Immunochemical techniques were used to investigate the protein-DNA crosslinking by ultraviolet (UV) and gamma radiation as well as 1,3-bis-(2-chloroethyl)-1-nitrosourea (BCNU) or cis- and trans-diamminedichloroplatinum II (cis-DDP and trans-DDP). Antisera to 0.35 M NaCl extract and 0.35 M NaCl residue of HeLa nuclei were employed. Both gamma and UV irradiation, exposure to cis- or trans-DDP and, to a lesser extent, BCNU, resulted in crosslinking of various antigens to the DNA. Although several antigens were crosslinked by all the employed agents, other exhibited agent-specific crosslinking patterns.  相似文献

Nuclear counts determined by crystal violet staining from samples of stationary or microcarrier cultures of hybridomas, CHO or Vero cells were consistently and significantly higher than cell concentrations determined by the trypan blue or Coulter counter methods. This difference was attributed to the presence of a significant proportion of binucleated cells, which are assumed to be 35% of the cell population in the stationary phase of Vero cultures. The proportion of such cells during exponential growth was variable. However, continuous sub-culture of these cells induced a degree of synchrony during growth which resulted in a cyclic variation of the difference between the cell and nuclei counting techniques. This data indicates that care should be taken in interpreting cell culture profiles based solely on crystal violet nuclei staining counts.  相似文献

Understanding the ecological role of species with overlapping distributions is central to inform ecosystem management. Here we describe the diet, trophic level and habitat use of three sympatric stingrays, Hypanus guttatus, H. marianae and H. berthalutzae, through combined stomach content and stable isotope (δ13C and δ15N) analyses. Our integrated approach revealed that H. guttatus is a mesopredator that feeds on a diverse diet of benthic and epibenthic marine and estuarine organisms, principally bivalve molluscs, Alpheus shrimp and teleost fishes. Isotopic data supported movement of this species between marine and estuarine environments. H. berthalutzae is also a marine generalist feeder, but feeds primarily on teleost fishes and cephalopods, and consequently occupies a higher trophic level. In contrast, H. marianae is a mesopredator specialized on shrimps and polychaetas occurring only in the marine environment and occupying a low niche breadth. While niche overlap occurred, the three stingrays utilized the same prey resources at different rates and occupied distinct trophic niches, potentially limiting competition for resources and promoting coexistence. These combined data demonstrate that these three mesopredators perform different ecological roles in the ecosystems they occupy, limiting functional redundancy.  相似文献

ABSTRACT

The identities of 18 dyes whose names begin with “Victoria” are described using their chemical structures, names and numerical identifiers. All are synthetic dyes originally synthesized in Germany during the late 19th century as colorants for textiles. Brief manufacturing details are included. All the colors of the rainbow are represented except indigo. Unusual properties including explosive tendency or toxicity are noted. Some of the applications as stains and for food coloring, anti-obesity medication and pigments for ball pen inks also are discussed  相似文献

Waste sugar beet pulp has been used as adsorbent for the removal of a hazardous cationic dye, Basic violet 16, from its aqueous solution. Adsorption of the dye was studied as function of time, pH of the solution, dosage of the adsorbent, sieve size of the particles, concentration of the dye, and temperature. The initial pH of the dye solution did not affect the chemistry of the dye molecule and the surface of beet pulp. Langmuir and Freundlich adsorption isotherms were successfully employed, and on the basis of these models, the thermodynamic parameters were evaluated. Adsorption of Basic violet 16 on beet pulp was found to be an exothermic reaction. Time contact studies showed that more than 80% adsorption of the dye is achieved in less than 1 h. Kinetics investigations confirmed both pseudo-first-order and pseudo-second-order behaviors; on the other hand, it shows that the intraparticle diffusion step is not the only rate-controlling step in all concentrations.  相似文献

