We hereby report studies that suggest a role for serum exosomes in the anchorage-independent growth (AIG) of tumor cells. In AIG assays, fetal bovine serum is one of the critical ingredients. We therefore purified exosomes from fetal bovine serum and examined their potential to promote growth of breast carcinoma cells in soft agar and Matrigel after reconstituting them into growth medium (EEM). In all the assays, viable colonies were formed only in the presence of exosomes. Some of the exosomal proteins we identified, have been documented by others and could be considered exosomal markers. Labeled purified exosomes were up-taken by the tumor cells, a process that could be competed out with excess unlabeled vesicles. Our data also suggested that once endocytosed by a cell, the exosomes could be recycled back to the conditioned medium from where they can be up-taken by other cells. We also demonstrated that low concentrations of exosomes activate MAP kinases, suggesting a mechanism by which they maintain the growth of the tumor cells in soft agar. Taken together, our data demonstrate that serum exosomes form a growth promoting platform for AIG of tumor cells and may open a new vista into cancer cell growth in vivo.  相似文献

We describe the case of a boy with psychomotor delay and dysmorphic features, with a germline 16q22.1 microdeletion identified by array-CGH. The deletion spans 0.24Mb and encompasses three genes (ZFP90, CDH3 and CDH1). The deletion has been demonstrated to be inherited from his mother who was affected by lobular breast cancer (LBC) without any other apparently phenotypic features. We suppose that the microdeletion, in particular ZFP90 which is cerebrally expressed, is causative for the boy's phenotype. Mental retardation in the affected boy can recognize several mechanisms such as variable expressivity, non-penetrance, multifactorial/polygenic inheritance, recessive inheritance, a second rearrangement event and epigenetics. Furthermore, we suggest that the deletion of the CDH1, a tumor suppressor gene, involved in hereditary diffuse gastric cancer (HDGC) and LBC predisposed the mother to the carcinoma.  相似文献

Solid surface fluorescence excitation-emission matrix (EEM) is developed a potential method to characterize soil organic matter (SOM). Solid surface EEM spectroscopy with parallel factor analysis (PARAFAC) and hierarchical cluster analysis (HCA) is used to extract fluorescent components, to seek latent factors, and to investigate spatial distribution of SOM. Soil samples were collected from four native halophyte and two furrow-irrigated soil profiles, i.e. Comm. Salicornia europaea (CSE), Comm. Suaeda glauca (CSG), Comm. Kalidium cuspidatum (CKC), Comm. Sophora alopecuroides (CSA), corn fields (CFD), and wheat fields (WFD). SOM contained six fluorescent components: microbial/terrestrial fulvic-like fluorescent components (C1), tryptophan-like/lignin-derived phenol fluorescent components (C2), terrestrial humic-like fluorescent component (C3), lignin oxidative degradation by-products (C4 and C5), and amino acids (C6). The C 4 and C5 were the representative components of SOM within the CSE, CSG, CKC, CSA and CFD soil profiles, while the C2 and C6 were dominated within the WFD soil profile. The C4, C5, C1 and C2 were latent factors, and they could roughly distinguish SOM within the whole saline soil profiles except the CFD. A humification index (H/L) deduced from the fluorescent components, was very suitable to indicate humification levels of SOM. Humification levels of SOM within the halophyte soil profiles decreased with soil depth, but the opposite trends within the furrow-irrigated soil profiles. The H/L was closely correlated with exchangeable sodium percentage (ESP), and humification levels increased with the decreasing ESP. Soil surface EEM may not only indicate organic matter fractions of saline soils, but may be transferred to other types of landscape.  相似文献

Aerobic granulation, a novel environmental biotechnological process, was increasingly drawing interest of researchers engaging in work in the area of biological wastewater treatment. Developed about one decade ago, it was exciting research work that explored beyond the limits of aerobic wastewater treatment such as treatment of high strength organic wastewaters, bioremediation of toxic aromatic pollutants including phenol, toluene, pyridine and textile dyes, removal of nitrogen, phosphate, sulphate and nuclear waste and adsorption of heavy metals. Despite this intensive research the mechanisms responsible for aerobic granulation and the strategy to expedite the formation of granular sludge, and effects of different operational and environmental factors have not yet been clearly described. This paper provides an up-to-date review on recent research development in aerobic biogranulation technology and applications in treating toxic industrial and municipal wastewaters. Factors affecting granulation, granule characterization, granulation hypotheses, effects of different operational parameters on aerobic granulation, response of aerobic granules to different environmental conditions, their applications in bioremediations, and possible future trends were delineated. The review attempts to shed light on the fundamental understanding in aerobic granulation by newly employed confocal laser scanning microscopic techniques and microscopic observations of granules.  相似文献

The Lab-to-Field (LTF) rating scheme is a straightforward method of relating the results of effluent toxicity tests to the field survey measurements and has proven to be a useful tool for interpretation of Environmental Effects Monitoring studies for pulp and paper mills in the province of Ontario. The LTF method uses the same five-level scale (level 1 for no or low response to level 5 for severe response) for rating the toxicity and field survey results. Regression analysis of LTF scores has revealed that the relationship between the Ceriodaphnia reproduction test and benthic invertebrate field survey measurements was significant (p < 0.001, r = 0.79). However, there were not sufficient data to determine if this can be used as a predictive tool. Nonetheless, Ceriodaphnia-to-benthic survey, Selenastrum-to-benthic survey and fathead-to-fish survey relationships were qualitatively rated strong or moderately strong in 94%, 75%, and 60% of the 16 studies, respectively. The LTF rating scheme would benefit from the use of a more sensitive species or life stage of fish to strengthen the sublethal test-to-fish survey relationship. Further validation of the lab-to-field relationship could be accomplished through the synoptic collection of effluent samples for sublethal tests and field measurements.  相似文献

