Abstract

Selenate and selenite are the most prevalent bioavailable selenium (Se) forms and most easily taken up by plants. Some studies indicate that they are differently absorbed and accumulated in plants and that selenium is toxic if accumulated at high concentrations. Toxicity is due to substitution of sulphur by selenium in cysteine and methionine aminoacids with alteration of the tertiary structure and catalytic activity of proteins and with inhibition of enzymes involved in chlorophyll biosynthesis. Moreover, the interaction between Se and thiol groups induces loss of efficiency of plant defence systems and increases the reactive oxygen species (ROS) production thus enhancing the oxidative stress. To further elucidate the role of Se in higher plants, in this study the antioxidative response to the phytotoxicity of selenite and selenate in Senecio scandens L. was evaluated. The data indicate that while selenite induces oxidative stress enhancing ROS production, lipid peroxidation and the oxidised forms of ascorbate and glutathione, selenate does not significantly affect the analysed pathways. This article outlines that the synergistic action of different antioxidant components is necessary to overcome the phytotoxicity of selenium in Senecio.  相似文献

Five isoflavone glycosides, named derriscandenosides A–E (1–5), were isolated from the stems of Derris scandens, together with ten known compounds comprising one isoflavone, two benzoic acid derivatives, three glucosyl isoflavones and four rhamnosyl-(1→6)-glucosyl isoflavones. The structures of the glycosides were assigned on the basis of spectroscopic data, especially of the acetate derivatives. Three known rhamnosyl-(1→6)-glucosyl isoflavones isolated from a crude fraction were retested for hypotensive activity with varying results.  相似文献

应用倍比稀释法检测18份千里光(Senecio scandens Buch-Ham)水煎剂对7种细菌的抗菌效果。最低抑菌浓度(minimal inhibitory concentration,MIC)检测结果表明,千里光对金黄色葡萄球菌的抗菌作用最强,对铜绿假单胞菌的抗菌效果最差。比较不同细菌的MIC值发现,温和气单胞菌共检测出4个不同的MIC值,金黄色葡萄球菌、铜绿假单胞菌、大肠埃希菌、甲型副伤寒杆菌、福氏痢疾杆菌和迟钝爱德华菌的MIC值为5个,迟钝爱德华菌的MIC值最多,表现出6个MIC梯度值。本研究结果提示千里光个体有效抗菌组分的多样性,也意味着千里光抗菌药理学性状为多基因控制的数量性状。  相似文献

Total proteins in the pollen of Humulus scandens Lour,one of the most popular aeroallergens in China,were analyzed by two-dimensional electrophoresis in the current study.The proteins were extracted by Trichloracetic acid (TCA) method,and then separated by isoelectric focusing as the first dimension and SDS-PAGE as the second dimension.The spots of proteins were visualized by staining with Coomassie Brilliant Blue.After analysis with software (ImageMaster 2D),122 different proteins were detected;isoelectric point (pI),Molecular weight (MW) and relativevolume of each protein in the pollen were also discovered.This is the first high-resolution,two-dimensional protein map of the pollen ofHumulus scandens Lour in China.Our finding has built a solid foundation for identification,characterization,gene cloning and standardization of allergenic proteins in the pollen ofHumulus scandens Lour for further studies.  相似文献

千里光(Senecio scandens Buch.-Ham.ex D.Don)是具有良好开发前景的传统抗菌中草药。为了探讨该物种有性生殖的细胞学机制,实现在物种的遗传育种过程中的指导作用,本研究从细胞学角度观察了千里光配子体发育、配子形成和双受精作用。结果表明,在生殖核分裂为精细胞的过程中,其位置和形态呈现差异,表现为"二态"精细胞;此外,雌配子体的形成符合孢子发育模式,成熟胚囊的形态结构为卵形或梨形,成熟的卵细胞和极核位于珠孔端。根据上述结果可推断,合子在随后二分裂时表现为"基细胞-顶细胞"极性,可能与"二态"精细胞导致的合子细胞质不均一有关。此外,从植物分类学角度,千里光的"二态"精细胞,胚囊极性和合子极性可成为菊科近缘植物分类的细胞学依据。  相似文献

