ABSTRACT

The effluents coming from the dye industries are colored and polluted, and the disposal of these wastes into receiving waters causes damage to the water as well as the environment. The use of rice husk for the removal of dye from wastewater has been explored in a stir tank reactor. The effects of operation variables such as adsorbent dosage, contact time, dye concentration, initial pH, and agitation on the removal of safranin were investigated in a stirred tank reactor. The combined effect of various process parameters on dye removal were analyzed using response surface methodology (RSM), and the modeling of the process parameter had been done using the artificial neural network simulation method. It was observed that response surface methodology can determine the optimization of the process parameters and the model derived from the simulation of the artificial neural network (ANN) (deviation from experimental results was ~0.09%) described the process variable efficiently. It was observed that at the initial solution pH of 6.28 and adsorbent dosage of 15.63 g L?1, dye removal of safranin was 97%.  相似文献

Recent increases in the ability to detect low levels of immunofluorescence have shown the need for highly purified primary immunoreagents. There are now reports of purification of monoclonal antibodies using HPLC with reverse phase columns. In this study we have utilized standard size exclusion HPLC to purify both biotinylated and non-biotinylated monoclonal antibodies from hybridoma culture supernatants. Results indicated that both biotinylated and non-biotinylated monoclonal antibodies retained their antigen binding capacity after purification, and were not different in this capacity from commercially available, affinity purified reagents. These findings indicate that size exclusion HPLC may be used in the purification of biologically active monoclonal antibodies, and suggest that this technique may be used in the large scale production of antibodies and their fragments, in antibody purification from ascites fluid, and in antisera quality control.  相似文献

Improved production of teicoplanin using adsorbent resin in fermentations   总被引:2,自引:0,他引:2  
AIMS: To use adsorbent resins in fermentations to eliminate toxic effects on growth, reduce feedback repression of production and assist in recovery of teicoplanin. METHODS AND RESULTS: An adsorbent resin was added to the culture broth for the adsorption of teicoplanin. Amberlite XAD-16, Diaion HP-20, charcoal and silica gel were investigated as adsorbent resins. The adsorbed teicoplanin was extracted from the resin by 80% methanol after fermentation. Antibiotic activity was quantified by the disc-agar diffusion assay against Bacillus substilis, and qualitative evaluations were based on HPLC using YMC-Pack ODS-A column. Diaion HP-20 was the most effective adsorbent resin when added at a concentration of 5% (w/v) in the inoculation stage. CONCLUSIONS: Addition of Diaion HP-20 in fermentations eliminated toxic effects on growth and reduced feedback repression of teicoplanin by adsorption. There was a 4.2-fold increase in the quantities of teicoplanin. Addition of adsorbent assisted in the recovery of teicoplanin by reducing the recovery steps. SIGNIFICANCE AND IMPACT OF THE STUDY: The results of this study provide useful information for the production of teicoplanin, a glycopeptide antibiotic produced by Actinoplanes teicomyceticus. Addition of adsorbent in fermentation increased productivity of teicoplanin by more than five times.  相似文献

Silica gel bead coated with macroporous chitosan layer (CTS-SiO2) was prepared, and the metal immobilized affinity chromatographic (IMAC) adsorbents could be obtained by chelating Cu2+, Zn2+, Ni2+ ions, respectively on CTS-SiO2, and trypsin could be adsorbed on the IMAC adsorbent through metal–protein interaction forces. Batch adsorption experiments show that adsorption capacity for trypsin on these IMAC adsorbent variated with change of pH. The maximal adsorption reached when the solution was in near neutral pH in all three IMAC adsorbents. Adsorption isothermal curve indicated that maximal adsorption capacity could be found in the Cu2+-CTS-SiO2 with the value of 4980 ± 125 IU g−1 of the adsorbent, while the maximal adsorption capacity for trypsin on Zn2+ and Ni2+ loaded adsorbent was 3762 ± 68 IU g−1 and 2636 ± 53 IU g−1, respectively. Trypsin immobilized on the IMAC beads could not be desorbed by water, buffer and salt solution if the pH was kept in the range of 5–10, and could be easily desorbed from the IMAC beads by acidic solution and metal chelating species such as EDTA and imidazole. The effect of chelated metal ions species on CTS-SiO2 beads on the activity and stability of immobilized trypsin was also evaluated and discussed. Trypsin adsorbed on Zn-IMAC beads retained highest amount of activity, about 78% of total activity could be retained. Although the Cu-IMAC showed highest affinity for trypsin, only 25.4% of the calculated activity was found on the beads, while the activity recovery found on Ni-IMAC beads was about 37.1%. A remarkable difference on stability of trypsin immobilized on three kinds of metal ion chelated beads during storage period was also found. Activity of trypsin on Cu-IMAC decreased to 24% of its initial activity after 1-week storage at 4 °C, while about 80% activity was retained on both Ni-IMAC and Zn-IMAC beads. Trypsin immobilized on Zn-CTS-SiO2 could effectively digest BSA revealed by HPLC peptide mapping.  相似文献

