摘    要:研究β-酪啡肽-7(β-casomorphin-7,β-CM-7)对链脲佐菌素诱导的糖尿病大鼠肾损伤的保护作用.结果显示:(1)β-酪啡肽-7降低糖尿病大鼠的采食量、饮水量和空腹血糖值,提高糖尿病大鼠的体重,但是差异不显著(P>0.05).(2)β-酪啡肽-7显著降低糖尿病大鼠尿糖、尿蛋白、血清肌酐、血清尿素氮、肾脏指数和血清中转化生长因子-β1(TGF-β1)的含量;Masson染色结果显示,β-酪啡肽-7治疗显著降低糖尿病大鼠肾脏的纤维化程度.(3)RT-RCR结果显示,β-酪啡肽-7显著降低糖尿病大鼠肾脏TGF-β1、Ⅰ型和Ⅲ型胶原蛋白基因的表达.Western印迹显示,β-酪啡肽-7显著降低糖尿病大鼠肾脏Ⅰ型和Ⅲ型胶原蛋白的表达.上述结果表明,β-酪啡肽-7能够减缓糖尿病大鼠肾脏功能和肾脏纤维化程度,其机制可能与降低TGF-β1含量、减少胶原蛋白在肾脏的沉积有关.

Abstract:The protective effects of β-casomorphin-7(β-CM-7)on renal injury of streptozotocin-induced diabetic rats were explored. The results showed that (1) β-CM-7 reduced the food intake, water intake, fasting blood glucose and increased the body weight, but the difference was not significant (P>0.05). (2) β-CM-7 reduced significantly urine glucose, urinary protein, serum creatinine, serum urea nitrogen, kidney index, and serum transforming growth facto β1(TGF-β1) in diabetic rats. Masson staining revealed that β-CM-7 treatment significantly reduced the degree of fibrosis in the kidney of diabetic rats. (3) RT-RCR showed that β-CM-7 reduced the expression of TGF-β1, collagenⅠ and collagen Ⅲ in kidney of diabetic rats. Western blot showed that β-CM -7 reduced the expression of collagen Ⅰ and collagen Ⅲ in the kidney of diabetic rats. These results indicated that administration of β-CM-7 attenuates renal injury and renal interstitial fibrosis caused by diabetes. The protective effects may result in inhibition of TGF-β1 expression and reduction of collagen deposition in the kidney.
