The Steinfeld in Lower Austria supports a population of European Nightjar (Caprimulgus europaeus) which was extensively studied during 1997 and 1998. The study area encompassed a pine forest of 20 km2. The population densities of 1.05 and 1.25 territories/km2, respectively, lies within the range found in central European populations. Annual monitoring until 2001 has shown the population to be stable. To gain an insight into habitat use of the species, various habitat-related parameters were measured inside and outside the territories, namely structure of trees, density of trees, structure of undergrowth vegetation and structure of clearings. Discriminant analysis was applied to assess the factors responsible for habitat choice of the Nightjar population. The findings showed that the Nightjars territories were frequently centered on a large clearing with an area of at least 0.7 ha. Clearings less than 50 m wide were not colonized. The requirement for a minimum width of a clearing in addition to a minimum area probably relates to better hunting conditions. Nightjars prefer trees where the lower edge of the crown is on average 4.38 m higher than at control points so that males can churr from dead branches immediately below the canopy. Such trees were found on the edge of clearings in the forest, and the edge of a clearing thus had a pronounced effect on the quality of a territory. In contrast to reports in the literature, neither the proportion of bare patches of ground nor the average height of undergrowth vegetation was found to be decisive for territory selection.  相似文献

Genetically modified plants, in the following referred to as genetically modified organisms or GMOs, have been commercially grown for almost two decades. In 2010 approximately 10% of the total global crop acreage was planted with GMOs (James, 2011). More than 30 countries have been growing commercial GMOs, and many more have performed field trials. Although the majority of commercial GMOs both in terms of acreage and specific events belong to the four species: soybean, maize, cotton and rapeseed, there are another 20 + species where GMOs are commercialized or in the pipeline for commercialization. The number of GMOs cultivated in field trials or for commercial production has constantly increased during this time period. So have the number of species, the number of countries involved, the diversity of novel (added) genetic elements and the global trade. All of these factors contribute to the increasing complexity of detecting and correctly identifying GMO derived material. Many jurisdictions, including the European Union (EU), legally distinguish between authorized (and therefore legal) and un-authorized (and therefore illegal) GMOs. Information about the developments, field trials, authorizations, cultivation, trade and observations made in the official GMO control laboratories in different countries around the world is often limited, despite several attempts such as the OECD BioTrack for voluntary dissemination of data. This lack of information inevitably makes it challenging to detect and identify GMOs, especially the un-authorized GMOs. The present paper reviews the state of the art technologies and approaches in light of coverage, practicability, sensitivity and limitations. Emphasis is put on exemplifying practical detection of un-authorized GMOs. Although this paper has a European (EU) bias when examples are given, the contents have global relevance.  相似文献

Abstract. A regional vegetation survey of the temperate grassy woodlands (temperate savanna) in Australia was designed to assess the effects of clearing and grazing on the composition of vegetation remnants and the adjacent pasture matrix. Vegetation was sampled across a range of habitats using 77 0.1024‐ha quadrats; the relative abundance of species was recorded. Classification analysis clustered the sites into three main groups that corresponded to intensity of grazing/clearing followed by groups based on underlying lithology (basalt, metasediment, granites). Using Canonical Correspondence Analysis, exogenous disturbance and environmental variables were related to the relative abundance of species; grazing intensity had the highest eigenvalue (0.27) followed by tree canopy cover (0.25), lithology (0.18), altitude (0.17) and slope (0.10). Based on two‐dimensional ordination scores, six species response groups were defined relating to intensity of pastoralism and nutrient status of the landscape. Abundance and dominance of native shrubs, sub‐shrubs, twiners and geophytes were strongly associated with areas of less‐intense pastoralism on low‐nutrient soils. The strongest effects on species richness were grazing followed by canopy cover. Continuously grazed sites had lower native species richness across all growth forms except native grasses. There was no indication that intermediate grazing intensities enhanced forb richness as a result of competitive release. Species richness for all native plants was lowest where trees were absent especially under grazed conditions. Canopy cover in ungrazed sites appeared to promote the co‐existence of shrubs with the herbaceous layer. Predicted declines in forb richness in treeless, ungrazed, sites were not detected. The lack of a disturbance‐mediated enhancement of the herbaceous layer was attributed to habitat heterogeneity at 0.1 ha sampling scale.  相似文献

Recently several DNA-binding fluotochromes have been used for demonstrating pollennuclei. However, the autofluorescence of pollen wall often obscured the fluorescence of nuclei, thus limited the use of this method. Methyl salicylate (MS) as a clearing agent has shownexcellent effect for observing embryo sac in whole-mounted ovules. This aroused me to trya combination of fluorescent staining with MS clearing in orded to make a better demonstration of the pollen nuclei. Mature 2-celled or 3-celled pollen of several angiosperm species stained with Hoechst 33258(H33258) and cleared (via ethanol dehydration) with MS showed clearcut fluorescence oftheir generative or sperm nuclei and vegetative nucleus. MS greatly decreased the wall fluorescence and increased the transparency of the pollen contents, meanwhile maintained the H33258stained fluorescence, consequently made the nuclei brighter under a darkened background. For example, in sunflower pollen a pair of elongated and winding sperm nuclei whichcould not be identified after simple H33258 staining were quite visible after MS clearing, inartificially germinated pollen tubes, the locomotion of nuclei from pollen grain into the tube,the sequence of generative and vegetative nucle travelling along the tube and the division of generative nucleus into two sperm nuclei could be well followed by this method. The present technique may be adoptable for observations on the processes of microsporogenesis and male gametophyte development, and rogenesis in cultured anthers, and also possiblyfor tracing the nuclear events during pollination-fertilization.  相似文献

