。研究结果表明:与模型组比较,各剂量组的CP和200、400μg/mL的WEE均能极显著提高HepG2细胞的细胞活力。100μg/mL的CP和WEE均能极显著降低细胞培养液的ALT水平;200和400μg/mL的CP和WEE均能显著降低细胞培养液的ALT、AST水平,同时提高胞内的CAT活力;200和400μg/mL的WEE及400μg/mL CP能明显提高胞内的SOD活力。此外,WEE各剂量组和400μg/mL CP中的胞内ROS水平显著下降。CP中含有甘露糖、鼠李糖、葡萄糖、半乳糖、岩藻糖5种单糖,摩尔比为1:0.1622:6.651:2.646:0.3929。WEE和CP能提高细胞的抗氧化应激能力,降低胞内ROS,对酒精诱导的HepG2细胞氧化损伤起到明显的保护作用,提示多糖是牛樟芝解酒保肝的重要活性成分之一。

Protective effect of Taiwanofungus camphoratus polysaccharides on alcohol-induced oxidative injury in HepG2 cells

Abstract:The protective effect of different doses (100, 200 and 400μg/mL) of water extract after extraction by ethanol (WEE) and crude polysaccharide (CP) fromTaiwanofungus camphoratus on alcohol-induced HepG2 cell injury was investigated. Compared with the model experimental group, various doses of CP and 200 and 400μg/mL of WEE could significantly improve cellular viability. 100μg/mL CP and WEE could significantly reduce the alanine aminotransferase (ALT) level in culture medium. 200 and 400μg/mL of CP and WEE could notably reduce ALT and aspartate aminotransferase (AST) level in culture medium, and simultaneously improve the intracellular catalase (CAT) activity. 200 and 400μg/mL WEE and 400μg/mL CP could remarkably increase the intracellular superoxide dismutase (SOD) activity. Treatment with various doses of WEE and 400μg/mL of CP conspicuously reduced the intracellular reactive oxygen species (ROS) level. CP was consisted of 5 monosaccharides, including mannose, rhamnose, glucose, galactose and fucose with the molar ratio of 1:0.1622:6.651:2.646:0.3929. Results proved that WEE and CP could increase the cellular response to oxidative stress and reduce the intracellular ROS level to protect HepG2 cell from oxidative injury induced by alcohol prominently, suggesting that CP was one of the main active ingredients responsible for drunkenness-alleviating and liver-protecting effects.
