An analogue of the periodogram method for unequally spaced data is presented with a view to resolving the frequency structure of the observations. The algorithm is explicitly based on the sequential least squares procedure. In particular, the key concept is that the with-in-plot spectral analysis can be augmented by the between-plot information to make inferences about common characteristics. It is also shown how the between-plot random variations can be incorporated into the multiple harmonic regression model. A detailed spectral analysis investigates the periodic fluctuations in four cardio-circulatory variables, measured by autorhythmometric observation by eight men at rest and extending over a time span of 2 years. The spectral curves show the existence of circadian and circaseptan rhythmicities. The amplitude modulation of the dian rhythm by circaseptan variation is assimilated with the rhythmicity of work during the week. The blood-pressure variables situate their maximum annual peak in the winter period. These quasi-periodic fluctuations appear to be related to the amount of physical activity performed in time by the subjects.  相似文献

A new molecular dynamics method for calculating free energy profiles for rare events is presented. The new method is based on the creation of an adiabatic separation between the reaction coordinate subspace and the remaining degrees of freedom within a molecular dynamics run. This is achieved by associating with the reaction coordinate(s) a high temperature and large mass, thereby allowing the activated process to occur while permitting the remaining degrees of freedom to respond adiabatically. In this limit, by applying a formal multiple time scale Liouville operator factorization, it can be rigorously shown that the free energy profiles are obtained directly from the probability distribution of the reaction coordinate subspace and, therefore, require no postprocessing of the output data. The new method is applied to a variety of model problems and its performance tested against free energy calculations using the "bluemoon ensemble" approach. The comparison shows that free energy profiles can be calculated with greater ease and efficiency using the new method.  相似文献

Ahsg (fetuin-A) is a 55-59 kDa phosphorylated glycoprotein synthesized in the adult predominantly by hepatocytes, from which it enters the circulation. When dysregulated, this glycoprotein operates to influence the clinical sequelae of insulin resistance-type 2 diabetes and cardiovascular disease. The pathological sequelae likely arise from two separable molecular “faces” of Ahsg—one acting at the level of the insulin receptor and a second face influencing ectopic biomineralization in the intima. A detailed understanding of these two functional faces of Ahsg is not yet clear for lack of structural studies. Ahsg has a physiological role in the biomineralization of bone, which when dysregulated can lead to ectopic calcification of soft tissues in the vasculature. Ahsg has a second physiological function in regulating how insulin signals through its receptor, a transmembrane tyrosine kinase. Dysregulation of this “face” of Ahsg results in morbid sequelae such as impaired glucose disposal and fatty liver. Ahsg binds to tandem fibronectin type 3 (Fn3) domains present in the 194 amino acid residue extracellular portion of the β-subunit of the insulin receptor, distant from the high-affinity pocket formed by two complementing α-subunits where insulin binds. Only two proteins are known to bind directly to the insulin receptor ectodomain - insulin and Ahsg - the former turns on the receptor's intrinsic tyrosine kinase (TK) activity, and the latter shuts it down. Recent X-ray crystallographic studies of the ectodomain of the insulin receptor now sharpen our understanding of the receptor's extracellular α-subunit and linked β-subunit. Ahsg genotype and its circulating level have been correlated with body morphometrics (obese versus lean and visceral adiposity) in epidemiological studies enrolling thousands of patients. Epidemiological studies from the clinic reveal high levels of circulating Ahsg in insulin resistance and diabetes. This review endeavors to explain how one protein can mediate diverse pathologies, but specifically addresses its metabolic “face” blunting insulin receptor activity, an action leading to insulin resistance.  相似文献

