This article reports the concentrations of hexachlorocyclohexanes (HCHs) and dichloro-diphenyl-trichloroethanes (DDTs) in fresh Gentoo penguin (Pygoscelis Papua) droppings deposited on the Ardley Island (62°13′S, 58°56′W), West Antarctica, and those in the island's two lake cores. In the fresh penguin droppings, metabolites were the predominant constituents of the DDT group, most likely due to the ban on DDT use in agriculture and the higher stability of the metabolites in the environment. In contrast, γ-HCH comprised 66% of the total HCHs, apparently due to the continuous use of Lindane in the Southern Hemisphere. The concentrations of HCHs and DDTs in the fresh droppings are many times higher than those in penguin plume, the latter currently being used as biomaterial for monitoring coastal contaminations in Antarctica. Results of this study indicate that penguin droppings have a good potential for use as another biomonitoring material in detecting the organochlorine pesticides and other contaminants in the maritime Antarctic. Because penguin droppings can be well preserved in lake sediments, they also provide a good and continuous historical record of these contaminants. The slight increase of DDTs since the 1990s, as observed in the aged penguin droppings in the sediment cores, suggests there have been new inputs of DDT into the Antarctic coastal ecosystem, probably from the illegal use of DDT in the Southern Hemisphere.  相似文献

Dichlorodiphenyltrichloroethane (DDT), hexachlorocyclohexane (HCH), and their isomers’ levels in residential soils were determined for the assessment of health risk in Korba, India. Observed concentrations of total HCH and total DDT in soils were more or less comparable with other parts of India and the world. ΣHCH and ΣDDT concentrations ranged between 0.9–20 μg kg?1 and 2–315 μg kg?1, respectively, which were lower than recommended soil quality guidelines indicating low ecotoxicological risk. Carcinogenic and non-carcinogenic impacts of HCH and DDT on human populations through soil ingestion were evaluated and presented. The incremental lifetime cancer risk (ILCR) for adults and children ranged between 7.8 × 10?10–1.6 × 10?7 and 4.1 × 10?9–8.2 × 10?7, respectively. Non-cancer health hazard quotient (HQ) ranged between 5.9 × 10?7–1.8 × 10?3 and 3.1 × 10?6–9.4 × 10?3, respectively, for adults and children. The estimated ILCR and HQ were within the safe acceptable limits of 10?6–10?4 and ≤1.0, respectively, indicating low risk to human populations from exposure to organochlorine pesticides (HCH and DDT) in the study area.  相似文献

Soil pollution with hexachlorocyclohexane (HCH) has caused serious environmental problems. Here we describe the targeted degradation of all HCH isomers by applying the aerobic bacterium Sphingobium indicum B90A. In particular, we examined possibilities for large-scale cultivation of strain B90A, tested immobilization, storage and inoculation procedures, and determined the survival and HCH-degradation activity of inoculated cells in soil. Optimal growth of strain B90A was achieved in glucose-containing mineral medium and up to 65% culturability could be maintained after 60 days storage at 30°C by mixing cells with sterile dry corncob powder. B90A biomass produced in water supplemented with sugarcane molasses and immobilized on corncob powder retained 15–20% culturability after 30 days storage at 30°C, whereas full culturability was maintained when cells were stored frozen at −20°C. On the contrary, cells stored on corncob degraded γ-HCH faster than those that had been stored frozen, with between 15 and 85% of γ-HCH disappearance in microcosms within 20 h at 30°C. Soil microcosm tests at 25°C confirmed complete mineralization of [14C]-γ-HCH by corncob-immobilized strain B90A. Experiments conducted in small pits and at an HCH-contaminated agricultural site resulted in between 85 and 95% HCH degradation by strain B90A applied via corncob, depending on the type of HCH isomer and even at residual HCH concentrations. Up to 20% of the inoculated B90A cells survived under field conditions after 8 days and could be traced among other soil microorganisms by a combination of natural antibiotic resistance properties, unique pigmentation and PCR amplification of the linA genes. Neither the addition of corncob nor of corncob immobilized B90A did measurably change the microbial community structure as determined by T-RFLP analysis. Overall, these results indicate that on-site aerobic bioremediation of HCH exploiting the biodegradation activity of S. indicum B90A cells stored on corncob powder is a promising technology.  相似文献

