Summary Synchronized transformed and reverse-transformed (by 10−3 M B2cAMP) CHO-K1 cells, growing adherent to plastic, are characterized by means of geometric and densitometric parameters at the level of both the entire cell and of the nuclei at various time intervals after selective mitotic detachment. Transformed and reverse-transformed cells triple-stained with Feulgen, Napthol Yellow S, and periodic acid-Schiff appeared very similar in terms of integrated optical density (IOD), related to either polysaccharides, protein, or DNA amount. On the other hand, a shift from a polygonal to a spindle-shaped morphology is accompanied by a significant decrease in both form factor and average optical density (AOD) of intact cell and nuclei, which are the most conspicuous measured changes caused by B2cAMP, in addition to a lengthening of the cell cycle duration. In both control and treated cells, important and parallel cell-cycle-dependent modulations of geometric and densitometric parameters are also observed, for both the cytoplasmic (i.e., cell morphometry) and DNA space (i.e., nuclear morphometry). Specifically, the modulation in nuclear morphometry during G1, S, G2, andM phases confirms previous findings on synchronized HeLa cells. The optical density threshold-dependence of geometric parameters shows that, while becoming fusiform, the cytoplasm of reverse-transformed cells had a particularly low optical density precisely in the polar area. Utilization of such an approach in the development of anobjective morphological classification of all cell lines grown as monolayers “in vitro” is also discussed.  相似文献

The effect of long-term (77 day) exposure of carp Cyprinus carpio to low concentrations of waterborne iron (1 mg Fe-dextran l−1) on accumulation and cellular distribution of iron in the liver was assessed using Perl's staining and histological observations and morphometric measurements. Accumulation of iron in the liver occurred after 14 days of exposure, when the majority of the iron was found in the sinusoidal endothelium with lower amounts in the cytoplasm and nuclei of hepatocytes. Upon further treatment the iron was predominantly distributed in the cytoplasm of the hepatocytes either as granules or diffusive iron and in macrophages.  相似文献

Morphometric variation among populations of Sarotherodon melanotheron melanotheron across its natural range (Mahin, epe, Lekki, Apese, Kuramo, Lagos, Iyagbe, Ologe, Badagry, Yewa – Nigeria; Be Lagoon – Togo; Idenau – Cameroon, Aheme – Benin; Portonovo – Benin) were studied to assess the level of differentiation of one of the populations that is currently isolated from the rest, the Apese Lagoon population. S. m. melanotheron showed subtle differentiation from some of the other lagoon populations. However, the pattern of variation did not reflect the level of geographical barriers or geographical distance among populations. The characters that form the basis of the differentiation observed were mostly those related to feeding, vision and respiration.  相似文献

The nutrient‐rich organic waste generated by ants may affect plant reproductive success directly by enhancing fruit production but also indirectly, by affecting floral traits related with pollinator attraction. Understanding how these soil‐nutrient hot spots influence floral phenotype is relevant to plant–pollination interactions. We experimentally evaluated whether the addition of organic waste from refuse dumps of the leaf‐cutting ant Acromyrmex lobicornis (Hymenoptera: Formicidae: Attini) alters floral traits associated with pollinator attraction in Eschscholzia californica (Ranunculales: Papaveraceae), an entomophilous herb. We analysed flower shape and size using geometric morphometric techniques in plants with and without the addition of refuse‐dumps soil, under greenhouse conditions. We also measured the duration of flowering season, days with new flowers, flower production and floral display size. Plants growing in refuse‐dumps soil showed higher flower shape diversity than those in control soil. Moreover, plants in refuse‐dumps soil showed bigger flower and floral display size, longer flowering season, higher number of flowering days and flower production. As all these variables may potentially increase pollinator visits, plants in refuse‐dumps soil might increase their fitness through enhanced attraction. Our work describes how organic waste from ant nests may enhance floral traits involved in floral attraction, illustrating a novel way of how ants may indirectly benefit plants.  相似文献

An extensive review of the genus Matrona is presented based on mitochondrial (COI) and nuclear (ITS) sequences from 150 samples which cover all the known taxa of this genus. The separation of two main clades (oreades group: M. oreades, M. corephaea and M. taoi; basilaris group: M. basilaris, M. nigripectus, M. cyanoptera, M. japonica and M. annina) is strongly supported. The classification of all traditional recognized species is confirmed. The Hainan population separates very well from mainland M. basilaris populations, which is also confirmed by geometric morphometric analysis of wing shape. Given the implications of the molecular analysis the genus Matrona is grouped into two subgenera: subgen. Matrona (type species M. basilaris) and D ivortia subgen. nov. (type species M. oreades). A new species, M . ( M .) mazu sp. nov. , from Hainan is described. Brief taxonomic notes on the nine recognized species of the genus are given. 东亚中更新世古人类在头骨、下颌骨、牙齿等解剖部位表现出不同程度的形态多样性,中期成员代表为直立人,而晚期成员的演化地位具有较大争议。为进一步了解东亚中更新世古人类内部的形态变异特点和为东亚中更新世晚期古人类分类提供依据,本文使用微分同胚的表面匹配(Diffeomorphic Surface Matching, DSM)和形态测量图(Morphometric map)对下颌第二臼齿(M2)釉质-齿质连接面的形状和齿冠侧面釉质厚度分布模式进行了量化分析。结果显示:1)东亚中更新世古人类与晚期人属成员(尼安德特人和现代人)存在较明显的形态差别;2)该时段晚期的东亚古人类相对中期直立人在侧面釉质厚度分布规律上具有独特性,并在釉质-齿质连接面的三维形状上与晚期人属成员更加接近。本文在以往对东亚中更新世古人类牙齿内外结构单个性状研究的基础上,使用三维形态测量方法进一步量化了M2釉质-齿质连接面三维形状和侧面釉质厚度分布模式两项重要特征的变异特点,这对未来该时段同类型牙齿的形态鉴定以及解决东亚中更新世晚期古人类的分类地位具有一定意义。  相似文献

Tenualosa ilisha, T. toli and Hilsa kelee are three Indian shad species of the family Clupeidae. In past, relationships among these shads have been explored in few morphological studies but still remain poorly understood. In this study, 120 specimens of three shad species were collected from the wild and a truss network system of 13 landmarks was interconnected to yield 77 size-adjusted characters. Discriminant function analysis (DFA) produced 13 discriminating variables in delineating the three species. The landmark based morphometric distance between origin of dorsal fin and posterior end of eye was the most important character in discriminating the three species. Three prominent characters extracted from DFA (distance between posterior end of operculum and posterior end of eye, distance between origin of dorsal fin and posterior end of eye and distance between insertion of pelvic fin and end of operculum) were used as traditional morphometric characters and their ratio to standard length were found significant in differentiating the three shad species. The present study strongly supports the efficiency of landmark-based morphological characters generated from digital images in discriminating the species and can be used as a rapid diagnostic tool in recognizing them.  相似文献
