According to biochemical and physiological characters, 77 strains ofChlorella were assigned to 12 taxa. The characters used are presence or absence of hydrogenase, formation of secondary carotenoids under nitrogen-deficient conditions, liquefaction of gelatin, products of glucose fermentation, ability to reduce nitrate, thiamine requirement, acid tolerance, salt tolerance, thermophily, and base composition of DNA (GC content). On the other hand, certain nutritional characters, i. e., utilization of organic carbon and nitrogen compounds, were found to be more or less strain-specific, highly variable, and therefore unsuitable for taxonomy.Presented at the symposium Speciation and the Species Concept during the XIIth International Botanical Congress, Leningrad, July 8, 1975.  相似文献

Aim To develop a new method for bioclimate mapping where the vegetation layer is the main source of climate information. Location The study area includes four subareas, all situated on the Varangerhalvøya peninsula in Finnmark, north‐easternmost Norway (70–71° N). The four subareas were chosen to represent most of the climatic, topographic, geomorphologic and botanic diversity along the arctic–boreal gradient in the area. The four meteorological stations in the area show a climatic gradient with mean July temperature ranging from 10.1 to 12.3 °C. Methods The new vegetation‐based method is based on the fact that most plant species and plant communities both in the Arctic and adjacent areas have a distribution pattern limited by temperature to some extent. The vegetation is mapped using Landsat TM data and a contextual correction process in a geographic information system. The mapped vegetation units are defined as temperature indicators based on their total distribution patterns and the temperature indicator value of their high frequency and dominant species. The indicator value and degree of cover of all thermophilous vegetation units, within each 500 × 500 m study unit, are combined in a Vegetation‐based Index of Thermophily, VItm. This new vegetation‐based method is based on the same basic idea as a recently published floristic‐based method for calculating a Floristic‐based Index of Thermophily, FItm. The VItm values are tested by comparison with the FItm values, and temperature data collected in the field during two growing seasons, and the differences are interpreted ecologically. Results Twenty‐one of the mapped vegetation units were defined as thermophilous and categorized in five groups of temperature indicators. The VItm values showed a strong positive linear relationship with the temperatures measured during the years 2001 and 2002, with r2 values of 0.79 and 0.85, respectively. The VItm values show a high linear relationship (r2 = 0.76) with the 71 study units where the FItm values were calculated. As interpreted from the relationship with temperature measurements and FItm values, the vegetation‐based method seems to work at a broad range of ecological conditions, with very dry, acidic sites being the most important exception. The VItm values are related to growing degree‐days of a normal year, and the four subareas are mapped, showing a diversity of 13 bioclimatic classes. The birch forest line is estimated to occur at about 980 °C‐days. The results show climatic gradients with temperatures increasing from the cold coast towards the interior, from wind‐exposed convex hills towards wind‐protected valleys, and from mountain plateaux towards south‐facing lowlands. The north‐easternmost study site at the coast is positioned within the arctic shrub tundra zone. Main conclusions The vegetation‐based method shows a strong positive correlation both with measured temperatures and the floristic‐based method within a broad range of different ecological conditions. The vegetation‐based method has the potential for bioclimatic mapping of large areas in a cost‐effective way. The floristic‐based method has higher accuracy and is more flexible than the vegetation‐based method, and the two methods seem to complement each other.  相似文献

In brown semidesert soil, thermophilic prokaryotic organisms identified as Streptomyces roseolilacinus and Silanimonas lenta were shown to play the main role in chitin transformation at 50°C. The phylogenetic positions of the isolated dominant chitinolytic microorganisms were determined on the basis of 16S rRNA gene sequencing. The consumption of chitin as a source of carbon and nitrogen by both the bacterium and the actinomycete was evident from considerable biomass accumulation, high emission of carbon dioxide, and presence in the medium of the chitinase exoenzyme.  相似文献

An extremely thermophilic (optimum growth at 88° C), anaerobic bacterium was isolated from a shallow submarine thermal spring. It appears to be an obligate heterotroph, capable of reducing sulfur to H2S. Oxygen sensitivity is apparent only at and above those temperatures where growth occurs, while the cultures retain their viability for long periods under air at 4° C. Insensitivity to chloramphenicol, vancomycin and streptomycin, and lack of muramic acid in its cell wall, indicate a possible affilitation of the new isolate to the thermoacidophilic archaebacteria. Cream‐colored streamers of Thermothrix thiopara were found at the sulfide‐oxygen interfaces of active tufa mounds where reducing geothermal groundwaters mixed with the oxidizing atmosphere. In the Jemez hot springs, the molar ratio of sulfide to oxygen was 0.3 to 0.8 at streamer locations within the interface. This corresponded to the optimum stoichiometric proportion (0.5) necessary for sulfur metabolism. The mechanism of cell positioning at the interface was studied by shifting the interface location with a plastic cover to extend reducing conditions from the mouth of the spring to the edge of the plastic. Macroscopically visible streamers of filamentous cells became established at the new interface within a period of eight days. They could then be reestablished at the original interface by removing the cover.