In the soil remediation process, the hydrophobic characteristics of pollutants and their affinity for soil matrix may be responsible for mass transfer limitations. The degradation of hexachlorocyclohexane (HCH) isomers present in a spiked soil by the white-rot Bjerkandera adusta was evaluated in a slurry system. Experiments in shaken flasks were performed to evaluate the action of the endogenous microflora, the adsorption of HCH on the fungal biomass and the potential synergic or antagonic actions between the microflora and the fungal biomass. The fungus significantly degraded the HCH isomers from the soil slurry in the following order: αγ>δ>β-HCH. The degradation process was further scaled in a 5-l reactor, where the solid load and concentration of the pollutant in the soil were evaluated. At optimal conditions, 100 g soil l−1 and 100 mg total HCH l−1, maximal degradations of 94.5%, 78.5% and 66.1% were attained after 30 d for γ-, α- and δ-HCH isomers, respectively, representing between 1.7 and 3.1-fold the values obtained at small scale. These results indicate that minimising mass transfer resistances is a key factor for HCH degradation from soil.  相似文献

The levels of organochlorine pesticides (OCPs) in the environment and foods have declined significantly in China since 1983, the year when their use in agriculture was officially banned. The levels today of these contaminants in soil, water, and agricultural products are well below 50 μg/kg in many parts of China. Yet in spite of such a decline, the current levels of pollution still pose a significant health risk to many people in China because OCPs are highly persistent and bioaccumulative substances. This preliminary study was conducted to monitor the levels of hexachlorocyclohexane (HCH) and dichloro-diphenyl-trichloroethane (DDT) in human and fish fats from southeast China in an effort to corroborate the above health concern. The data reconfirmed that pollution by HCH and DDT, which are the major components of OCPs, is a serious ecological problem in the study region, and likely in other parts of China as well. The HCH contents in the human samples were between 0.25 and 2.20 mg/kg, with an average of 0.90 mg/kg. The DDT contents in these human samples were much higher, between 0.54 and 9.22 mg/kg with an average of 3.50 mg/kg. The major isomers found in the human samples were β-HCH and p, p′-DDE, representing 94.9 to 99.5% of total HCH and 72.3 to 96.7% of total DDT, respectively. The HCH and DDT content in the grass carp samples averaged 0.03 and 0.67 mg/kg, respectively. The levels of bioaccumulation reflected in these human and fish samples were considered substantial, particularly when residents in the study region continue to be exposed to the OCPs remaining so persistently in their environment. It is therefore important for the health authorities to launch a large-scale investigation into the potential health and ecological impacts of OCP pollution in China. The data also suggested that the current national standards might need to be lowered for the levels of OCPs in the environment in China.  相似文献

Background

Sphingobium spp. are efficient degraders of a wide range of chlorinated and aromatic hydrocarbons. In particular, strains which harbour the lin pathway genes mediating the degradation of hexachlorocyclohexane (HCH) isomers are of interest due to the widespread persistence of this contaminant. Here, we examined the evolution and diversification of the lin pathway under the selective pressure of HCH, by comparing the draft genomes of six newly-sequenced Sphingobium spp. (strains LL03, DS20, IP26, HDIPO4, P25 and RL3) isolated from HCH dumpsites, with three existing genomes (S. indicum B90A, S. japonicum UT26S and Sphingobium sp. SYK6).

Results

Efficient HCH degraders phylogenetically clustered in a closely related group comprising of UT26S, B90A, HDIPO4 and IP26, where HDIPO4 and IP26 were classified as subspecies with ANI value >98%. Less than 10% of the total gene content was shared among all nine strains, but among the eight HCH-associated strains, that is all except SYK6, the shared gene content jumped to nearly 25%. Genes associated with nitrogen stress response and two-component systems were found to be enriched. The strains also housed many xenobiotic degradation pathways other than HCH, despite the absence of these xenobiotics from isolation sources. Additionally, these strains, although non-motile, but posses flagellar assembly genes. While strains HDIPO4 and IP26 contained the complete set of lin genes, DS20 was entirely devoid of lin genes (except linKLMN) whereas, LL03, P25 and RL3 were identified as lin deficient strains, as they housed incomplete lin pathways. Further, in HDIPO4, linA was found as a hybrid of two natural variants i.e., linA1 and linA2 known for their different enantioselectivity.

