Methylamines are frequently present in high concentrations in biological samples, but their separation and quantification are difficult. Data presented show that methylamines commonly occurring in biological material can be uniquely identified and quantified by proton nuclear magnetic resonance spectroscopy by recording spectra at both neutral and acid pH. Use of a high sensitivity probe permits this analysis even in the presence of high water concentrations, allowing accurate quantification with minimum preparative technique. The method was tested on tissues of the dogfish. Trimethylamine oxide was found in amounts ranging from 42 mmol kg−1 fresh weight in liver, up to 115 mmol kg−1 fresh weight in heart. Betaine was found to range from 10 mmol kg−1 fresh weight in liver to 49 mmol kg−1 fresh weight in brain. Creatine was not found in heart or liver, but was present in body wall muscle and in brain. Further analysis using high-performance liquid chromatography allowed determination of urea/methylamine ratios, which ranged from 1.9 in liver to 3.7 in body wall muscle. Summary Hydrozoa replace used-up nematocytes (cnidocytes) by proliferation and differentiation from interstitial stem cells (i cells). Repeated pulsed exposure ofHydra to elevated levels of unprotonated ammonia leads to successive loss of the various types of nematocytes: first of the stenoteles, then of the isorhizas and finally of the desmonemes. The loss is due to deficits in supply; the number of nematoblasts and differentiating intermediates is reduced. In the hydroidHydractinia the main process leading to numerical reduction was observed in vivo: mature nematocytes as well as precursors emigrate from their place of origin into the gastrovascular channels where they are removed by phagocytosis. This is a regular means by which these animals down-regulate an induced surplus of nematocytes. With lower effectiveness, pulses of methylamine, trimethylamine and glutamine also induce elimination of the nematocyte lineages. In the long term the population of nerve cells, which are permanently but slowly renewed from interstitial neuroblasts, decreases, too. After 2 months of daily repeated treatment the density of the Arg-Phe-amide-positive nerve cells was reduced to 50% of its normal level. Thus, ammonia induces down-regulation of all interstitial cell lineages. The temporal sequence of the ammonia-induced loss reflects the diverse rates with which the various i cell descendants normally are renewed.  相似文献

Abstract Extremely halophilic methanogens and a rodshape homo-acetogen were isolated from cyanobacterial mats of Sivash, Crimea. Trimethylamine served as a single source of energy for both groups. Methanogenic strains obtained constitute a continuum of forms, from moderately halophilic to those living at salt saturation. A new homo-acetogen, Acetohalobium arabaticum growing in the salinity range of 10–25% produced acetate, mono-, di- and trimethylamines from betaine. A limited number of substrates including formate, H2+ CO2, CO, lactate, pyruvate and hystidine was utilized. A trophic chain is discussed for methanogenesis in hypersaline environments.  相似文献
