Rising atmospheric CO2 concentrations are predicted to have a significant impact on global phytoplankton populations. Of particular interest in freshwater systems are those species that produce toxins or impact water quality, though evidence for how these species, and many others, will respond is limited. This study investigated the effects of elevated CO2 (1,000 ppm) relative to current atmospheric CO2 partial pressures (400 ppm), on growth, cell size, carbon acquisition, and photophysiology of five freshwater phytoplankton species including a toxic cyanophyte, Raphidiopsis raciborskii, from Lake Wivenhoe, Australia. Effects of elevated CO2 on growth rate varied between species; notably growth rate was considerably higher for Staurastrum sp. and significantly lower for Stichococcus sp. with a trend to lower growth rate for R. raciborskii. Surface area to volume ratio was significantly lower with elevated CO2, for all species except Cyclotella sp. Timing of maximum cell concentrations of those genera studied in monoculture occurred in the lake in order of CO2 affinity when free CO2 concentrations dropped below air equilibrium. The results presented here suggest that as atmospheric levels of CO2 rise, R. raciborskii may become less of a problem to water quality, while some species of chlorophytes may become more dominant. This has implications for stakeholders of many freshwater systems.  相似文献

The effect of four microalgal chlorophytes, Chlorella vulgaris,Scenedesmus quadricauda, Chlorococcum sp. and Ankistrodesmus convolutus, on the survival, larval development and adultbody size of the mosquito Aedes aegypti was investigated. The percentage mortality of larvae fed with C. vulgaris, Chlorococcum sp.or S. quadricauda after six days was 100%, 84% and 88%,respectively. Delayed pupation and body size reduction of the mosquitoesfed with C. vulgaris, Chlorococcum sp. and S. quadricaudawere observed. In contrast, larvae fed with A. convolutus werebigger than those fed with normal insectory feed. The study showed thatC. vulgaris, S. quadricauda and Chlorococcum sp. have potentialto be used as larvicidal agents.  相似文献

When the chlorophyte alga Dunaliella tertiolecta Butcher is placed in darkness, a form of programmed cell death with many similarities to apoptosis is induced, including the induction of caspase‐like proteases. Many uncertainties about the regulation and mediators that participate in the process remain. To examine the relationship between caspase‐like activities and different apoptotic events (i.e., phosphatidylserine [PS] translocation), increases in membrane permeability and numbers of dead cells revealed by SYTOX‐green staining, and the generation of reactive oxygen species (ROS), we used the broad‐range caspase inhibitor Boc‐D‐FMK to block the activity of the whole class of caspase‐like proteins simultaneously. In the presence of the inhibitor, ROS were not produced, and cells did not die. Loss of membrane asymmetry, indicated by external labeling of PS by annexin V, was apparent at midstages of light deprivation, although it did not conform to the typical pattern for PS exposure observed in metazoans or vascular plants, which occurs at early stages of the apoptotic event. Thus, we have evidence for a link between ROS and cell death involving caspase‐like enzymes in an alga. The fact that caspase‐like inhibitors prevent not only cell death, but also ROS and loss of cell membrane integrity and asymmetry, suggests that caspase‐like proteases might have regulatory roles early in cell death, in addition to dismantling functions.  相似文献

The biosynthesis of nutritionally important polyunsaturated fatty acids (PUFAs) in phytoplankton is influenced by environmental temperature. We investigated the potential of climate warming to alter lipid dynamics of Scenedesmus obliquus (Turpin) Kütz. by comparing lipid and fatty acid (FA) profiles as well as FA metabolism (using [1‐14C] acetate) at 20°C and 28°C. We documented an overall decline (53%–37%) in the proportion of n‐3 PUFA (in particular, of α‐linolenic acid [ALA; 18:3n‐3]), and a concomitant increase in saturated fatty acids (SAFAs) in total lipids (TLs) at 28°C, consistent with enhanced incorporation of radioactivity from [1‐14C] acetate into total 16:0, 18:1, and decreased incorporation into 18:2 and 18:3 FA (from 36% to 22% of the total) at 28°C. Glycerophospholipids were also affected by warming; ALA and stearidonic acids (SDAs; 18:4n‐3) both decreased (by 13% and 15%, respectively) in phosphatidylcholine (PC) and (by 24% and 20%, respectively) in phosphatidylethanolamine (PE). The characteristic FA in phosphatidylglycerol (PG; 16:1n‐13t) increased (by 22%) at 28°C. The activities of desaturases, which add double bonds to FA moieties, comprised the major suite of reactions affected by the temperature increase in TL and polar lipid (PL) classes. Climate modelers predict an increase in the number of extreme heat days in summer at temperate latitudes, with parallel projected increases in water temperatures of shallow water bodies. Our results suggest that the overall decrease in the essential n‐3 FA ALA in S. obliquus at higher water temperatures may lower food quality for higher tropic levels, adding another climate‐warming stress.  相似文献

