Summary A direct comparison of microspore culture and anther culture was made in Brassica napus using F1 crosses of Regent (canola) by Golden (rapeseed), and their reciprocals, as well as a hybrid between Reston and a highly embryogenic, canola-quality breeding line (G231) as donor plants. The study confirmed that microspore culture can be ten times more efficient than anther culture for embryo production. Embryo yields from cultures initiated from the Reston x G231 were four-fold greater than those initiated from the Regent x Golden crosses, and significant differences were also detected among cultures initiated from the different Regent x Golden crosses. These results illustrate the influence that donor plant genotype has on embryo production. However, superior embryogenic potential among donor material was not always coincident with superior plant production. The average haploid-todiploid ratio in microspore-derived regenerates was 21 for the population obtained from the Regent x Golden crosses but 11 for the Reston x G231 cross. For both types of material, the frequency of diploids increased upon repeated cycles of explanting. A field study showed that there were no differences between the populations of anther-derived and microspore-derived spontaneous diploid and doubled haploid lines, with respect to the days required for them to flower or to mature. The information is valuable for canola breeding programs considering the use of haploidy.  相似文献

High percentages of micro-calli and micro-derived embryos were produced from isolated asparagus microspores at late uninucleate stage on MS liquid medium supplemented with 1.0 mg l–1 2,4-D and 0.5 mg l–1 BA. Two types of calli, namely compact callus (CC) and loose callus (LC), were found. Plantlets were regenerated via organogenesis, when these calli were transferred onto MS solid medium supplemented with 1.0 mg l–1 BA and 0.2 mg l–1 IBA 6 weeks. Embryos were produced from liquid cultured microspores, or from solid cultured micro-calli. The frequencies of haploid plant production from organogenesis and embryogenesis were compared. Effects of plant growth regulators on callus production, plantlet regeneration, and haploid plant production were tested. 采用不完全双列杂交设计研究了 6个母本3个父本甜玉米自交系籽实含糖量的配合力效应。结果表明,父母本一般配合力和特殊配合力效应均方都显著。根据各亲本的表现把亲本5033归为一般配合力高而特殊配合力方差大的最理想类型;亲本5012和5011属一般配合力高而特殊配合力方差小的类型;亲本5018、5034、5028和5024为一般配合力低,但特殊配合力方差高的类型; 亲本5023和5009属一般配合力效应和特殊配合力方差都低的类型,最缺少实用价值。  相似文献

研究了大麦(Hordeum vulgare L.)花药-花粉培养中不同预处理对花药内源激素(ABA,IAA,IPA)含量和过氧化物酶活性的影响。结果表明:1. 经甘露醇预处理不同天数,同一种基因型的3种内源激素的变化和不同基因型的同一种内源激素的变化规律十分相似,均是在预处理初期含量急剧增加,最高值在0.5或1 d 处。以后开始逐渐下降;最后,保持在一定的水平上。2. 在甘露醇预处理过程中,两种基因型花药过氧化物酶活性的变化规律也十分相似。在预处理前期(从开始到第3天)活性呈直线上升,第3天达到最大值。从第3天到第5天活性减弱。最后,活性又开始增强。3. 在低温预处理过程的初期(2~5 d) 两种基因型花药过氧化物酶活性都出现第一个小峰。在第14天(“Harrington”)或第21天(“Igri”)又出现第二个峰值,但后者较高。在预处理的后期(从第28天到第35天)两种基因型花药过氧化物酶的活性又呈现上升的趋势。同甘露醇预处理后期变化一致  相似文献

含单核中后期至后期花粉的蚕豆离体花药,经过2mM丁酸钠24小时预处理后,再漂浮培养于pH5.8或7.0的液体培养基。以不经预处理的作为对照。结果如下: 1.培养后9天内,丁酸钠预处理和培养基pH值并不明显影响花粉退化百分率。2.培养初期,pH7.0显著促进小孢子不等分裂。3.丁酸钠预处理抑制培养初期的小孢子有丝分裂,而后又显著增加小孢子均等分裂百分率。4.丁酸钠预处理导致小孢子有丝分裂类型的趋向改变。本文还对丁酸钠导致有丝分裂类型趋向改变的可能原因,进行了讨论。  相似文献