摘要 目的:探讨清热化痰、宣肺解痉法治疗支气管哮喘的临床疗效及对气道呼吸参数、生活质量的影响。方法:选取我院2019年10月到2021年9月收治的60例支气管哮喘患者作为研究对象,分为观察组与对照组,每组30例。对照组予吸入布地奈德福莫特罗粉吸入剂,观察组在对照组基础上增加清热化痰宣肺解痉法治疗,对比两组患者临床疗效,治疗前与治疗1个月后的中医证候积分变化,气道呼吸参数变化,炎症因子以及生活质量变化。结果:观察组总有效率较对照组高(P<0.05);两组患者治疗前中医证候积分对比无差异(P>0.05),治疗后降低,且观察组低于对照组(P<0.05);两组患者治疗前第1秒用力呼气容积(FEV1)、用力肺活量(FVC)、第1s用力呼气量/用力肺活量(FEV1/FVC)最大呼气流速(PEF)对比无明显差异(P>0.05),治疗后两组患者FVC、FEV1、FEV1/ FVC、PEF均有提升,且观察组高于对照组(P<0.05);两组患者治疗前肿瘤坏死因子-α(TNF-α)、转化生长因子-β1(TGF-β1)、基质金属蛋白-9(MMP-9)、血管内皮生长因子(VEGF)、干扰素-γ(IFN-γ)、白细胞介素-4(IL-4)水平对比无明显差异(P>0.05),治疗后两组患者TNF-α、TGF-β1、MMP-9、VEGF、IFN-γ、IL-4水平均明显降低,且观察组低于对照组(P<0.05);两组患者治疗前生活质量评分对比无差异(P>0.05),治疗后1个月生活质量相关评分均降低,且观察组较对照组低(P<0.05)。结论:支气管哮喘患者在常规西医治疗基础上增加清热化痰宣肺解痉法治疗可减轻患者症状。另外可改善患者呼吸功能,降低炎症因子反应,提升患者生活质量。  相似文献

A new stain-clearing procedure has been developed for embryological observations on whole mounted specimens. Ovules of Helianthus annus and Nicotiana tabacum as well as ovaries of Oryza sativa were stained with diluted Ehrlich's hematoxylin for a proper short time, followed by steps of washing and dehydration, and finally cleared and mounted in methyl salicylate. When observed by ordinary bright-field microscopy, the embryo sacs before fertilization and the embryos and endosperms after fertilization were clearly visible. The gynogenic embryoids induced in unpollinated rice ovaries in vitro were also finely detectable. The Ehrlich's hematoxylin-methyl salicylate technique has the merits of rapidity in specimen preparation, high contrast and three dimensional view, needlessness of phase- or interference-contrast equipment, and the feasibility for a wide range of materials. The special significance of this technique for in vitro embryological studies is emphasized.  相似文献

Toxoplasma gondii is an obligate, intracellular parasite with a broad host range, including humans and rodents. In both humans and rodents, Toxoplasma establishes a lifelong persistent infection in the brain. While this brain infection is asymptomatic in most immunocompetent people, in the developing fetus or immunocompromised individuals such as acquired immune deficiency syndrome (AIDS) patients, this predilection for and persistence in the brain can lead to devastating neurologic disease. Thus, it is clear that the brain-Toxoplasma interaction is critical to the symptomatic disease produced by Toxoplasma, yet we have little understanding of the cellular or molecular interaction between cells of the central nervous system (CNS) and the parasite. In the mouse model of CNS toxoplasmosis it has been known for over 30 years that neurons are the cells in which the parasite persists, but little information is available about which part of the neuron is generally infected (soma, dendrite, axon) and if this cellular relationship changes between strains. In part, this lack is secondary to the difficulty of imaging and visualizing whole infected neurons from an animal. Such images would typically require serial sectioning and stitching of tissue imaged by electron microscopy or confocal microscopy after immunostaining. By combining several techniques, the method described here enables the use of thick sections (160 µm) to identify and image whole cells that contain cysts, allowing three-dimensional visualization and analysis of individual, chronically infected neurons without the need for immunostaining, electron microscopy, or serial sectioning and stitching. Using this technique, we can begin to understand the cellular relationship between the parasite and the infected neuron.  相似文献

Question: What is the effect of fuel management practices in the recovery capacity of seeder‐dominated shrublands? Location: Ulex parviflorus shrubland localities in Mediterranean regions of eastern Spain. Methods: We applied prescribed burning and brush‐chipping as fuel management techniques in three young and three mature shrublands, and evaluated the effects in the following four years. Results: Canopy opening by the treatments allowed increasing species richness through the four years of secondary succession. The treatments produced a change in community structure and dominant species, from the woody seeder Ulex parviflorus to the resprouter grass Brachypodium retusum. Vegetation response was conditioned by both shrubland developmental stage and treatment applied. Burning resulted in more severe modification of the ecosystem, increasing bare soil cover. Four years after fuel management in different aged Mediterranean gorse shrublands, vegetation response followed a similar pattern with the exception of the young, brush‐chipped shrublands. The treatments applied for controlling Ulex parviflorus were seen to be very effective, with the exception of brush‐chipping in young shrublands. Conclusions: Selective brush‐chipping in middle‐aged or mature gorse shrublands would combine a drastic reduction in fire hazard with ecosystem conservation and regeneration.  相似文献
