Anomalous sedimentation behavior has been observed for high molecular weight duplex DNA's in sucrose gradients. The sedimentation rate of DNA's having molecular weights of 108 or higher is influenced by high centrifugal fields. The change in the sucrose sedimentation coefficient due to this effect, SRPMsuc-S0suc, is equal to 1 × 10?48M3.65( The anomalous behavior is not influenced by DNA concentration at sufficiently low concentrations. Because of the smallness of the coefficient this effect has not been previously detected for DNA's the size of T2 or smaller at rotor speeds below 40000 RPM. For example, the relative sedimentation coefficient of T2 DNA at 65 000 RPM is only 9% less than at 10000 RPM. However, the sedimentation profile of heterogeneous high molecular weight [(100 – 350) × 106] E. coli DNA is severely altered even at moderate rotor speeds (37000 RPM). Therefore, it seems advisable to use low rotor speeds when sedimenting high molecular weight DNA's.  相似文献

Pregnenolone (3β-hydroxy-5-pregnen-20-one) and DHA (3β-hydroxy-5-androsten-17-one), substrates for 3β-hy-droxysteroid dehydrogenase (3β-HSD), with KM values of 15–40 nM, were ineffective inhibitors of 5-ene-3-ketosteroid isomerase (isomerase), with KI values >40 μM in each case. Progesterone and androstenedione (4-androstene-3, 17-dione), 3β-HSD inhibitors with KI values of 5.0 μM and 0.8 μM respectively, were also relatively ineffective inhibitors of isomerase, with KI values of 30 μM and 16.5 μM respectively. Exposure of microsomes to hydrogen peroxide, which significantly increases the KM for pregnenolone as a 3β-HSD substrate, had no effect on the KM for the isomerase substrate 5-pregnene-3, 20-dione.It is concluded that the data do not support the common site concept with regard to the conversion of pregnenolone to progesterone by human placental microsomes.  相似文献

Summary Variation in concentrations of N, K, Mg, and Ca in vegetative-buds, leaves, twigs, and male catkins of trembling aspen (Populus tremuloides Michx.), determined throughout a growing season in central Minnesota, reflected the different physiological roles of the nutrients. Freeze-fracture electron microscopy was used to study water content related freezing resistance in Grand Rapids lettuce seeds. Consistent and recognizable conformational changes occurred in lipid-water phases of lettuce seeds at different moisture contents. In air-dry lettuce seed cotyledons, the lipids lying in spherical lipid bodies near the cell wall appeared amorphous, while the structure was crystalline above 20% water content. The lipid bodies interassociated into membrane bilayers in seeds containing 20 to 25% water. Such lyotropic phase transitions in membrane lipids during lettuce seed hydration are believed to contribute to the biphasic freezing behavior observed in lettuce seeds at different moisture contents and to provide a natural freezing tolerance mechanism for highly desiccated plant tissues such as seeds.  相似文献

Regeneration of many late-successional tree species depends on specialized safe sites. The primary objective was to investigate the roles of seedbed and moisture retention as dimensions of safe sites for the early regeneration of drought-sensitive northern white cedar (Thuja occidentalis). We hypothesized that rates of germination, survival, and growth of T. occidentalis are unlikely to differ among seedbed types under conditions of abundant water, but that differences are likely to emerge as water becomes more limited. In a 67-d greenhouse experiment, cedar seeds were sown on logs, leaf litter, and soil of cedar and paper birch (Betula papyrifera) canopy origin. Seedbeds were subjected to three water treatments. Among the water treatments, highest germination rates occurred within the high water treatment, although germination on cedar litter was comparable to that of the low water treatment. Higher germination and survival rates occurred on decayed logs than other natural seedbeds for medium (P = 0.001) and low (P < 0.0001) water treatments. Germination on birch logs occurred at higher rates than on cedar logs within the low water treatment (P = 0.04). Seedling growth for the medium water treatment was lower on leaf litter than any other type of seedbed (P < 0.01). Results generally demonstrated that the interplay between seedbed and moisture retention is a component of safe sites for T. occidentalis regeneration.  相似文献