Cadmium forms neutral, lipophilic CdL20 complexes with diethyldithiocarbamate (L = DDC) and with ethylxanthate (L = XANT). In a synthetic solution and in the absence of natural dissolved organic matter (DOM), for a given total Cd concentration, uptake of these complexes by unicellular algae is much faster than the uptake of the free Cd2+ cation. The objective of the present study was to determine how this enhanced uptake of the lipophilic CdL20 complexes was affected by the presence of natural DOM (Suwannee River humic acid, SRHA). Experiments were performed with Cd(DDC)20 and Cd(XANT)20 at two pH values (7.0 and 5.5) and with the three chlorophytes [Chlamydomonas reinhardtii P. A. Dang., Pseudokirchneriella subcapitata (Korshikov) Hindák, Chlorella fusca var. vacuolata Shihira et R. W. Krauss]. Short‐term uptake (30–40 min) of the CdL20 complexes was followed in the absence and presence of SRHA (6.5 mg C · L?1). Acidification from pH 7.0 to 5.5 decreased CdL20 uptake by the three algae, in the presence or absence of humic acid (HA). The dominant effect of the HA was to decrease Cd uptake, due to its interaction with the CdL20 complexes in solution. However, if uptake of the free CdL20 complexes was compared in the presence and absence of HA, in four of eight cases initial uptake rate constants (ki) were significantly higher (P < 0.05) in the presence of the HA, suggesting the operation of an interfacial effect of the HA at the algal cell membrane, favoring uptake of CdL20. Overall, the experimental results suggest that neutral metal complexes will be less bioavailable in natural waters than they are in synthetic laboratory media in the absence of natural DOM.  相似文献

Exponentially growing cultures of Thalassiosira fluviatilis Hustedt and Dunaliella tertiolecta Butcher were exposed to 4 min temperature shocks of 5° to 20°C above ambient (20°C). Photosynthetic carbon fixation, changes in in vivo fluorescence and fluorescence on the addition of the herbicide DCMU (3-(3,4-dichlorophenyl)-1,1-dimethylurea) were measured over the subsequent 24 h. The fluorescence ratio (R, DCMU-enhanced fluorescence/in vivo fluorescence) paralleled changes in photosynthesis over this period; both were significantly reduced (P < 0.05) by temperature shocks of +15° and +20° C, but +5° and +10° C treatments had no inhibitory effect on either relative to the control. The instantaneous response obtained with the fluorescence ratio indicates that the technique might be applicable to routine bioassay procedures and thus replace the time consuming methods now used for the estimation of 14C-incorporation and growth rates.  相似文献

