We studied the effect of long-term nitrogen fertilisation on wood chemistry at two boreal sites in Finland: the northern site (Kemijärvi) and the southern site (Heinola). N-fertilisation was repeated in five-year intervals from the 1960s. Norway spruce (Picea abies L. Karst.) trees that had been planted in 1938 and 1954, in the northern and the southern site, respectively, were harvested in October 2002. Altogether 20 trees, in five different size classes, either unfertilised or fertilised, were felled. Wood sections at breast height, consisting of five consecutive annual rings, from six (Kemijärvi) or five (Heinola) points with different distances from the pith were examined. Differences in growth between the northern and southern sites were marked in favour of the southern site. In the northern site fertilisation had clearly increased the diameter growth, while in the southern site fertilisation had no effect. Nitrogen fertilisation resulted in slight changes in wood chemistry that included increased nitrogen concentrations in the northern site and extractives in the southern site. Stem wood had higher concentrations of extractives, starch, and uronic acids, and lower concentration of cellulose, in the northern than in the southern site. Changes in the stem wood chemistry along radial axis were marked. The changes in wood chemistry are discussed in relation to the physiological function and also how the changes can influence the suitability of wood for different end-use purposes. 相似文献

Cellobiose hydrolysis by β-d-glucosidase (β-d-glucoside glucohydrolase EC 3.2.1.21) can become the rate-limiting step in the hydrolysis of cellulosic wastes because of inhibition phenomena involving other enzymes of the cellulase complex. Enhancement of the overall rate can therefore be obtained by increasing the amount of β-d-glucosidase present in the reactor. Unfortunately, the thermal stability of β-d-glucosidase is rather poor compared to and cellobiohydrolase. A novel stabilization method is proposed that exploits the polarization phenomena that take place in an unstirred ultrafiltration membrane enzymatic reactor. As much as a 20-fold increase in half-life compared to the native enzyme is obtained by injecting small amounts of hydroxyethyl cellulose into the system. The interactions of the cellulose molecule with several anions, including acetate , alkyl phosphate, tetrafluoroborate and hexafluorophosphate anions which are most commonly involved in the imidazolium ionic liquids (ILs), have been studied by performing density functional theory calculations. Based on calculated geometries, energies, IR characteristics, and electronic properties of the cellulose-anion complexes, it is found that the strength of interactions of anions with cellulose follows the order: acetate anion > alkyl phosphate anion > tetrafluoroborate anion > hexafluorophosphate anion, which is consistent with the experimentally observed solubility trend of cellulose in the corresponding imidazolium-based ILs. The present study may provide basic aids to some extent for understanding the dissolution behavior of cellulose in the imidazolium-based ILs. 相似文献

A brief review is given of the changing views over the years, as knowledge of wall structure has developed, concerning the mechanism whereby cellulose chains may be oriented. This leads to an examination of current concepts, particularly those concerning microtubules. It is shown that none of the mechanisms suggested whereby microtubules might cause orientation of cellulose microfibrils is consistent with the known range of molecular architectures found in plant cell walls. It is further concluded that any mechanism which necessitates an indissoluble link between the plasmalemma and the cellulose-synthesising complex at the tip of a microfibril is unacceptable. A new proposal is presented in which it is speculated that both microtubules and microfibrils are oriented by a mechanism separate from both. It is shown that if two vectors are contemplated, one parallel to cell length and one at right angles, and a sensor exists on the plasmalemma surface which responds to changes in the vectors, then all known wall structures may be explained. β-Glucosidases activated by glucose and xylose are uncommon yet intriguing enzymes that may enhance cellulose saccharification efficiency, and are of interest for application in bioethanol production processes. The molecular mechanisms of activation are completely unknown, and the aim of this study was the kinetic and biophysical characterization of the stimulation of a β-glucosidase from Humicola insolens by glucose and xylose. The effects of the monosaccharides were concentration dependent, where in a stimulatory range (0.1–50 mmol L−1), the activity increased up to 2-fold; in a stimulatory-inhibitory range (50–450 mmol L−1 glucose or 50–730 mmol L−1 xylose), the enzyme continued to be stimulated, but the activity was lower than maximal. Above 450 mmol L−1 glucose or 730 mmol L−1 xylose, increasing inhibition occurred. Dynamic light scattering confirmed that the enzyme is monomeric (54 kDa) and kinetic, intrinsic tryptophan fluorescence emission and far ultraviolet circular dichroism analyses indicated that the enzyme possesses a catalytic site (CS) and a modulator binding site (MS). Glucose or xylose binding to the MS induces conformational changes that stimulate the catalytic activity at the CS. Glucose and xylose may compete with the substrate for the CS while the substrate competes with the monosaccharides for binding to the MS. The stimulation of the enzymatic activity by glucose and xylose, which compete for the same sites on the enzyme molecule, is not synergistic. These data reveal allosteric interactions between the MS and the CS in H. insolens β-glucosidase that result in fine modulation of the catalytic activity by the monosaccharides. A kinetic model was developed that accurately described the experimental data for enzyme stimulation by glucose and/or xylose. Understanding the regulatory mechanisms of the enzyme activity, with the aid of kinetic models, may be useful for the application of the enzyme in cellulose hydrolysis processes. 相似文献

Experiments with dyed and undyed cotton, using different cellulose concentration and with or with out mechanical action showed a high effect of those factors. The process with mechanical action have a higher weight-loss. Dyed cotton with vat and sulfur dyes have almost die same weight-loss than undyed cotton, but on reactive dyed cotton the cellulase hydrolysis is shorter. It was also verified a decrease of hydrolysis extent with an increase of the reactive dye concentration on the cotton substrate. Cellulolytic bacteria became established 12 days after birth in the caecum and colon of conventionally-reared mice fed a diet containing 5 p. 100 crude cellulose (Weende). Their population reached a level between 10(6) and 10(7) bacteria per gram of digestive contents in 25-day-old animals. However, variations between animals were very large; 20 to 50% of the individuals were free of cellulolytic bacteria. A low cellulolytic population was observed in adult mice fed a cellulose-free diet. The amount of cellulose in the diet and its nature (crude or pure cellulose) affected the number of cellulolytic bacteria: the higher the percentage of cellulose in the diet, the higher the number of cellulolytic bacteria, in particular with crude cellulose-containing diet. 相似文献