We collected 729 Hypanus guttatus from the northern coast of the state of Rio Grande do Norte (RN), of which 196 were used to estimate age and growth. Ninety-five were male (12.7 to 57.0 cm disc width; WD) and 101 were female (13.0 to 88.5 cm WD); females were significantly larger than males. Cross sections of vertebrae showed band-pairs ranging from 0 to > 14 in females and from 0 to 9 in males. New-borns presented an opaque edge at birth in vertebrae without a birthmark. The average percentage of error (APE; %E) for the entire sample provided evidence that ages were repeatable. The mean monthly marginal increment (IM) indicates annual band-pair formation from August to November. The annual cycle model for one band-pair deposition provided the best fit to data based on the AIC, with peaks between August and October, similar to that found in the IM analysis, suggesting an annual formation pattern. A multi-model approach that included four models based on the observed mean WD at age indicated a modified von Bertalanffy growth model as the best for describing the species growth: W0 (WD at birth) = 14.6 cm for both sexes; females W = 98.61 cm (95% CI = 87.34–114.61 cm); k = 0.112 year−1 (CI = 0.086–0.148 year−1); males W = 60.22 cm (CI = 55.66–65.35 cm); k = 0.219 year−1 (CI = 0.185–0.276 year−1). The age-at-maturity in males and females is 5 years and 7 years, respectively. The age composition shows that most (84%) specimens were aged 0 to 2 years. The information provided here is essential for analytical assessments of H. guttatus, which is subject to significant fishing pressure mainly on new-borns and juveniles.  相似文献

In order to observe the cytotoxicity of Acanthamoeba spp., which were isolated from contact lens containers as ethiological agents for the probable amoebic keratitis in Korea, the crystal violet staining method and LDH release assay were carried out. In the crystal violet staining method, among eight contact lens container isolates, isolate 3 (Acanthamoeba KA/LS5) showed 83.6% and 81.8% of cytotoxicity, and isolate 7 (Acanthamoeba KA/LS37) showed 28.2% and 25.1% of cytotoxicity, in 1 mg/ml and 0.5 mg/ml lysate treatments, respectively. Acanthamoeba culbertsoni and A. healyi showed 84.0% and 82.8% of cytotoxicity. Similar results were observed in A. castellanii and A. hatchetti which showed 83.6% and 75.5% of cytotoxicity. Acanthamoeba royreba and A. polyphaga showed 9.0% and 1.7% of cytotoxicity. In the LDH release assay, isolate 3 (20.4%) showed higher cytotoxicity than other isolates in 1 mg/ml lysate treatment. The results provide that at least isolate 3 has the cytotoxic effect against CHO cells and seems to be the pathogenic strain.  相似文献

A simple pH-induced electrochemical switch based on an i-motif structure is developed by using crystal violet as a selective electrochemical probe for the i-motif structure. Thiol-modified cytosine-rich single-strand oligonucleotide (C-rich ssDNA) can be self-assembled on the gold electrode surface via gold–sulfur interaction. Crystal violet is employed as an electrochemical probe for the i-motif structure because of its capability of binding with the i-motif structure through an end-stacking mode. In acidic aqueous solution, crystal violet may approach the electrode surface owing to the formation of the i-motif structure, resulting in an obvious signal, so-called “ON” state. Whereas in neutral or basic aqueous solution, the i-motif structure unfolds to dissociative single strand, which causes crystal violet to leave from the electrode surface, and a weak signal is obtained, so-called “OFF” state. In addition, in the range of pH 4.6–7.3, the increase in current has a good linear relationship (R = 0.989) with pH value in the testing solutions. This pH-driven electrochemical switch has the advantages of simplicity, sensitivity, high selectivity, and good reversibility. The potential of spent biomass of a hydrogen producing cyanobacterial strain Nostoc linckia from a hydrogen fermentor was studied for decolorization of a tri-phenylmethane dye, crystal violet. The waste cyanobacterial biomass immobilized in calcium alginate was used as a biosorbent and the process variables were optimized for maximum dye removal using the statistical response surface methodology (RSM). Batch mode experiments were performed to determine the kinetic behavior of the dye in aqueous solution allowing the computation of kinetic parameters. Influence of interacting parameters like temperature (25-35 °C), pH (4-8), initial dye concentration (100-200 mg/L) and cyanobacterial dose (0.2-0.4 g) on dye removal were examined using central composite design (CCD) which included two additional levels for each parameter. Second-order polynomial regression model, was applied which was statistically validated using analysis of variance. Ability of the immobilized biomass to decolorize the dye was maximum (72%) at pH 8.0, temperature 35 °C, 200 mg/L initial dye concentration and 0.2 g cyanobacterial dose. Adsorption of the dye on cell surface was further confirmed by scanning electron micrographs of the biomass before and after dye loading. FT-IR studies revealed that decolorization was due to biosorption mediated mainly by functional groups like hydroxyl, amide, carboxylate, methyl and methylene groups present on the cell surface.  相似文献