Ion-exchange chromatography relies on electrostatic interactions between the adsorbent and the adsorbate and is used extensively in protein purification. Conventional ion-exchange chromatography uses ligands that are singly charged and randomly dispersed over the adsorbent, creating a heterogeneous distribution of potential adsorption sites. Clustered-charge ion exchangers exhibit higher affinity, capacity, and selectivity than their dispersed-charge counterparts of the same total charge density. In the present work, we monitored the transport behavior of an anionic protein near clustered-charge adsorbent surfaces using fluorescence correlation spectroscopy. We can resolve protein-free diffusion, hindered diffusion, and association with bare glass, agarose-coated, and agarose-clustered peptide surfaces, demonstrating that this method can be used to understand and ultimately optimize clustered-charge adsorbent and other surface interactions at the molecular scale.  相似文献

A novel bioreactor with an internal adsorbent was developed for the simultaneous fermentation and recovery of prodigiosin-like pigment produced from Serratia sp. KH-95 as a model product in one bioreactor. The pigment concentration recovered in the internal adsorbent was 13.1 g l–1, which was 1.8-fold higher than that obtained in a bioreactor with an external adsorbent.  相似文献

A robust, naturally evolving methanotrophic community in landfill cover soil (LFCS) can be the simplest way to mitigate landfill methane emission. In this study, bacterial community composition in LFCS and methane oxidation potential of enriched methanotrophic consortium, in comparison to that of axenic Methylosinus sporium, was investigated. Growth and methane oxidation of the consortium was studied in liquid phase batch experiments under varying temperature (20–40°C), pH (5–10), headspace CO2, and in presence of porous adsorbent (1.3 cm3 sponge cubes). The 16S rRNA gene analysis revealed presence of both type-I and type-II methanotrophs along with few obligate methylotroph in LFCS. Though the optimal growth condition of the consortium was at 30°C and pH 7, it was more resilient in comparison to M. sporium. With increasing availability of porous adsorbent, methane consumption by the consortium was significantly improved (p < 0.001) reaching a maximum specific methane oxidation rate of 11.4 μmol mg?1 biomass h?1. Thus, inducing naturally thriving methanotrophs in LFCS is a better alternative to axenic methanotrophic culture in methane emission management.  相似文献

Flow-ELISA measurements of the monoclonal antibody concentration in cultivation broth containing murine hybriboma cells were carried out using a small expanded-bed column (0.5×2.5 cm) charged with protein A. A new specialised pellicular agarose/stainless steel matrix designed for high flow rates with fast mass transport properties was used. Special care was taken to get an efficient flow distribution. The axial dispersion coefficient was very low (2×10–6 m2 s–1 for latex particles at a linear velocity of 10 cm min–1). Breakthrough curves for polyclonal IgG on the protein A-derivatised support (at 2–11 cm min–1) further emphasised its advantageous properties. No significant change in dynamic capacity was found over the entire speed range.  相似文献

以甜菜渣、果胶、海藻酸钠为原料 ,与表氯醇在碱性条件下交联 ,制备了果胶酶亲和吸附剂并应用这几种亲和吸附剂对草酸青霉果胶酶和黑曲霉果胶酶进行亲和层析实验。结果表明 :用小于 60目的甜菜渣粉末和海藻酸钠制备的果胶酶亲和吸附剂 (分别简写为SBP吸附剂和SA吸附剂 )具有较好的纯化果胶酶性能 ,其中SBP吸附剂性能稳定 ,而SA吸附剂膨胀系数较大 ,在流动相改变时引起流速不稳。  相似文献