The effects of an E-strain fungus (Wilcoxina mikolae var. mikolae) and an ectomycorrhizal fungus (Hebeloma crustuliniforme) on growth and water relations of balsam poplar were examined and compared in the present study. Balsam poplar roots inoculated with W. mikolae var. mikolae (Wm) exhibited structures consistent with ectendomycorrhizal (EEM) associations, including a mantle surrounding the outside of the root and an extensive Hartig net that was located between cortical cells and extended to the vascular cylinder. Roots colonized with H. crustuliniforme (Hc) developed a mantle layer, indicative of an ectomycorrhizal (ECM) association, around the outer part of the root, but no distinct Hartig net was present. Wm-colonized balsam poplar also showed increased shoot growth, stomatal conductance (g s), and root volumes compared with non-inoculated and Hc-inoculated plants. However, Hc-inoculated plants had higher root hydraulic conductivity (L pr) compared with non-inoculated plants and Wm-inoculated plants. These results suggest that L pr was not a growth-limiting factor in balsam poplar and that hyphal penetration of the root cortex in itself may have little influence on root hydraulic properties.  相似文献

For the purpose of evaluating the stability of municipal solid waste (MSW) excavated from a landfill, dissolved organic matter was extracted and characterized by physicochemical and spectroscopic methods. Results showed that dissolved organic carbon concentration, ratio of dissolved organic carbon to dissolved organic nitrogen, and specific ultraviolet absorbance at 254 nm were in the range of 0.383-3.502 g kg−1, 0.388-3.693 and 2.700-4.629 L mg−1 m−1, respectively, indicating the stability of MSW. Results obtained from Fourier transform infrared spectra have demonstrated that the stability of excavated MSW was characterized by disappearance of some easily biodegradable compounds; and the 1635/1406 ratio varied from 0.979 to 1.840 and was higher than that of the matured compost. The excitation-emission matrix spectra have shown that the principal components in excavated MSW comprised humic substances and the MSW was stable by the presence of a peak with wavelength pair of ∼280/420 nm.  相似文献

The formation of disinfection by-products (DBPs), including both nitrogenous DBPs (N-DBPs) and carbonaceous DBPs (C-DBPs), was investigated by analyzing chlorinated water samples following the application of three pretreatment processes: (i) powdered activated carbon (PAC) adsorption; (ii) KMnO(4) oxidation and (iii) biological contact oxidation (BCO), coupled with conventional water treatment processes. PAC adsorption can remove effectively the precursors of chloroform (42.7%), dichloroacetonitrile (28.6%), dichloroacetamide (DCAcAm) (27.2%) and trichloronitromethane (35.7%), which were higher than that pretreated by KMnO(4) oxidation and/or BCO process. The removal efficiency of dissolved organic carbon by BCO process (76.5%)--was superior to that by PAC adsorption (69.9%) and KMnO(4) oxidation (61.4%). However, BCO increased the dissolved organic nitrogen (DON) concentration which caused more N-DBPs to be formed during subsequent chlorination. Soluble microbial products including numerous DON compounds were produced in the BCO process and were observed to play an essential role in the formation of DCAcAm in particular.  相似文献

As part of the Canadian Environmental Effects Monitoring program under the National Metal Mining Effluent Regulation, there is a requirement to conduct sublethal toxicity tests twice per year for the first three years. These first three years (2003 to 2005) were considered a period of initial monitoring and resulted in test endpoints for each of the required standardized methods on a fish, an aquatic plant, an invertebrate, and an algal species. On a national level (based on 1648 valid results), the test from most to least sensitive was: the inhibition of reproduction with Ceriodaphnia dubia, the growth inhibition (frond number) with Lemna minor, the inhibition of cell yield with Pseudokirchneriella subcapitata, the growth inhibition (dry weight) with Lemna minor, the growth inhibition of fathead minnows, and the effect on embryo viability with rainbow trout. This sensitivity ranking changed when tests were further evaluated on a geographical region and mine-type basis (e.g., base metal, precious metal, uranium, iron ore). Site-specific examples show how sublethal toxicity data are being used to track changes in effluent quality, choosing a final discharge point, monitoring multiple discharges to the same watercourse, and to identify study design weaknesses by comparing laboratory results to field survey conclusions.  相似文献

The present study was conducted to characterize fluorophores in the fish body using three-dimensional fluorescence fingerprints (3D-FFs) and to utilize these 3D-FFs obtained from frozen horse mackerel (Trachurus japonicus) fillets to predict early post-mortem changes. Alive fish were sacrificed instantly, preserved in ice until 2 days, and then filleted, vacuum packed, and frozen. Subsequently, 3D-FFs of the frozen fillets were acquired using F-7000 aided with a fiber probe. Post-mortem freshness changes were tracked by measuring adenylate energy charge (AEC) values and nicotinamide adenine dinucleotide (NAD and NADH) content. Partial least squares regression models for predicting AEC values and NADH content in frozen fish meat showed good fittings, with R2 of 0.90 and 0.85, by utilizing eight and five excitation wavelengths, respectively, based on their fluorescence features acquired from standard fluorophores. This novel approach of 3D-FFs could be utilized as an efficient technique for at-line monitoring of frozen fish quality.  相似文献