Chemostat-grown cells of the chlorophyte Dunaliella tertiolecta (Butcher) exposed to triphenyltin were examined using transmission electron microscopy. Following a 1-h exposure to 21 and 84μM triphenyltin, mitochondria underwent structural damage and the thylakoid membranes of a small proportion of cells spread from the usual compact arrangement. Uptake of lipophilic metal complexes by freshwater algae has recently been shown to be pH dependent. Here we look at different physiological aspects that could influence the diffusion of the lipophilic Cd complex, Cd(diethyldithiocarbamate)20 (Cd(DDC)20), into algal cells at different exposure pH values. Changes in cell membrane permeability were assessed as a function of pH for three species of green algae [Chlamydomonas reinhardtii P. A. Dang., Pseudokirchneriella subcapitata (Korshikov) Hindák, and Chlorella fusca var. vacuolata Shihira et R. W. Kraus] using two neutral, nonionic probes, fluorescein diacetate (FDA) and D‐sorbitol. In parallel experiments, we exposed algae to inorganic Cd or to Cd(DDC)20 and monitored Cd intracellular metal distribution, together with phytochelatin synthesis. For the three algal species acclimated at pH 5.5 (w/wo DDC 1 μM) and exposed at this pH, their permeability to FDA and D‐sorbitol was consistently lower than for algae growing at pH 7.0 and exposed at this pH (P < 0.001). The ratio of the FDA hydrolysis rate measured at pH 7.0 with respect to the rate measured at pH 5.5 (both in the presence of DDC) correlated with the ratio of the Cd(DDC)20 initial internalization rate constant obtained at pH 7.0 versus that obtained at pH 5.5 (three algae species, n = 9, r = 0.85, P = 0.004). Our results strongly suggest that acidification affects metal availability to algae not only by proton inhibition of facilitated metal uptake but also by affecting membrane permeability.  相似文献

Acclimation to UV radiation (UVR) was examined in a unicellular chlorophyte isolated from fumarole fields adjacent to Mt. Erebus, Antarctica. Long‐term exposure to UVR (14 days) initially reduced the efficiency of photosynthetic energy conversion measured as the dark adapted quantum yield of PSII fluorescence (Fv/Fm) when compared with cultures not exposed to UVR. However, the UVR exposed cultures recovered to pre‐UVR exposure levels of photosynthetic efficiency by the end of the experimental period. Additionally, neither growth rates nor cell size were significantly affected by exposure to UVR. In contrast, the maximum rate of carbon fixation on a chl a basis was significantly reduced at the end of the experiment and was accompanied by a significant decrease in chl a concentrations. These results suggest a transient effect of UVR on primary photochemistry consistent with damage to PSII, possibly at the D1 protein, with more chronic effects on carbon fixation that did affect maximum photosynthetic capacity. Ultrastructural and molecular (18s rDNA) data show that this isolate from the fumoral fields of Mt. Erebus, Antarctica is a member of the genus Scenedesmus.  相似文献

The temperature dependence of NADH:NR activity was examined in several marine phytoplankton species and vascular plants. These species inhabit divergent thermal environments, including the chromophytes Skeletonema costatum (12–15° C), Skeletonema tropicum (18–25° C), Thalassiosira antarctica (?2 to 4° C), and Phaeocystis antarctica (?2 to 4° C), the green alga Dunaliella tertiolecta (14–28° C), and the vascular plants Cucurbita maxima (20–35° C) and Zea mays (20–25° C). Despite the difference in growth habitats, similar temperature response curves were observed among the chromophytic phytoplankton, with temperatures optimal for NR activity being between 10–20° C. In contrast, the chlorophyll b‐containing alga and vascular plants exhibited optimal temperatures for NR activity above 30° C. Such dramatic differences in NR thermal characteristics from the two taxonomic groups reflect a divergence in NR structure that may be associated with the evolutionary diversification of chromophytes and chlorophytes. Further, it suggests a potential contribution of the thermal performance of NR to the geographic distributions, seasonal abundance patterns, and species composition of phytoplankton communities. NR partial activities, which assess the individual functions of Mo‐pterin and FAD domains, were evaluated on NR purified from S. costatum to determine the possible causes for high temperature (>20° C) inactivation of NR from chromophytes. It was found that the FAD domain and electron transport among redox centers were sensitive to elevated temperatures. S. costatum cells grown at 5, 15, and 25° C exhibited an identical optimal temperature (15° C) for NADH:NR activity, whereas the maximal NR activity and NR protein levels differed and were positively correlated with growth temperature and growth rate. These findings demonstrate that thermal acclimation of NO3? reduction capacity is largely at the level of NR protein expression. The consequences of these features on NO3? utilization are discussed.  相似文献
