A crude inhibitor for pancreatic lipase was extracted from soybean seeds. The lipase activity decreased curvilinearly with an increase in inhibitor concentration. At a low inhibitor concentration, enhanced inhibition was observed by the co-existence of protein such as bovine serum albumin in the reaction mixture. The lipase activity was inhibited immediately after the addition of inhibitor which did not cause the significant destraction of substrate emulsion. The lipase activities of Aspergillus niger, Rhizopus delemar and castor bean seeds were also inhibited. The inhibition was observed when various oil substrates such as soybean oil, linseed oil, olive oil emulsions and Ediol were used, and the extent of inhibition varied among them. Column chromatography of inhibitor on Sephadex G–100 showed that the molecular weight of a main peak of inhibitor was estimated as about 80,000. 相似文献

Related or distant species of cultivated cs are a large pool of many desirable genes. Gene transfer from these species through conventional breeding is difficult owing to post- and pre-zygotic sexual incompatibilities. Somatic hybridization via protoplast fusion is a possible alternative for gene transfer from these species to cultivated crops. Since the early days of somatic hybridization many intergeneric somatic hybrids have been developed through symmetric fusion, asymmetric fusion and microfusion. Somatic hybrids are mainly selected by using markers such as specific media or fusion parents with special features, biochemical mutants, antibiotic resistance and complementation strategy. The hybridity of the regenerants is determined based on morphological, cytological and molecular analysis. The inheritance patterns of nuclear and cytoplasmic genomes in the somatic hybrids are diverse. Nuclear DNA from both fusion parents co-exists congruously in some hybrids with translocation and rearrangement of chromosomes, but spontaneous elimination of chromosomes from either or both fusion parents has been observed very often. In asymmetric fusion, chromosome elimination is an important issue that is a complicated process influenced by many factors, such as irradiation dose, phylogenetic relatedness, ploidy level of fusion parent and regenerants. As for chloroplast genome, uniparental segregation is mainly detected, though co-existence is also reported in some cases. The mitochondrial genome, in contrast to chloroplast, undergoes recombination and very frequent rearrangements. Somatic cell fusion has potential applications for crop genetic improvement by overcoming sexual incompatibility or reproductive barriers, and by realizing novel combinations of nuclear and/or cytoplasmic genomes. 相似文献

Agriculture faces challenges to fulfil the rising food demand due to shortage of arable land and various environmental stressors. Traditional farming technologies help in fulfilling food demand but they are harmful to humans and environmental sustainability. The food production along with agro-environmental sustainability could be achieved by encouraging farmers to use agro-environmental sustainable products such as biofertilizers and biopesticides consisting of live microbes or plant extract instead of chemical-based inputs. The eco-friendly formulations play a significant role in plant growth promotion, crop yield and repairing degraded soil texture and fertility sustainably. Mineral solubilizing microbes that provide vital nutrients like phosphorus, potassium, zinc and selenium are essential for plant growth and development and could be developed as biofertilizers. These microbes could be plant associated (rhizospheric, endophytic and phyllospheric) or inhabit the bulk soil and diverse extreme habitats. Mineral solubilizing microbes from soil, extreme environments, surface and internal parts of the plant belong to diverse phyla such as Ascomycota, Actinobacteria, Basidiomycota, Bacteroidetes, Chlorobi, Cyanobacteria, Chlorophyta, Euryarchaeota, Firmicutes, Gemmatimonadetes, Mucoromycota, Proteobacteria and Tenericutes. Mineral solubilizing microbes (MSMs) directly or indirectly stimulate plant growth and development either by releasing plant growth regulators; solubilizing phosphorus, potassium, zinc, selenium and silicon; biological nitrogen fixation and production of siderophores, ammonia, hydrogen cyanide, hydrolytic enzymes and bioactive compound/secondary metabolites. Biofertilizer developed using mineral solubilizing microbes is an eco-friendly solution to the sustainable food production system in many countries worldwide. The present review deals with the biodiversity of mineral solubilizing microbes, and potential roles in crop improvement and soil well-being for agricultural sustainability. 相似文献