Summary A simple procedure is described for the in vitro production of tobacco (Nicotiana tabacum L.) pollen from microspores isolated just before entering mitosis. During a 3-day culture period in a liquid medium containing pyrimidine nucleosides these microspores develop into young pollen grains to the stage of starch deposition. Pollen maturation and transition to dormancy is achieved during a further 2- to 3-day culture period in the same medium stepwise supplemented by a concentrated solution of sucrose and l-proline. Upon transfer of the pollen to a simple germination medium containing sucrose and boric acid, up to 40% of the grains were observed to produce relatively long tubes. The in vitro-matured pollen grains can be stored at-20° C either suspended in 1.17 M sucrose and 100 mM l-proline or separated from the medium on filter paper discs. The stored pollen germinated both in vitro and on the stigma, the pollen tubes grew through the style into the ovary and pollination produced up to 300 viable seeds per pod. The procedure is of interest for pollen developmental studies and various fields of pollen manipulation, such as in vitro pollen selection.  相似文献

对水稻(OryzasativaL.)小孢子发育过程中微管变化的研究表明,微管在小孢子不同发育阶段呈现多样性。在花粉母细胞内,微管形成许多粗束和分支,围绕着核分布形成一个网络。花粉母细胞经第一次减数分裂形成二分体。在每一个二分体细胞内,有许多微管束,从核周辐射至细胞质各部位;在细胞质存在一个疏松的微管束网络。二分体经第二次减数分裂形成四分体,在每一个四分体细胞内,微管束呈辐射状,从核膜辐射入细胞质内。四分体形成后不久,四分体的四个细胞便分开,每一个细胞变成一个独立的小孢子。在早期的小孢子细胞内,微管束呈疏松网状分布。其中有些微管束朝向胞质一个小突起聚集。当小孢子进入中期发育阶段,在胞质的小突起部位微管束密度增大。小突起最终形成为萌发孔。当小孢子发育至成熟期,细胞内的微管束变得纤细,而网络则变得紧密。之后的发育阶段(即花粉发育不同阶段)因荧光标记难以进入细胞,无法获得清晰的图像。  相似文献

A procedure for direct gene transfer into isolated microspores of rapeseed (Brassica napus L.) and the production of fertile transgenic plants is presented. By modifying the microspore culture method and adopting the firefly luciferase (Luc) gene as a non-destructive marker, we could obtain stably transformed androgenetic embryos from bombarded microspores. Luc-positive embryos were easily isolated from the large non-transformed population using a high-sensitivity bioluminescent image analyzer. PCR and Southern blot analyses confirmed that the introduced transgene was integrated stably into the genome of the selected embryos. Diploidized plants obtained from the haploid embryos were self-pollinated, and all of the offspring tested were Luc-positive, indicating rapid fixation of the transgene which is characteristic of doubled haploids. Biogenesis and function of the lipidic structures of pollen grains   总被引:31,自引:0,他引:31  
 Pollen grains contain several lipidic structures, which play a key role in their development as male gametophytes. The elaborate extracellular pollen wall, the exine, is largely formed from acyl lipid and phenylpropanoid precursors, which together form the exceptionally stable biopolymer sporopollenin. An additional extracellular lipidic matrix, the pollen coat, which is particularly prominent in entomophilous plants, covers the interstices of the exine and has many important functions in pollen dispersal and pollen-stigma recognition. The sporopollenin and pollen coat precursors are both synthesised in the tapetum under the control of the sporophytic genome, but at different stages of development. Pollen grains also contain two major intracellular lipidic structures, namely storage oil bodies and an extensive membrane network. These intracellular lipids are synthesised in the vegetative cell of the pollen grain under the control of the gametophytic genome. Over the past few years there has been significant progress in elucidating the composition, biogenesis and function of these important pollen structures. The purpose of this review is to describe these recent advances within the historical context of research into pollen development. 应用甘蓝型油菜DH系保604为材料研究小孢子胚发生过程,结果表明,在小孢子离体培养1~5d内,随培养天数增加,小孢子的存活率迅速下降,部分小孢子培养后出现细胞膨大和分裂,并沿2-细胞。“f”形3细胞,多细胞原体,胚柄球形胚,心形胚最终发育成鱼雷形胚,一般在心形胚阶段,胚柄脱离胚主体部分游离到培养基中,大多数膨大的细胞不能分裂或分裂后停止发育或发育异常。  相似文献
