Summary Susceptibility toPhomopsis stalk disease ofPlantago lanceolata genotypes, sampled in three different populations with a variable degree of infection by the fungusPhomopsis subordinaria, was determined under greenhouse conditions. Susceptibility of the host varied within, but not among populations. No relationship between the intensity of the disease in the field and the mean susceptibility of the host genotypes sampled at those locations could be established. Host susceptibility appeared to be composed of the host genotypes sampled at those locations could be established. Host susceptibility appeared to be composed of different (uncorrelated) plant characteristics. Determining whether host genotypes are highly or slightly susceptible can only be achieved by field trials, where the plants are exposed to the whole set of disease inducing factors. The relevance of host susceptibility to the intensity of disease in the field is discussed in relation to the variation in pathogenicity of the fungus and the variation in environmental factors prevailing inP. lanceolata populations underP. subordinaria pathogen pressure. 本病系我国杨树新发生的一种病害。病原菌的分生孢子有2种形状,无色透明、单胞,呈纺缍形孢子大小为1.64—2.74×9.59—13.7μm。线形孢子顶端弯钩状大小为1.37—1.78×15—27.4μm。病原菌适宜生长的培养基为PDA、麦芽粉、豆饼粉培养基;温度为25—32℃和pH6—10。本病于4月下旬发病,5月中下旬分生孢子成熟,6月中下旬为分生孢子飞散高峰期。经室内采用多菌灵、甲基托布津、福美砷的200倍液浸泡插穗30、60、90分钟处理均有药害,采用100—200倍液浸泡10分钟无药害,对病害有防效。造林地幼树可采用上述药剂的100倍液进行刮破病斑涂抹,防效可达87.5%。  相似文献

A survey of die-back disease of neem was done in different agro climatic regions of Tamil Nadu, India using Global Positioning System (GARMIN 12). Twigs of Azadirachta indica (Neem) infected with die-back were collected from different regions of Tamil Nadu, India and they were further analyzed to determine the pathogen. Phomopsis azadirachtae the causal organism was isolated on malt extract agar from die-back infected neem twigs. They were identified by conventional and molecular methods. Phomopsis genus specific primers (5.8S r-DNA) were then used for the confirmation of P. azadirachtae – the causative agent of die-back of neem by Polymerase chain reaction (PCR). Studies revealed the amplification of expected 141bp DNA in P. azadirachtae isolated from the diseased trees of different regions of Tamil Nadu confirming the causal organism of die-back of neem. Studies revealed a very high incidence of die-back in most of the places of Tamil Nadu. Hand held GPS was used in the study which would help in continuous monitoring of the diseased trees.  相似文献

The growth of Brevibacterium flavum No. 2247 was inhibited over 90% at a concentration above 1 mg/ml of α-amino-β-hydroxyvaleric acid, a threonine analogue, and the inhibition was reversed by the addition of l-threonine, and to lesser extent by l-leucine, l-isoleucine, l-valine and l-homoserine. l-Methionine stimulated the inhibition. Several mutants resistant to the analogue produced l-threonine in the growing cultures. The percentage of l-threonine producer in the resistant mutants depended on the concentration of the analogue, to which they were resistant. The best producer, strain B-183, was isolated from resistant strains selected on a medium containing 5 mg/ml of the analogue. Mutants resistant to 8 mg/ml of the analogue was derived from strain B-183 by the treatment with mutagen, N-methyl-N’-nitro-N-nitrosoguanidine. Among the mutants obtained, strain BB-82 produced 13.5 g/liter of l-threonine, 30% more than did the parental strain. Among the resistant mutants obtained from Corynebacterium acetoacidophilum No. 410, strain C-553 produced 6.1 g/liter of l-threonine. Several amino acids other than l-threonine were also accumulated, and these accumulations of amino acids were discussed from the view of regulation mechanism of l-threonine biosynthesis.  相似文献

Diaporthe helianthi is the causal agent of sunflower stem canker, a serious pathogen of sunflower in Europe but recorded sporadically in Italy. The genetic diversity of D. helianthi isolates from different geographic origins (Argentina, France, Italy, Yugoslavia, Romania) was investigated using IGS sequences. A 400 bp fragment of the portion of the IGS region flanking the 5' end of the 18S gene was amplified from each isolate. The aligned nucleotide sequences showed intraspecific sequence homology from 99-100% among French/Yugoslavian isolates to 95-100% among Italian isolates. French/Yugoslavian isolates shared 90-92% sequence homology with Italian isolates. The phylogenetic tree obtained from the aligned data revealed three separate groups. Group 1 included all isolates from France and former Yugoslavia and one isolate from Argentina; Group 2 included all Italian isolates and one isolate from Argentina. The most distantly related isolate was that from Romania (Group 3). The average genetic distances among isolates within Group 1 and within Group 2 were 0.22 and 3.29 respectively. The analysis showed that all isolates originating from countries where severe outbreaks of the disease are reported annually (France and former Yugoslavia) form a well defined taxon characterized by relatively low variability. This group is distinct from the group formed by isolates originating from Italy, whose variability is relatively much higher. Results obtained revealed a marked differentiation among pathogen isolates, and members of Group 1 seem not yet to have spread into Italian sunflower-growing areas.  相似文献

