At the end of the spring 1987 growing season, the mycoparasite Sporidesmium sclerotivorum was applied at 0, 0.2, 2 or 20 kg ha‐1 to lettuce plants infected with Sclerotinia minor. Disease incidence was monitored in the same plots for five subsequent crops (three fall and two spring crops) without additional application of either pathogen or mycoparasite. Logistic growth curves were fitted to the data to describe disease progression over time for each inoculum level within each of the five crops. Within each crop, increasing the quantity of mycoparasite inoculum resulted in positive horizontal displacement of the curve with respect to time. As quantities of inoculum of S. sclerotivorum increased, inflection points of the disease progress curves increased at a decreasing rate. Thus, additional mycoparasite inoculum resulted in ever‐smaller increases in inflection point, and after a certain threshold level of mycoparasite inoculum (< 0.2 kg ha‐1), increases in inflection point did not result in meaningful increases in harvestable lettuce. Maximum rates of disease increase were not different among the treatments within each crop, but were different between crops. Maximum rates of disease increase averaged 3.4, 3.4, 2.1, 3.6 and 1.5% day‐1 for the fall 1987, spring 1988, fall 1988, spring 1989, and fall 1989, respectively. At all inoculum levels, the fall epidemics began later after planting than the spring epidemics.  相似文献

为了研究核盘菌的分类和生理分化问题,对供试的28个核盘菌株进行了可溶性蛋白质,芳香基酯酶和酸性磷酸酯酶同工酶电泳谱带以及紫外光吸收峰图形的试验研究,其结果,可分为五种类型。其中属于核盘菌属有四个种,即核盘菌Sclerotinia sclerotiorum(Lib.)de Bary(=Whetzelinia sclerotiorum(Lib.)Korf & Dumont),三叶草核盘菌S.trifoliorum Erikss.,细辛核盘菌(S.asari Wu et C.R.Wang和人参菌核病菌Sclerotinia sp。人参菌核病的另一分离菌和油菜菌核病的一个分离菌其谱带与灰葡萄孢(Botrytis cinerea)谱带相似,应归属于与葡萄孢属(Botrytis)相应的核盘菌。在核盘菌S.sclerotiorum种群中,南方菌系与北方菌系的可溶性蛋白质稍有差异,芳香基酯酶和酸性磷酸酯酶活性强度也稍有不同,可能存在生理分化现象。  相似文献

通过试验分析,结果得出,油菜菌核病菌生长温度范围在0~34℃之间,23℃以下菌丝生长速度与温度呈正相关,25℃以上则呈负相关,23~25℃最适宜,菌丝和菌核致死温度分别为45℃和75℃.该菌对酸碱度适应范围很广,PH1~14范围内只在PH_1条件下不能生长,其最适PH值为5~6。病菌利用铵态氮源能力最强,硝态氮源次之,不能利用亚硝态氮;对氨基酸利用能力以精氨酸最强,其次是酪氨酸、天门冬氨酸和谷氨酸,赖氨酸最差。植株内精氨酸和赖氨酸比值大的白菜型油菜“黄鳝籽”病情扩展最快。  相似文献

Growth of Coniothyrium minitans on potato dextrose broth was compared with that on an inexpensive molasses-yeast liquid medium at 18-22°C in static culture. Biomass and conidial production were, in general, similar, although the rate of biomass production was quicker and conidial production was slightly greater per unit volume of medium in the molasses-yeast medium. Air-dried biomass from molasses-yeast liquid culture containing mycelia, pycnidia and conidia of C. minitans was mixed (12%, w/w) with kaolin to give a kaolin-biomass dust. The ability of C. minitans to survive and subsequently infect and reduce the viability of sclerotia of Sclerotinia sclerotiorum from this kaolin-biomass dust was found to be little affected by storage for 48 weeks between 4 and 15°C but was decreased by higher storage temperatures. The kaolin-biomass dust preparation did not differ from a standard maizemeal-perlite inoculum of C. minitans in its ability to infect sclerotia of S. sclerotiorum or reduce their viability or carpogenic germination in glasshouse and field pot bioassays. Further, when either inoculum was applied once to glasshouse soil naturally infested with S. sclerotiorum prior to planting three successive crops of lettuce, the pattern of disease control, reduction of sclerotial numbers/ plot, infection of sclerotia, reduction of sclerotial viability and survival in soil were similar for both inocula. The potential for the commercial development of liquid-culture-produced inocula of C. minitans is discussed.  相似文献

The adventitious shoots in three populations of Cirsium arvense in sheep-grazed pastures were treated in October (spring) 1991 with a mycelium/wheat formulation of Sclerotinia sclerotiorum and the fates of mapped shoots were followed over the growing season. In untreated plots, deaths through natural causes were compensated for by births (emergence of new shoots above the soil) throughout the growing season, but, on plots treated with S. sclerotiorum, deaths from the induced disease exceeded births for 35 days following treatment, causing the shoot population to decline markedly. Disease-induced deaths occurred only among shoots present at the time of treatment; there was no evidence of transfer of the pathogen to shoots emerging after the treatment was applied. A life-table analysis showed that only 8% of the adventitious shoots emerging during the growing season survived to seeding on treated plots, compared with 28% on the untreated plots; most mortalities occurred in shoots at the vegetative stage of development. The dry mass of propagative roots in autumn was reduced to 35% of that on the untreated plots by the pathogen and the density of shoots emerging the following spring was reduced to a similar extent. The results of this study indicate that S. sclerotiorum has potential as a mycoherbicide for C. arvense in sheep-grazed pasture in New Zealand.  相似文献

