激素对转基因雪莲毛状根植株再生及类黄酮产生的影响   总被引:2,自引:0,他引:2  
为了研究外源激素GA3和IAA对3个转基因新疆雪莲类黄酮高产毛状根系C17、C27、C46的植株再生及其总黄酮含量的影响,在培养基中添加不同浓度的GA3和IAA,结果发现,GA3浓度高于1.0 mg/L时,可诱导毛状根系产生不定芽,其中以GA3浓度为2.0 mg/L时,转基因毛状根系C17的不定芽再生率最高,可达82%。高压液相色谱以及紫外分光光度法测定结果表明,与未用激素处理的毛状根和它的再生植株相比,外源激素GA3和IAA能显著提高毛状根培养物中芹菜素和总黄酮的含量。毛状根系的组织干重与类黄酮的含量没有相关性,但毛状根系的再生率与类黄酮的含量几乎呈反相关性。  相似文献

Monitoring is indispensable for the optimization and simulation of biotechnological processes. Hairy roots (hr, plant tissue cultures) are producers of valuable relevant secondary metabolites. The genetically stable cultures are characterized by a rapid filamentous growth, making monitoring difficult with standard methods. This article focuses on the application of laser speckle photometry (LSP) as an innovative, non‐invasive method to characterize Beta vulgaris (hr). LSP is based on the analysis of time‐resolved interference patterns. Speckle interference patterns of a biological object, known as biospeckles, are characterized by a dynamic behavior that is induced by physical and biological phenomena related to the object. Speckle contrast, a means of measuring the dynamic behavior of biospeckles, was used to assess the biospeckle activity. The biospeckle activity corresponds to processes modifying the object and correlates with the biomass growth. Furthermore, the stage of the cultures’ physiological development was assessed by speckle contrast due to the differentiation between active and low active behavior. This method is a new means of monitoring and evaluating the biomass growth of filamentous cultures in real time. As a potential tool to characterize hairy roots, LSP is non‐invasive, time‐saving, can be used online and stands out for its simple, low‐cost setup.  相似文献

Rhodiola quadrifida is a rare mountain medicinal plant whose root extracts are used in traditional Chinese medicine as a hemostatic, antitussive, and tonic in the treatment of gynecological diseases. The aim of the study was to obtain R. quadrifida cultures at different degrees of differentiation in vitro and compare their growth characteristics and the content of salidroside and rosavin. Hairy roots were obtained by incubating cotyledons and hypocotyls in a suspension of Agrobacterium rhizogenes strain A4. The presence of the rolB and rolC genes was proven by polymerase chain reaction. The obtained roots were cultivated in Murashige-Skoog medium (MS). Calluses were obtained from the hairy roots in MS medium with the addition of hormones: 3 mg/L 2,4 D and 0.5 mg/L BAP. The presence of the main secondary metabolites of R. quadrifida, salidroside and rosavin, in calluses and salidroside in hairy roots by HPLC/MS was confirmed. The content of salidroside in callus culture was significantly higher than in hairy roots, 0.158 and 0.047%, respectively. The content of rosavin in callus culture was 0.07%. The content of rosavin and salidroside in callus culture was close to the level of these substances in the rhizomes of R. quadrifida plants growing in vivo, making this culture promising for its possible biotechnological use.  相似文献

The demand for natural and nonpersistent insecticides is increasing day by day. Plant cell cultures could be an alternative to conventional methods of production of insecticides from field-grown plants. In vitro cultured plant cells produce a wide array of insecticides as a part of their secondary metabolism. Their ability to synthesize key enzymes and the manipulation of these could lead to the enhanced production of many insecticides of industrial importance. The development of a high-yielding hairy root culture system for thiophenes, nicotine, and phytoecdysones is of considerable interest. In this article, the current literature on various factors that influence the growth, production, and secretion of six insecticidal compounds, namely, pyrethrins, azadirachtin, thiophenes, nicotine, rotenoids, and phytoecdysones which have been prospects for the scale-up of cell cultures, genetic engineering to obtain transgenic plants, and metabolically engineered plants for increased production of bio-molecules, has been discussed. Environmental safety clearance and the future prospects of application of bio-molecules for plant-derived insecticides are presented.  相似文献

An unstable anthraquinone was isolated from hairy root cultures of Sesamum indicum after preventing light throughout all experimental procedures. The structure of the (Z)-isomer of a previously isolated anthraquinone was determined to be 2-[(Z)-4-methylpenta-1,3-dien-1-yl]anthraquinone by spectroscopic methods. Abstract

Betulin (B) and betulinic acid (BA) are two triterpenoids with a wide range of biological and medicinal activities in different organs of Betula pendula. This research aimed to increase the accumulation of B and BA in the hairy root culture of B. pendula by seven biotic and abiotic elicitors. Hairy root was induced in the stem’s inner bark of B. pendula using the C58C1 strain in the WPM (Woody Plant Medium). The effects of different concentrations of elicitors and different time of root harvest in hairy root culture of B. pendula showed that highest level of growth index (GI), B, and BA was acquired in treated hairy roots with chitosan (CTS), chlorocholine chloride (CCC) and chitosan nano-fiber (CTS NF). Highest GI of B. pendula hairy roots was 13 that was obtained in the roots treated with CTS 150?mg l?1 on the 8th day. The highest content of BA was 1.3?mg g?1 DW after treatment with 1?mg l?1CCC on the 4th and 6th days and 200?mg l?1CTS NF on the 10th day. The highest B content (0.94?mg g?1DW) was obtained in the treated hairy root by 2?mg l?1 CCC after 4 and 6?days.  相似文献

Different germ-cell stages of Drosophila males with a double marked Y-chromosome and either a normal X- or a ring-X chromosome were irradiated with X-rays, inducing the following aberrations: chromosome loss, chromosome gain (XYX-females), partial Y loss and isochromosomes of the Y-chromosome.Doses of 520 rad in spermatocytes and spermatids and 2600 rad in sperm, produced the same effect in these stages with regard to the chromosome loss in the males with a normal X, and the following results were obtained: (a) The partial Y loss in postmeiotic stages is small in comparison with spermatocytes in both stocks. This could mean that in spermatocytes this aberration is determined by exchange processes which can only be induced and/or detected in premeiotic stages. (b) In spermatocytes and mature sperm of males with a ring-X chromosome, the chromosome loss was 2.9 times greater than in those with a normal X. In spermatids of the males with a ring-X the rate of loss was only 1.5 times greater. In spermatocytes of either males with a ring-X or a normal X a similar high rate of isochromosomes could be induced. However, in spermatids and mature sperm the rate of induction of isochromosomes was found to be very small. These results seem to indicate that in mature sperm the rejoining of breaks in the Y-chromosome takes place before, and in the X-chromosome usually after the replication. If in post-meiotic stages of Drosophila the X- and Y-chromosomes existed as chromatid-like subunits then in spermatids these should behave as a structural unit.In sperm we were able to induce similar frequencies of individuals with a single isochromosome type in all body cells as of individuals with two types of isochromosomes (isochromosome mosaics). This result seems to indicate that after irradiation of sperm one of the first two division nuclei is lethal in an proportion of the zygotes.  相似文献
