兔急性肠道缺血再灌注损伤模型的建立

摘    要:目的:建立小肠急性缺血再灌注损伤模型,确定合适的肠系膜上动脉阻断时间.方法:将70只新西兰兔随机按不同的肠系膜缺血时间(0、15、30、45、60min)分为A、B、C、D、E5组,每组14只,取8只用于恢复血供2h后留取各组兔下腔静脉血标本及肠组织,检测血清中MDA含量的变化,光镜下观察小肠组织形态学变化并对小肠黏膜损伤程度进行评分.另6只用于术后24h、 48h及72h生存率的观察.结果:A、B、C组的术后生存率均>83.3%.C、D、E组的MDA含量及肠黏膜损伤评分与A组比较,差异均有显著性(F=12.13~280.24,p<0.01).结论:肠系膜缺血30min,再灌注2h是建立兔小肠急性再灌注损伤的合适时间.

Abstract:Objective: To establish the model of acute small intestinal isehemia-reperfusion injury and to determine the propor blocking time of superior mesenteric arteries (SMA). Methods: Seventy New Zealand rabbits were randomly divided into five groups - A,B, C, D, and E, based on the mesentery isehemia time (0, 15, 30, 45 and 60min), with 14 rabbits in each group. 2 hours after recovery of blood supply, eight of the rabbits in each group were chosen, by detecting MDA content changes in serum, to observe the histomorphological changes of small intestine under light microscope and to score on the basis of the injury degree of small intestine mucosa. The rest six rabbits in each group were used for postoperative observation of the survival rates at 24h, 48h and 72h. Results:The The survival rates were all higher than 83.3% in group A, B and C. Compared with group A, there was a significant increase in group C, D and E on MDA content and score of injury of intestinal mucosa (F= 12.13~280.24,p<0.01). Conclusion: 30 min of mesentery ischemia and 2h of reperfusion are the proper time to establish the acute small intestinal reperfusion injury in rabbits.