The widespread production of cash crops can result in the decline of forests, peatlands, rice fields and local community land. Such unwanted land‐use and land‐cover (LULC) change can lead to decreased carbon stocks, diminished biodiversity, displaced communities and reduced local food production. In this study, we analysed to what extent four main commodities, namely, palm oil, pulpwood, rice and rubber, can be produced in North and East Kalimantan in Indonesia without such unwanted LULC change. We investigated the technical potential of four measures to mitigate unwanted LULC change between 2008 and 2020 under low, medium and high scenarios, referring to the intensities of the mitigation measures compared with those implemented in 2008. These measures are related to land sparing through (i) the improvements of yields, (ii) chain efficiencies, (iii) chain integration and (iv) the steering of any expansion of these commodities to suitable and available underutilised (potentially degraded) lands. Our analyses resulted in a land‐sparing potential of 0.4–1.2 Mha (i.e. 24–62% of the total land demand of the commodities) between 2008 and 2020, depending on the land‐use projection of the four commodities and the scenario for implementing the mitigation measures. Additional expansion on underutilised land is the most important mitigation measure (45–62% of the total potential), followed by yield improvements as the second most important mitigation measure (32–46% of the total potential). Our study shows that reconciling the production of palm oil, pulpwood, rice and rubber with the maintenance of existing agricultural lands, forests and peatlands is technically possible only (i) under a scenario of limited agricultural expansion, (ii) if responsible land zoning is applied and enforced and (iii) if the yields and chain efficiencies are strongly improved. 相似文献

Mediterranean environments are characterised by cool wet winters and hot dry summers. While native vegetation in Mediterranean-climatic zones usually comprises a mixture of perennial and annual plants, agricultural development in the Mediterranean-climatic region of Australia has led to the clearing of the perennial vegetation and its replacement with annual crops and pastures. In the Mediterranean environments of southern Australia this has led to secondary (dryland) salinisation. In order to slow land degradation, perennial trees and pasture species are being reintroduced to increase the productivity of the saline areas. The annual crops and pastures that form the backbone of dryland farming systems in the Mediterranean-climatic zone of Australia are grown during the cool wet winter months on incoming rainfall and mature during spring and early summer as temperatures and rates of evaporation rise and rainfall decreases. Thus, crop and pasture growth is usually curtailed by terminal drought. Where available, supplementary irrigation in spring can lead to significant increases in yield and water use efficiency. In order to sustain production of annual crops in Mediterranean environments, both agronomic and genetic options have been employed. An analysis of the yield increases of wheat in Mediterranean-climatic regions shows that there has generally been an increase in the yields over the past decades, albeit at a lower rate than in more temperate regions. Approximately half of this increase can be attributed to agronomic improvements and half to genetic improvements. The agronomic improvements that have been utilised to sustain the increased yields include earlier planting to more closely match crop growth to rainfall distribution, use of fertilisers to increase early growth, minimum tillage to enable earlier planting and increase plant transpiration at the expense of soil evaporation, rotations to reduce weed control and disease incidence, and use of herbicides, insecticides and fungicides to reduce losses from weeds, insects and disease. Genetic improvements include changing the phenological development to better match the rainfall, increased early vigour, deeper rooting, osmotic adjustment, increased transpiration efficiency and improved assimilate storage and remobilisation. Mediterranean environments that are subjected annually to terminal drought can be both environmentally and economically sustainable, but to maximise plant water use efficiency while maintaining crop productivity requires an understanding of the interaction between genotypes, environment and management. 相似文献

Phage-displayed synthetic antibody libraries were built on a single human framework by introducing synthetic diversity at solvent-exposed positions within the heavy chain complementarity-determining regions (CDRs). The design strategy of mimicking natural diversity using tailored codons had been validated previously with scFv libraries, which produced antibodies that bound to antigen, murine vascular endothelial growth factor (mVEGF), with affinities in the 100nM range. To improve library performance, we constructed monovalent and bivalent antigen-binding fragment (Fab) libraries, and explored different CDR-H3 diversities by varying the amino acid composition and CDR length. A Fab with sub-nanomolar affinity for mVEGF was obtained from a library with CDR-H3 diversity designed to contain all 20 naturally occurring amino acids. We then expanded the library by increasing the variability of CDR-H3 length and using tailored codons that mimicked the amino acid composition of natural CDR-H3 sequences. The library was tested against a panel of 13 protein antigens and high-affinity Fabs were obtained for most antigens. Furthermore, the heavy chain of an anti-mVEGF clone was recombined with a library of light chain CDRs, and the affinity was improved from low nanomolar to low picomolar. The results demonstrated that high-affinity human antibodies can be generated from libraries with completely synthetic CDRs displayed on a single scaffold. 相似文献

The results of the application of the principles of Life Cycle Inventory (LCI) to the production paths of Oxo-alcohols and
LAB from kerosene are shown here, taking into account the improvements made within our production technologies.
The case studies detailed in this study show the most relevant achievements, regarding our processes, in terms of environmental
quality and demonstrate that the correct application of LCI methodology is decisive to obtain considerable advantages, as