Abstract Sunflower plants were inoculated with a virulent isolate of Sclerotinia sclerotiorum and with the same isolate nutritionally conditioned to produce small amounts of oxalic acid. The preconditioned isolate behaved as hypovirulent. Tomato plants were inoculated with four S. sclerotiorum isolates of increasing virulence. A close correlation among disease severity, accumulation of oxalic acid, decrease in pH and inhibition of polyphenoloxidase in both infected host tissues was demonstrated. Oxalic acid production as an important factor of virulence in S. sclerotiorum is emphasized and its effect on the phenolic metabolism of the host via inhibition of polyphenoloxidase is suggested.  相似文献

向日葵菌核病拮抗菌的筛选、鉴定及防效测定   总被引:1,自引:1,他引:0  
[背景] 由核盘菌(Sclerotinia sclerotiorum)引起的菌核病是影响向日葵产量的重要病害,近年来在我国内蒙古和甘肃等地频繁发生。[目的] 挖掘能够对向日葵菌核病进行生物防治的拮抗菌株和有效方法。[方法] 用4种不同的培养基通过稀释涂布法对向日葵健康植株的根际土壤菌群进行分离,利用平板对峙实验筛选出对核盘菌有抑制作用的菌株。选取拮抗作用较强的菌株进行向日葵离体叶片防效测定,采用形态学特征、生理生化特性结合16S rRNA基因序列分析进行菌种鉴定,并配制成不同单菌剂和复合菌剂进行盆栽实验,测定活体防效。[结果] 从土壤中共分离出142株菌,从中筛选到12株抑菌圈明显的拮抗菌株。其中拮抗菌Bacillus sp.NM63、JQ134、J7、J33和Streptomyces sp.Z9、ZX6抑菌圈直径大于25 mm,这6株菌在向日葵离体叶片防效测定实验中效果显著。菌株NM63、JQ134、J7、Z9、J33和ZX6单菌剂盆栽实验的防治效果依次为79.06%、74.10%、70.72%、67.83%、65.11%和57.11%。菌株配比为Z9:NM63:JQ134:J7=1:1:1:1的复合发酵菌剂Ⅰ对向日葵盆栽的活体防效为81.43%,菌株配比为Z9:NM63:JQ134:J7=1:2:2:1的复合发酵菌剂Ⅱ对向日葵盆栽的活体防效为85.88%。[结论] 筛选鉴定出多株对核盘菌具有较强抑制作用的拮抗菌株,复合菌剂Ⅱ对向日葵菌核病的防治具有显著效果。  相似文献

菌核是核盘菌Sclerotinia spp.在土壤中的主要存活形式和菌核病的主要初侵染源,在土壤中可存活8年以上,其数量和存活状况直接影响着菌核病的发生和危害程度。本研究以雪腐核盘菌Sclerotinia nivalis菌株SS-TB为材料,分析了菌核萌发的影响因素、致死温度以及土壤温度对菌核存活的影响。结果表明,未成熟菌核较成熟菌核更容易萌发;菌核萌发的最佳温度为20-25℃、pH为3.0-4.0、土壤含水量为20%-45%。菌核长时间浸泡水中对其存活不利,浸泡30d以后,存活率开始急剧下降,至47d时存活率为0。雪腐核盘菌菌核具有较强的耐高温特性,随着温度和处理时间的增加,菌核萌发率呈下降趋势。菌核在水浴中85℃ 5min、80℃ 10min、75℃ 10min、70℃ 30min、65℃ 120min、60℃ 180min时全部丧失活力。在土壤温度30℃和35℃处理5周、40℃和45℃处理4周时菌核全部失去活力。该研究结果为通过水旱轮作和土壤高温处理来防治西洋参菌核病提供了理论基础。  相似文献

Sunflower cultivation is affected seriously by Sclerotinia sclerotiorum (Lib.) de Bary in Iran, particularly north-western areas. Because of economic and environmental harms by chemical control, it is necessary to develop cultivars with adequate genetic resistance for reduction of yield losses. The purpose of this study was to find an effective method of inoculation with S. sclerotiorum under field evaluations. Three stem-inoculation techniques including: 1 – mycelium plug, 2 – oxalic acid solution (OAS) and 3 – infested wheat seeds with Sclerotinia mycelium were employed under field conditions. Four genotypes including Ghalami (local variety in market), Confeta, Allstar and Master were used in this study. The lesion length, lesion width and lesion as up and down leading on the stem from inoculation site were measured after 3, 7, 10 and 14 days of inoculation. The analysis of variance showed significant difference between all employed techniques and incubation days after inoculation. Mycelial plug (MP) inoculation technique produced significantly more developed lesions on the treated stems. In spite of this effect, Master variety demonstrated reasonable resistance reaction against the disease. The progress of disease in wounded treatments was also faster than the non-wounded ones. And, the shortest time to obtain significant differences between varieties was 10?days after inoculation. By comparison of results of lesion length at flowering and seed-filling stages, the more obvious effectiveness of the disease was observed at the second stage. Finally, there were negative correlations between mean temperature and mean lesion length in all three inoculation methods.  相似文献
