 利用细胞培养的方法,测定五肽胃泌素(Boc-β-Ala-Trp-Met-Asp-phe·NH_2)、葡萄糖-1-脱氧果糖四肽(GIe-1-Deoxyflu-Trp-Met-Asp-phe·NH_2)及Boc-三肽(Boc-Met-Asp-Phe·NH_2)等胃泌素类似物对~3H-胸腺嘧啶核苷(~3H-TdR)参入胃粘膜细胞DNA合成的影响。实验结果表明五肽胃泌素对胃粘膜细胞DNA合成有明显促进作用,而泌酸活性比五肽胃泌素还强的葡萄糖-1-脱氧果糖四肽却没有这种作用。进一步证明胃泌素的营养作用是独立于泌酸活性而存在的,二者有着不同的作用机理。  相似文献

 本文用HPLC分离五肽胃泌素(Pentagastrin,Boc-β-Ala-Trp-Met-Asp-Phe·NH_2,PG_(1-5))及其N-端无保护基的TFA·五肽(β-Ala-Trp-Met-Asp-Pne·NH_2·TFA,TFA·PA_(1-5))被胃泌酸细胞膜和肝匀浆降解之产物,并对它们的氨基酸组成作了分析,又用薄层层析作了进一步验证,从而提出了酶促降解时可能的酶切位点。实验结果表明,N-端的Boc-基团能大幅度增加对酶降解的抵抗力,这从一个方面解释了为什么商品五肽胃泌素的泌酸活性大于其它胃泌素类似物的原因。  相似文献

Summary. This paper describes the enzymatic synthesis of the C-terminal fragment H-Gly-Trp-Met-Asp-Phe-NH2 of cholecystokinin. Immobilized enzymes were used for the formation of all peptide bonds except thermolysin. Beginning the synthesis with phenylacetyl (PhAc) glycine carboxamidomethyl ester (OCam) and H-Trp-OMe by using immobilized papain as biocatalyst in buffered ethyl acetate, the dipeptide methyl ester was then coupled directly with Met-OEt·HCl by -chymotrypsin/Celite 545 in a solvent free system. For the 3+2 coupling PhAc-Gly-Trp-Met-OEt had to be converted into its OCam ester.The other fragment H-Asp(OMe)-Phe-NH2 resulted from the coupling of Cbo-Asp(OMe)-OH with H-Phe-NH2·HCl and thermolysin as catalyst, followed by catalytic hydrogenation.Finally PhAc-Gly-Trp-Met-Asp-Phe-NH2 was obtained in a smooth reaction from PhAc-Gly-Trp-Met-OCam and H-Asp(OMe)-Phe-NH2 with -chymotrypsin/Celite 545 in acetonitrile, followed by basic hydrolysis of the -methyl ester. The PhAc-group is removed with penicillin G amidase and CCK-5 is obtained in an overall isolated yield of 19.6%.  相似文献

This study describes the interaction between gastrin and Helicobacter pylori. Human gastrin amino acids 4-17 were found to be the minimal growth-stimulating sequence. Gastrin from other mammals did not stimulate bacterial growth. When human serum was used to stimulate bacterial growth in brucella broth, gastrin was shown to be a necessary and sufficient growth-stimulating factor. Competition for the gastrin effect by pentagastrin and cholecystokinin (CCK-8) resulted in inhibition of bacterial growth. This effect was mediated by the four C-terminal amino acids which are shared by gastrin, CCK-8 and pentagastrin. In conclusion, the interaction between gastrin and H. pylori was shown to be specific, essential, and dependent on a defined gastrin sequence.  相似文献

The effects of an amino acid derivative (N-benzoyl-l-argininamide), four small peptides (Phe-Gly-Phe-Gly, gastrin-related peptide (Trp-Met-Arg-Phe-NH2), tetragastrin (Trp-Met-Asp-Phe-NH2), pentagastrin (Boc-βAla-Trp-Met-Asp-Phe-NH2)) and one medium-sized peptide. glucagon (29 residues), on the gel-to-liquid crystalline transition of a multilamellar suspension of dimyristoylphosphatidylcholine have been studied by means of high-sensitivity differential scanning calorimetry. At low concentrations of added solutes, the temperature at which the excess apparent specific heat in the gel-to-liquid crystalline phase transition of the lipid is maximal is lowered by an amount proportional to the total concentration of the peptide, with proportionality constants ranging from ?0.018 K mM?1 for Phe-Gly-Phe-Gly to ?3.1 K mM?1 for the gastrin-related peptide. The lipid mixtures involving the first two solutes listed above exhibited approximately symmetrical curves of excess apparent specific heat vs. temperature. The curves for the other solutes were asymmetric, and could be well represented as the sum of either two or three two-state curves. The asymmetry, which was especially pronounced in the cases of pentagastrin and glucagon, thus appeared to be due to the presence of components having lower and/or higher transition temperatures than that of the lipid. Pentagastrin and glucagon (R.M. Epand and J.M. Sturtevant, Biochemistry 20 (1981) 4603) have much smaller effects on the gel-to-liquid crystalline phase transition of dipalmitoylphosphatidylcholine than on that of the dimyristoyl analog.  相似文献