ABSTRACT In apparent response to recent periods of global warming, some migratory birds now arrive earlier at stopover sites and breeding grounds. However, the effects of this warming on arrival times vary among locations and species. Migration timing is generally correlated with temperature, with earlier arrival during warm years than during cold years, so local variation in climate change might produce different effects on migration phenology in different geographic regions. We examined trends in first spring arrival dates (FADs) for 44 species of common migrant birds in South Dakota (1971–2006) and Minnesota (1964–2005) using observations compiled by South Dakota and Minnesota Ornithologists’ Unions. We found significant trends in FAD over time for 20 species (18 arriving earlier and two later) in South Dakota and 16 species (all earlier) in Minnesota. Of these species, 10 showed similar significant trends for both states. All 10 of these species exhibited significantly earlier arrival, and all were early spring migrants, with median FADs before 10 April in both states. Eighteen of the 44 species showed significant negative correlations of FADs with either winter (December–February) or spring (arrival month plus previous month) temperatures in one or both states. Interestingly, spring temperatures in both South Dakota and Minnesota did not warm significantly from 1971–2006, but winter temperatures in both states warmed significantly over the same time period. This suggests that the warmer winters disproportionately affected early spring migrants, especially those associated with aquatic habitats (seven of the 10 species showing significantly earlier spring arrival in both states). The stronger response to climate change by early spring migrants in our study is consistent with the results of several other studies, and suggests that migrants, especially early migrants, are capable of responding to local temperature conditions experienced on wintering grounds or along the migration route.  相似文献

ABSTRACT North temperate species on the southern edge of their distribution are especially at risk to climate-induced changes. One such species is the moose (Alces alces), whose continental United States distribution is restricted to northern states or northern portions of the Rocky Mountain cordillera. We used a series of matrix models to evaluate the demographic implications of estimated survival and reproduction schedules for a moose population in northeastern Minnesota, USA, between 2002 and 2008. We used data from a telemetry study to calculate adult survival rates and estimated calf survival and fertility of adult females by using results of helicopter surveys. Estimated age- and year-specific survival rates showed a sinusoidal temporal pattern during our study and were lower for younger and old-aged animals. Estimates of annual adult survival (when assumed to be constant for ages >1.7 yr old) ranged from 0.74 to 0.85. Annual calf survival averaged 0.40, and the annual ratio of calves born to radiocollared females averaged 0.78. Point estimates for the finite rate of increase (λ) from yearly matrices ranged from 0.67 to 0.98 during our 6-year study, indicative of a long-term declining population. Assuming each matrix to be equally likely to occur in the future, we estimated a long-term stochastic growth rate of 0.85. Even if heat stress is not responsible for current levels of survival, continuation of this growth rate will ultimately result in a northward shift of the southern edge of moose distribution. Population growth rate, and its uncertainty, was most sensitive to changes in estimated adult survival rates. The relative importance of adult survival to population viability has important implications for harvest of large herbivores and the collection of information on wildlife fertility.  相似文献

Abstract: There is interest in expanding eastern wild turkey (Meleagris gallopavo silvestris) populations north of their current range. We hypothesized that winter survival and food availability are primary determinants in setting the northern extent of wild turkey distribution. To test our hypothesis, we translocated wild turkey females north of their present range into central Minnesota, USA, and compared survival in areas with supplemental food in the form of corn food plots versus areas with no supplemental food. During 2 winters with below-average snow, winter survival was higher for females with supplemental food. In one winter with above-average snow depths, survival was extremely low even with supplemental food. Supplemental food could augment survival during mild winters if wildlife managers arrange with farmers to, annually, retain standing corn near roosting habitat, but food plots may only partially offset effects of deep snow. Managers should critically evaluate northern habitats, long-term costs of sustained feeding, and potential outcomes of concentrating animals and introducing wild animals into new ecosystems. Winter survival may delimit the northern range of wild turkeys, though annual survival rates may also be important and need further research.  相似文献
