Abstract: The effect of ATP, Mg2+, or MgATP on the release of luteinizing hormone-releasing hormone (LH-RH) from hypothalamic granules was examined under in vitro conditions. Granules, isolated from adult male hypothalami, were incubated at 37°C in a buffered (pH 7.8) medium containing 0.15 m -KCl. The addition of ATP to the incubation mixture did not stimulate the release of LH-RH. In contrast, the addition of MgATP stimulated the release of LH-RH, the release being 62% greater than control. The addition of Mg2+ to the incubated granules also stimulated the release of LH-RH. However, the magnitude of this Mg2+-stimulated release of LH–RH was significantly ( P < 0.01) lower than that of the MgATP-stimulated release, indicating that ATP stimulates LH-RH release in a Mg2+-dependent manner. As both MgATP and Mg2+ alone stimulated LH-RH release, we characterized further these two release processes by incubating the granules under one of the following conditions: incubation at 4°C in a buffered medium containing 0.15 m -KCl or incubation at 37°C in a medium that does not contain KCl. Under these two incubation conditions, the MgATP-stimulated release of LH-RH was not manifested, whereas the Mg2+-stimulated release of LH-RH was manifested. On the basis of these differences, we propose that two different processes can lead to the release of LH-RH from isolated hypothalamic granules: one process involves ATP and Mg2+ (MgATP) and another process involves Mg2+ alone.  相似文献

Two novel small molecule gonadotropin-releasing hormone (GnRH) receptor antagonists (12 and 13) of the furamide-class were synthesized and evaluated in vitro for their receptor binding affinities for the rat GnRH receptor. Radiolabeling with no carrier added fluorine-18 of the appropriate precursors was investigated in a one-step reaction. Log P (Octanol/PBS pH 7.4) and serum stability of the compounds were investigated. The antagonists showed low nM affinity for the rat GnRH receptor. 18F-radiolabled compounds were obtained in high radiochemical purity (>95%) and specific activity (>75 GBq/μmol). These findings suggest this class of compounds holds promise as potential probes for PET targeting of GnRH-receptor expression.  相似文献

The molecular circadian clock mechanism is highly conserved between mammalian and avian species. Avian circadian timing is regulated at multiple oscillatory sites, including the retina, pineal, and hypothalamic suprachiasmatic nucleus (SCN). Based on the authors’ previous studies on the rat ovary, it was hypothesized that ovarian clock timing is regulated by the luteinizing hormone (LH) surge. The authors used the chicken as a model to test this hypothesis, because the timing of the endogenous LH surge is accurately predicted from the time of oviposition. Therefore, tissues can be removed before and after the LH surge, allowing one to determine the effect of LH on specific clock genes. The authors first examined the 24-h expression patterns of the avian circadian clock genes of Bmal1, Cry1, and Per2 in primary oscillatory tissues (hypothalamus and pineal) as well as peripheral tissues (liver and ovary). Second, the authors determined changes in clock gene expression after the endogenous LH surge. Clock genes were rhythmically expressed in each tissue, but LH influenced expression of these clock genes only in the ovary. The data suggest that expression of ovarian circadian clock genes may be influenced by the LH surge in vivo and directly by LH in cultured granulosa cells. LH induced rhythmic expression of Per1 and Bmal1 in arrhythmic, cultured granulosa cells. Furthermore, LH altered the phase and amplitude of clock gene rhythms in serum-shocked granulosa cells. 目的研究羊骨胶原肽(sheep bone collagen peptide,SBCP)对类固醇诱导的去卵巢大鼠催乳素(pro-lactin,PRL)和促黄体生成素(luteinizing hormone,LH)分泌脉冲的影响。方法 6周龄子宫颈未开口的青春期SD大鼠实施双侧卵巢摘除手术,康复1周或3周后以类固醇替代方法诱导PRL和LH脉冲,处理组同时按1000 mg/(kg.d)的剂量以SBCP灌胃,通过颈导管采集血样,利用放射免疫技术测定各组大鼠外周血中的LH和PRL浓度。结果对于术后康复1周的大鼠,SBCP对LH脉冲振幅起到增强作用,而对PRL脉冲没有影响;对于术后康复3周的大鼠,SBCP对LH和PRL脉冲振幅均表现为降低作用,但卵巢摘除后立即给予SBCP可减弱这种降低作用。结论雌激素和孕激素替代注射的同时,补充羊骨胶原肽,不仅仍可诱导LH和PRL脉冲的产生,还可预防由于卵巢摘除带来的骨代谢紊乱,免疫力低下等不良影响,因而是对现有类固醇替代方法的改良。  相似文献

利用在体注射实验和放射免疫测定法,研究了多巴胺能药物对性腺处于再发育期虎纹蛙的促性腺激素释放激素(GnRH)及促黄体激素(LH)分泌活动的影响。结果是:多巴胺(DA)及其激素剂阿扑吗啡(APO)可显著降低血浆LH水平;而多巴胺的拮抗剂-地欧酮(DOM)可显著增加垂体LH含量。DA对脑中cGnRH-Ⅱ的合成有抑制作用,而OM对其mGnRH的释放有一定的刺激作用。结果表明:DA可在脑及垂体水平分别抑制虎纹蛙GnRH和LH的释放,DA对LH释放的抑制作用很可能是通过D2受体实现的。  相似文献