Introduction  – Two recent mass spectrometry‐based reports concerning Senecio scandens yielded remarkably dissimilar pyrrolizidine alkaloid constituents. In both studies, and in a related analysis of Senecio scandens and Tussilago farfara using micellar electrokinetic chromatography, the presence of hazardous N‐oxides of the alkaloids was either not considered or was inadequately considered. This raises concerns about the effectiveness of the methodologies used in these, and similar, studies in assessing the pyrrolizidine alkaloid content and the safety of food, food supplements and medicines for human use. Objective  – To highlight essential analytical requirements for confident assessment of pyrrolizidine alkaloid‐related safety of food and herbal products for human use. Methodology  – Direct infusion‐ESI MS and HPLC‐ESI MS were used to analyse samples derived from liquid–liquid partitioning experiments and from strong cation exchange, solid‐phase extraction of pyrrolizidine alkaloids and their N‐oxides. Results  – A simple solvent partitioning experiment using pure senecionine and senecionine‐N‐oxide, two constituents reported in one of the mass spectrometry‐based studies of S. scandens, clearly demonstrated the inadequacy of the reported method to detect and quantitate hazardous pyrrolizidine alkaloid N‐oxide components. A preliminary LCMS analysis of commercially‐prepared extracts of comfrey roots (Symphytum officinale and S. uplandicum s. l.) was used as a model to highlight the analytical importance of N‐oxides in the safety assessment of pyrrolizidine alkaloid‐containing medicinal herbs. Conclusions  – This study highlighted significant differences in the reported identification of pyrrolizidine alkaloids from the same plant species, and clearly demonstrated the inadequacy of some procedures to include N‐oxides in the assessment of pyrrolizidine alkaloid‐related safety of food and herbal products. A quantitative method using HPLC-MS(2) has been developed for the determination of adonifoline, one of the retronecine-type hepatotoxic pyrrolizidine alkaloids in Senecio scandens Buch.-Ham. ex D. Don., a traditional Chinese herb. Using an orthogonal design test, a simple and rapid sample extraction method was developed. HPLC analysis was conducted using a C(18) column as stationary phase and a mixture of acetonitrile and aqueous formic acid as mobile phase. Good linearity for adonifoline was found in the concentration range 0.12-4.18 microg/mL, and the HPLC-MS/MS method was shown to be appropriate, in terms of sensitivity, precision and reproducibility. The quantities of adonifoline in extracts of 18 plant samples from different collection sources and from different parts (flowers, leaves, thick stems, slim stems and roots) of S. scandens were determined using the newly developed HPLC/MS(2) analysis.  相似文献

对8月和10月采集的葎草全草、嫩头、叶和茎中多糖含量进行测定,并对各部位提取纯化的多糖进行体外抗氧化作用研究。结果表明,8月份嫩头中多糖含量最高,达到(42.897±2.996)mg/g;浓度为5mg/mL的各部位葎草多糖溶液,对Fenton反应生成的羟自由基具有较强的清除能力,达85.15%~98.52%;对邻苯三酚自氧化法产生的超氧阴离子自由基也有较好的抑制能力,达57.15%~67.54%。葎草多糖对羟自由基和超氧阴离子自由基具有较高的清除能力。  相似文献

从攀援孔药花全草95%乙醇提取物中首次分离得到19个化合物,通过波谱数据或与已知物对照,它们分别鉴定为:(2S,3S,4R)-2-[(2R)-2-羟基-二十一酰胺基]-二十一烷-1,3,4-三醇(1)、(2S,3S,4R)-2-二十四酰胺基-十八烷-1,3,4-三醇(2)、胡萝卜甙(3)、β-谷甾醇(4)、(20S,22E,24R)-5α,8α-表二氧-麦角甾-6,22-二烯-3β-醇(5)、6β-羟基-豆甾-4-烯-3-酮(6)、十六烷酸-1-甘油酯(7)、桦木酸(8)、大黄素(9)、二十二烷酸-1-甘油酯(10)、对羟基苯甲醛(11)、十七烷酸-1-甘油酯(12)、金色酰胺醇乙酸酯(13)、十九烷酸-1-甘油酯(14)、棕榈酸(15)(、E)-p-香豆酸(16)、(22E,24S)-24-甲基-5α-胆甾-7,22-二烯-3β,5α,6β-三醇(17)、2-去氧-β-蜕皮激素(18)和auranamide(19)。  相似文献