Calcite crystals, pyrite crystals, and membrane enrichment vials were incubated on both sides of the interface and the kinetics of colonization determined. The preferential attachment of rod‐shaped cells to pyrite appeared to be the mechanism by which cells located themselves where pyrite occurred in situ, upstream from the interface. The formation of filamentous cells from rod‐shaped cells was induced by oxygen‐limited growth conditions. This moved the cells slightly downstream and directly within the interface.  相似文献

Reverse gyrase, an enzyme of uncertain funtion, is present in all hyperthermophilic archaea and bacteria. Previous phylogenetic studies have suggested that the gene for reverse gyrase has an archaeal origin and was transferred laterally (LGT) to the ancestors of the two bacterial hyperthermophilic phyla, Thermotogales and Aquificales. Here, we performed an in-depth analysis of the evolutionary history of reverse gyrase in light of genomic progress. We found genes coding for reverse gyrase in the genomes of several thermophilic bacteria that belong to phyla other than Aquificales and Thermotogales. Several of these bacteria are not, strictly speaking, hyperthermophiles because their reported optimal growth temperatures are below 80 degrees C. Furthermore, we detected a reverse gyrase gene in the sequence of the large plasmid of Thermus thermophilus strain HB8, suggesting a possible mechanism of transfer to the T. thermophilus strain HB8 involving plasmids and transposases. The archaeal part of the reverse gyrase tree is congruent with recent phylogenies of the archaeal domain based on ribosomal proteins or RNA polymerase subunits. Although poorly resolved, the complete reverse gyrase phylogeny suggests an ancient acquisition of the gene by bacteria via one or two LGT events, followed by its secondary distribution by LGT within bacteria. Finally, several genes of archaeal origin located in proximity to the reverse gyrase gene in bacterial genomes have bacterial homologues mostly in thermophiles or hyperthermophiles, raising the possibility that they were co-transferred with the reverse gyrase gene. Our new analysis of the reverse gyrase history strengthens the hypothesis that the acquisition of reverse gyrase may have been a crucial evolutionary step in the adaptation of bacteria to high-temperature environments. However, it also questions the role of this enzyme in thermophilic bacteria and the selective advantage its presence could provide.  相似文献

Three features of the heat shock response, reorganization of protein expression, intracellular accumulation of trehalose, and alteration in unsaturation degree of fatty acids were investigated in the thermophilic fungus Chaetomium thermophile and compared to the response displayed by a closely related mesophilic species, C. brasiliense. Thermophilic heat shock response paralleled the mesophilic response in many respects like (i) the temperature difference observed between normothermia and the upper limit of translational activity, (ii) the transient nature of the heat shock response at the level of protein expression including both the induction of heat shock proteins (HSPs) as well as the repression of housekeeping proteins, (iii) the presence of representatives of high-molecular-weight HSPs families, (iv) intracellular accumulation of trehalose, and finally (v) modifications in fatty acid composition. On the other hand, a great variability between the two organisms was observed for the proteins expressed during stress, in particular a protein of the HSP60 family that was only observed in C. thermophile. This peptide was also present constitutively at normal temperature and may thus fulfil thermophilic functions. It is shown that accumulation of trehalose does not play a part in thermophily but is only a stress response. C. thermophile contains less polyunsaturated fatty acids at normal temperature than C. brasiliense, a fact that can be directly related to thermophily. When subjected to heat stress, both organisms tended to accumulate shorter and less unsaturated fatty acids.  相似文献

Many ectothermic vertebrates raise their preferred body temperature after feeding, likely expediting digestion. However, most studies documenting this phenomenon have relied upon laboratory thermal gradients, which grossly oversimplify an animal's environment. We explored the validity of thermal gradient methodology by investigating post-prandial thermophily in an Australian pygopodid lizard (Lialis burtonis Gray). Mean body temperatures did not differ between fed and unfed lizards in field enclosures. Feeding influenced body temperature in a thermal gradient, but in opposite directions depending on details of the methodology. When we introduced L. burtonis into the gradient at the warmer end, fed lizards had higher body temperatures than unfed conspecifics. However, the opposite was true when lizards were introduced at the cooler end. These contrasting results indicate that lizards with food in their stomachs did not seek out higher temperatures, but instead were more sedentary than unfed lizards. Our study highlights the need for caution in interpreting thermal gradient results unaccompanied by field data, and it demonstrates how minor changes in equipment design or procedures can significantly alter conclusions from laboratory studies.  相似文献

In the presence of a vertical thermal gradient, juvenile three‐spined sticklebacks Gasterosteus aculeatus and minnows Phoxinus phoxinus positioned themselves higher in the water column compared with adult conspecifics. This result was consistent regardless of whether age cohorts were tested separately or together. Furthermore, juveniles but not adult fishes positioned themselves higher in water column in the presence of a thermal gradient compared with those in the absence of a thermal gradient. Juvenile G. aculeatus and adult fish of both species did opt to position themselves higher in the water column in the hours immediately following a feeding event relative to their positions in the same gradient when they had not fed.  相似文献