Second Harmonic Generation (SHG) microscopy dates back to 1974, but effective biological use of the technique has a history of barely 10 years. It is now widely used to image collagen in many different applications, and is becoming useful for imaging myosin and some polysaccharides. A separate line on research has focussed on SHG dyes, which can provide high-speed indication of membrane potential and are now in use in neurobiology. In order to gain insight into the effect of watershed conditions on fluctuations in stream water temperature, we statistically analyzed water temperature data for 1 year, using root mean square (Rms) and harmonic (A Amplitude, φ delay time) methods. The average values of delay time (days) between air and water temperatures (T a and T w) of small (< 0.5 ha), medium (0.5–100 ha) and large (> 100 ha) watersheds were 4.53 ± 0.82 days, 11.83 ± 3.88 days and 4.45 ± 1.52 days, respectively. Fluctuations in stream water temperature expressed by Rms (Rms T w/Rms T a) and harmonic methods (A −T w/A −T a) in the medium-sized watersheds with moderate slope gradients were 0.37 ± 0.09 and 0.56 ± 0.14, respectively. These values increased in the larger watersheds with low slope gradients, including five large rivers covered by various landscapes, with their averages of 0.53 ± 0.09 and 0.78 ± 0.09, respectively, indicating the influences of solar radiation and heat transfer processes. In the smaller watersheds with high slope gradients, these values were 0.73 ± 0.02 and 0.87 ± 0.03, respectively, suggesting that shorter passage time affected water temperatures. With respect to forest type, these values at badly managed hinoki forest watersheds (0.45 ± 0.04 and 0.73 ± 0.07) were larger than those at broadleaf forest (0.34 ± 0.04 and 0.51 ± 0.12) and well-managed hinoki forest (0.33 ± 0.04 and 0.51 ± 0.07) watersheds, indicating different proportions of flow paths.  相似文献

本文用二次谐波成像技术(second harmonic generation SHG)来研究飞秒激光切削后角膜结构的变化.在生物学研究,材料科学等方面都有很广泛应用的SHG成像技术能在不破坏的角膜情况下获得高对比度的角膜层析图像,分辨率为500 nm,实验装置是利用现有的双光子显微镜.本文还根据成像结果评价了飞秒激光在角膜切削中的质量,为飞秒激光微米级的精确切削和临床应用提供了实验支持.  相似文献

Wang, Ke, and Brown (2003, Biometrics59, 804-812) developed a smoothing-based approach for modeling circadian rhythms with random effects. Their approach is flexible in that fixed and random covariates can affect both the amplitude and phase shift of a nonparametrically smoothed periodic function. In motivating their approach, Wang et al. stated that a simple sinusoidal function is too restrictive. In addition, they stated that "although adding harmonics can improve the fit, it is difficult to decide how many harmonics to include in the model, and the results are difficult to interpret." We disagree with the notion that harmonic models cannot be a useful tool in modeling longitudinal circadian rhythm data. In this note, we show how nonlinear mixed models with harmonic terms allow for a simple and flexible alternative to Wang et al.'s approach. We show how to choose the number of harmonics using penalized likelihood to flexibly model circadian rhythms and to estimate the effect of covariates on the rhythms. We fit harmonic models to the cortisol circadian rhythm data presented by Wang et al. to illustrate our approach. Furthermore, we evaluate the properties of our procedure with a small simulation study. The proposed parametric approach provides an alternative to Wang et al.'s semiparametric approach and has the added advantage of being easy to implement in most statistical software packages.  相似文献

无论对于自然的还是人为管理的生态系统,天气都是最重要的一种环境条件。用系统分析方法对生态系统进行的研究使天气模拟越来越成为非常必要的了。本文首先介绍了一个美国天气模拟模型(WGEN)的数学原理。它用马尔柯夫链和Γ-分布组成降水量子模型,产生逐日降水量。用弱平稳过程和调和分析为工具建立另一个子模型,产生逐日最高温度、最低温度和太阳辐射量。通过常规的统计学方法或灰靶白化方法可以估计模型参数。于是,就可由WGEN生出Alabama州天气模型ALWGEN和北京天气模型BJWGEN。考虑到篇幅有限,本文没有具体介绍上述估计方法,也只是扼要介绍了用Monte Carlo Bootstrap方法进行模型校验和验证的问题。最后,简单地讨论了天气模拟模型的应用。如IPM研究中的风险分析,生态系统管理决策,农业生态区域规划等课题,对天气模型都有现实的或潜在的需要。  相似文献

We report the imaging of tendon with Interferometric Second Harmonic Generation microscopy. We observe that the noncentrosymmetric structural organization can be maintained along the fibrillar axis over more than 150 μm, while in the transverse direction it is ~1–15 μm. Those results are explained by modeling tendon as a heterogeneous distribution of noncentrosymmetric nano‐cylinders (collagen fibrils) oriented along the fibrillar axis. The preservation of the noncentrosymmetric structural organization over multiple tens of microns reveals that tendon is made of domains in which the ratio between fibrils with positive and negative polarity is unbalanced.