A controlled trial investigating the effect on conception of administration of 250 μg of gonadotropin-releasing hormone (GnRH) at artificial insemination (AI) in dairy cows in seasonal or split calving herds was conducted. Time of detection of estrus, body condition, extent of estrous expression, treatment, breed, age and milk production from the most recent herd test of the current lactation was recorded. Cows were tested for pregnancy with fetal aging between 35 and 135 days after AI. Sixteen herds provided 2344 spring-calved cows and 3007 inseminations. Logistic regression adjusting for clustering at herd level was used to examine the effect of treatment for first (2344) and second (579) inseminations separately. For first AI, treatment significantly improved conception rate in cows with milk protein concentrations of 3.75% or greater and for cows with milk protein concentrations between 3.00% and 3.50% and less than 40 days calved; increased conception rate from 41.2% to 53.4%. Treatment reduced conception rates in cows with milk protein concentrations of 2.75% or less. Treating only cows identified as responding positively to treatment (11% of all study cows) was estimated to increase first service conception rate in herds from 48.1% to 49.4%. There was no significant effect of treatment on conception to second AI, nor any significant interactions. These findings indicate that GnRH at AI should be limited to the sub-group cows most likely to respond. The positive effect of GnRH at AI may be mediated through improved oocyte maturation and/or improved luteal function, rather than by reducing AI-to-ovulation intervals.  相似文献

摘要 目的:分析脐带间充质干细胞对卵巢早衰家兔的治疗效果及机制研究。方法:经腹腔连续注射 2 d 环磷酰胺50 mg/(kg?d)建立卵巢早衰家兔模型。将建模成功的10只家兔随机分成模型组和治疗组,每组5只。建模一周后,治疗组家兔每天经耳缘静脉注射5×106/mL脐带间充质干细胞混悬液2 mL,连续注射3 d。模型组家兔经耳缘静脉注射等量无菌生理盐水。于治疗后 0 d、7 d、14 d和28 d,取家兔静脉血检查血清激素表达水平。于治疗后28 d,检测家兔卵巢中生长卵泡数、封闭卵泡数、黄体数、富半胱氨酸蛋白61(CYR61)和结缔组织生长因子(CTGF)mRNA及蛋白质相对表达量。结果:治疗前,模型组和治疗组家兔血清雌二醇(E2)、促卵泡生成素(FSH)、FSH/黄体生成素(LH)、抑制素B(INHB)和抗苗勒管激素(AMH)、均无显著差异(P>0.05)。与模型组相比,治疗后治疗组家兔血清E2和INHB水平显著上升(P<0.05),FSH水平显著下降(P<0.05),FSH/LH均无显著差异(P>0.05)。随着治疗时间延长,治疗组家兔血清E2和FSH水平周期性波动。治疗28 d后,与模型组相比,治疗组家兔血清AMH水平显著升高(P<0.05);卵巢组织中CYR61和CTGF mRNA及蛋白质相对表达量均显著升高(P<0.05);生长卵泡数显著升高(P<0.05);封闭卵泡数和黄体数均显著降低(P<0.05)。结论:静脉注射脐带间充质干细胞可通过上调CYR61和CTGF的表达,促进卵泡生成,恢复卵巢功能,达到治疗卵巢早衰的临床效应。  相似文献

A highly active inhibitor of the binding of estrone and estradiol-17β to rat alpha-fetoprotein is demonstrated for the first time in embryo, immature and adult rat sera as well as in fetal and adult human sera. The competitive character and the narrow specificity of this inhibition effect is shown. The major compound responsible for this activity is isolated by successive column Sephadex LH20 and thin layer chromatography : it is characterized as a nonpolar, nonphenolic, dialysable and thermostable substance, unreactive towards anti-estrone and anti-estradiol-17β anti-bodies. The possible biological role of an endogenous non-estrogen ligand of rodent fetoproteins is discussed.  相似文献

Summary The effects of implantation of testosterone propionate (TP) in various sites in the hypothalamus on the photoperiodically induced vernal premigratory functions in the White-crowned Sparrows were investigated in order to assess the role of the hypothalamo-hypophysial-testicular axis in the induction of these responses.Implantation of glass capillary tubes containing TP in the basal infundibular nucleus (IN), in the median eminence, or in the pars distalis inhibited the photoperiodically induced increase in plasma levels of luteinizing hormone (LH), as measured by radioimmunoassay, and testicular growth. The effective implants significantly lowered the levels of LH in birds held on nonstimulatory short days. These TP implants apparently inhibited release from the pars distalis of both LH and follicle-stimulating hormone (FSH). It is concluded that the site of sensitivity in the negative feedback by testosterone is either the basal IN or the pars distalis, or both. The implants of TP that inhibited the increase in plasma LH and testicular growth completely did not prevent the birds from fattening.These investigations were supported, in part, by research grants from the National Institutes of Health (HD-6527) and National Science Foundation (BMS 79-13933) to Professor Donald S.