Conclusion

The bacteria isolated from HCH dumpsites provide a natural testing ground to study variations in the lin system and their effects on degradation efficacy. Further, the diversity in the lin gene sequences and copy number, their arrangement with respect to IS6100 and evidence for potential plasmid content elucidate possible evolutionary acquisition mechanisms for this pathway. This study further opens the horizon for selection of bacterial strains for inclusion in an HCH bioremediation consortium and suggests that HDIPO4, IP26 and B90A would be appropriate candidates for inclusion.

Aim: To screen and identify bacteria from contaminated soil samples which can degrade hexachlorocyclohexane (HCH)‐isomers based on dechlorinase enzyme activity and characterize genes and metabolites. Methods and Results: Dechlorinase activity assays were used to screen bacteria from contaminated soil samples for HCH‐degrading activity. A bacterium able to grow on α‐, β‐, γ‐ and δ‐HCH as the sole carbon and energy source was identified. This bacterium was a novel species belonging to the Sphingomonas and harbour linABCDE genes similar to those found in other HCH degraders. γ‐Pentachlorocyclohexene 1,2,4‐trichlorobenzene and chlorohydroquinone were identified as metabolites. Conclusions: The study demonstrates that HCH‐degrading bacteria can be identified from large environmental sample‐based dehalogenase enzyme assay. This kind of screening is more advantageous compared to selective enrichment as it is specific and rapid and can be performed in a high‐throughput manner to screen bacteria for chlorinated compounds. Significance and Impact of the Study: The chlorinated pesticide HCH is a persistent and toxic environmental pollutant which needs to be remediated. Isolation of diverse bacterial species capable of degrading all the isomers of HCH will help in large‐scale bioremediation in various parts of the world.  相似文献

In this study, we compared the estrogenic action pattern of beta-HCH to another ErbB2 activating agent, heregulin beta1 (HRG), along with 17beta-estradiol and epidermal growth factor, to understand the similarities and differences of their action mechanisms. Not surprisingly, most of the initial test results indicated that the two agents, beta-HCH and HRG, are remarkably similar in several estrogenicity tests. However, in-depth biochemical studies revealed that there are some distinct differences between these two compounds in affecting ErbB2 and ErbB3 at early stages of their action. Immunocoprecipitation and Western blot studies indicated that beta-HCH mainly promotes dimerization of ErbB2 and ErbB3 at early time points, whereas HRG causes their dimerization with a rapid and significant rise in their tyrosine phosphorylation levels. These results indicate that, while both beta-HCH and HRG act through ErbB2, their initial actions differ. To understand the long-term consequence of such differential actions of these two agents, we tested the effect of a number of standard pathway specific inhibitors on their actions to induce foci formation after 2 weeks of exposure. The most conspicuous difference between beta-HCH and HRG in MCF-7 foci formation test was their response to 4-hydroxytamoxifen and LY294002, a PI3K inhibitor, i.e., the action of beta-HCH was inhibited by 4-hydroxytamoxifen but stimulated by LY294002, whereas that of heregulin was suppressed by LY294002 but stimulated by 4-hydroxytamoxifen. It appears, therefore, that the action of the latter relies more heavily on the PI3K route as compared to that of the former which has been shown to mainly act through p42/44 MAPK. These differences may account for their differential sensitivities to 4-hydroxytamoxifen.  相似文献

The soil contact effects of several commonly used agricultural insecticides on 2nd instar and adultForficula auricularia (L.) were studied in the laboratory. The earwigs were individually confined on soil surfaces sprayed under a track sprayer. Pirimicarb had no effect on either adult or 2nd instar earwigs. The LD50,s of 2nd instar earwigs for γ HCH, fenitrothion, DDT and dimethoate were all lower than their normal field rates. Second instars were more susceptible to dimethoate and cypermethrin than adults. The pyrethroids cypermethrin and deltamethrin had LD50,s above normal field rates. However, high levels of knockdown were induced by both insecticides at concentrations below normal application rates. The effects of this in the field remain unknown.  相似文献

Clostridium rectum strain S-17 converts -1,2,3,4,5,6-hexachlorocyclohexane (HCH) related compounds to chlorobenzenes. The metabolites from -1,2,3,4,5,6-hexachlorocyclohexene and -1,3,4,5,6-pentachlorocyclohexene are identified as 1,2,4-trichlorobenzene and 1,4-dichlorobenzene, respectively. ATP synthesis, converting these chlorinated compounds, is observed in the cell suspension of C. rectum as indicated by luciferase-luciferin reaction and phosphorylation of 32P-labeled phosphate.