Abstract: Calystegines are alkaloidal glycosidase inhibitors. They accumulate predominantly in young and meristemic parts of Calystegia sepium (Convolvulaceae). C. sepium, bindweed, infests meadows and cereal fields and is difficult to control chemically. Fungal pathogens against C. sepium are established as mycoherbicides. Stagonospora convolvuli LA39 attacks C. sepium and does not affect crop plants, but young plants of C. sepium are less susceptible to the fungus. The interaction of Stagonospora convolvuli with calystegines was investigated. Further, endophytic fungi of several classes were isolated from wild-grown Calystegia sepium leaves, and selected strains were tested for interaction with calystegines. Fungal growth on agar containing calystegines was not affected considerably. Plants in climate chambers were infected with an endophyte, Phomopsis, and with the fungal pathogen, Stagonospora convolvuli. Calystegine levels were measured in infected and non-infected plant tissues. Accumulation depended on developmental stage of the plant tissue and was not influenced by infection. Acid invertase was measured from fungal mycelia and from infected and non-infected plant tissues. Fungal acid invertase activity was not inhibited by 10 mM calystegine B2, while invertase from C. sepium leaves was inhibited. It is concluded that calystegines do not inhibit fungal development and sucrose consumption under the conditions of the present investigation, but may act by redirection of plant carbohydrate metabolism.  相似文献

人工培养条件下,用光学显微镜和透射电子显微镜对球壳孢目(Sphaerosidales)的Ascochyta,Phyllosticta,Phomopsis,Septoria四属进行个体发育研究,明确了其产孢方式:Ascochyta和Phyllosticta为内壁芽生瓶梗式;Phomopsis为全壁芽生瓶梗式;Septoria为全壁芽生合轴式,这一结果为属级分类提供了可靠的依据。产孢方式是属内共同具有的稳定特征,分生孢子的形成方式类型、产孢细胞及分生孢子的形态是划分属的首要标准。以寄主植物属为基础确定的同属大多数种在个体发育上无明显差别,分生孢子的形态及大小是分种的一个重要依据。  相似文献

为了探究桐花树内生真菌在抑菌方面的价值,该文以内生真菌发酵产物的抑菌作用为评价指标筛选活性菌株,采用生物活性跟踪方法结合多种色谱技术分离活性菌株的化学成分,通过波谱与文献数据比对鉴定单体化合物结构,并利用微孔板法测定单体化合物的抑菌活性。结果表明:(1)从桐花树分离得到的16株内生真菌分属2纲7目10科10属,镰刀菌属(Fusarium)为优势菌属。内生真菌GXIMD02029和GXIMD02039的发酵产物对枯草芽孢杆菌、表皮葡萄球菌、耐甲氧西林金黄色葡萄球菌、藤黄微球菌、粘性放线菌和金黄色葡萄球菌有不同程度的抑制作用,GXIMD02038发酵产物对耐甲氧西林金黄色葡萄球菌、藤黄微球菌和金黄色葡萄球菌有抑制作用。(2)7个化合物从内生真菌Phomopsis sp. GXIMD02029中被分离并鉴定为(15R)-acetoxydothiorelone A(1)、cytosporone B(2)、pestalotiopsone H(3)、pestalotiopsone B(4)、4-Hydroxybenzaldehyde(5)、p-Hydroxybenzoic acid(6)、N-(2-phenylethyl)acetamide(7)。(3)化合物1和2有不同程度的抑菌作用,化合物1对枯草芽孢杆菌、表皮葡萄球菌、耐甲氧西林金黄色葡萄球菌的MIC值为16.25 SymbolmA@ g·mL-1,对藤黄微球菌和粘性放线菌的MIC值为7.812 5 SymbolmA@ g·mL-1,对金黄色葡萄球菌的MIC值为31.25 SymbolmA@ g·mL-1。化合物2对藤黄微球菌的MIC值为62.5 SymbolmA@ g·mL-1,对枯草芽孢杆菌、表皮葡萄球菌、耐甲氧西林金黄色葡萄球菌、粘性放线菌的MIC值为125 SymbolmA@ g·mL-1,对金黄色葡萄球菌的MIC值为250 SymbolmA@ g·mL-1。该文筛选了3株活性菌株,首次报道化合物1具有抗菌活性,为桐花树内生真菌在抗菌价值方面提供了依据。  相似文献

Microsphaeropsis amaranthi and Phomopsis amaranthicola are potential biological control agents for several Amaranthus species. In an effort to understand the initial infection processes with these pathogens, a study was conducted of the conidial germination and germ tube length (μm) on the weed leaf surfaces at 21 °C and 28 °C. Weeds included Amaranthus rudis, A. palmeri, A. powellii, A. retroflexus, A. spinosus, A. hybridus, and A. albus. For P. amaranthicola, conidial germination and germ tube length varied among the seven weed species at both temperatures, while for M. amaranthi the differences in germ tube lengths were significant among weed species only at 21 °C. While the conidia of M. amaranthi and P. amaranthicola germinated on the leaf surfaces of all seven weed species, temperature appeared to impact the number and length of germ tubes on the leaf surfaces. The percentage of germinated conidia and the length of germ tubes at both temperatures were often greater for M. amaranthi than for P. amaranthicola. In order for the fungal pathogen to successfully infect and kill a weedy host, conidia must germinate and form a germ tube, two processes that vary with host species and temperature for M. amaranthi and P. amaranthicola. The extent to which successive infection processes, e.g., penetration, invasion and colonization, contribute to host specificity warrants study.  相似文献
