Toxic and nontoxic peptides were isolated from the cyanobacterium Microcystis aeruginosa PCC 7806 by a procedure including extraction of cells with water-saturated 1-butanol, chromatography of the extract on silica gel plates and high performance liquid chromatography (HPLC) on Partisil-5. The toxin was shown to be only a minor constituent, being negatively charged and thus separable by electrophoresis, within the HPLC-purified fraction. It contained erythro-β-methyl-D-Asp, D-Glu, D-Ala, L-Leu, and L-Arg known to be part of the Microcystis peptide-toxin with Mr 994. The major part of the HPLC-purified fraction was assigned, however, to a nontoxic peptide with a Mr of 956. Partial hydrolysis studies of the nontoxic peptide(s) revealed amino acid sequences composed of D-Glu, N-methyl-Phe, and 3,4-dehydro-Pro, aside from the common L-amino acids. Cyclic linkage in the nontoxic peptide(s) appears likely.  相似文献

Cultures of the cyanobacterium Microcystis firma show rhythmic uptake and release of ammonia under conditions of carbon limitation. The massive removal of ammonia from the medium during the first light phase has little impact on the intracellular pH: a pH shift of less than 0.2 U towards the alkaline can be measured by in vivo 31P NMR. Furthermore, the energy status of the cells remains regulated. In vivo 15N NMR of M. firma, cultivated either with labelled nitrate or ammonia as the sole nitrogen source, reveals only gradual differences in the pool of free amino acids. Additionally both cultivation types show -aminobutyric acid, acid amides and yet unassigned secondary metabolites as nitrogen storing compounds. Investigating the incorporation of nitrogen under carbon limitation, however, only the amide nitrogen of glutamine is found permanently labelled in situ. While transamination reactions are blocked, nitrate reduction to ammonia can still proceed. Insertion of foreign DNA into Ti plasmid-derived vectors in Agrobacterium tumefaciens is currently the most frequently used strategy for generating transgenic plants in a wide variety of species. Limitations of the host range of Agrobacterium restrict its usefulness in many cases, particularly when dealing with monocotyledonous plants. The objective of this presentation is to briefly discuss the efficiency of the transformation process utilized by Agrobacterium tumefaciens , potential barriers to efficient transformation by Agrobacterium that result in limitation of its useful host range, and how an understanding of the successful Agrobacterium /plant cell interaction might lead to advances in a variety of DNA delivery methodologies.  相似文献

Abstract Microcystis aeruginosa (Synechocystis ) is a unicellular cyanobacterium that performs oxygenic photosynthesis. We found two novel sets of repetitive sequences, A (REP-A) and B (REP-B), on the M. aeruginosa K-81 genomic DNA, which consisted of distinct motifs of tandem repeated sequences located in the up- and downstream regions of the orf1 structural gene, respectively. Genomic Southern hybridization revealed multicopies of REP-A and -B on the genome. Furthermore, genomic Southern blots of cyanobacteria species with the REP-A and -B probes revealed that different hybridization signals appeared on the genomic DNAs of all 12 Microcystis strains, but no signal appeared on those of Synechocystis sp. PCC 6803, Synechococcus sp. PCC 7942, and Anabaena sp. PCC 7120.  相似文献

Over a period of several days, rhythmic changes in extracellular NH+4 concentration take place in cultures of the cyanobacterium Microcystis firma (Bré et Lenorm.) Schmidle, strain Gromov/St. Petersb. 398, under conditions of restricted CO2 supply and light/dark alternation. The changes are enhanced by nitrate supply. Among the various processes generating intracellular NH+4 (NH44 uptake, NO3 reduction, protein and amino acid degradation, photorespiration), NO3 reduction appears as the one most important. This can be concluded from experiments with and without nitrate and/or ammonium in the medium. In the presence of saturating CO2, continuous light, or continuous darkness, rhythmic NH+44 oscillations are not induced. Studies of the incorporation of NH+4 nitrogen by in vivo 15N-NMR show that if CO2 is supplied, 15N is accumulated in several components with the following time course: in the first hour in Gln (δ), in the second hour in the α-amino groups of most nonbranched amino acids, in the third hour in γ-aminobutyric acid (GABA), Orn (δ) and Lys (ε), and in the sixth hour in Ala. Carbon limitation, however, results in accumulation of label in the amide nitrogen of glutamine only.  相似文献

The desmid Staurastrum luetkemuellerii Donat et Ruttner and the cyanobacterium Microcystis aeruginosa Kütz. were grown in mixed cultures with various phosphate (Pi) additions. One pulse of Pi each day (semi-continuous cultures) favored M. aeruginosa whereas S. luetkemuellerii was favored when the same quantity of Pi was supplied continuously (chemostats). Both species coexisted under P limitation provided that the nutrient was supplied in an appropriate mode. The ability of each species to compete for P depended on their Pi uptake characteristics and their capability to retain the accumulated Pi. High affinity in uptake at low Pi concentrations contributed considerably to the growth eficiency of S. luetkemuellerii under continuous supply of PiM. aeruginosa was, however, consistently superior to S. luetkemuellerii in accuniulatiug the newly added P, but had a high rate of Pi release. In both -types of cultures, a net high of P went from M. aeruginosa to S. luetkemuellerii. The kinetic characteristics of the two species were used to simulate the outcome of competition experiments. Simulations agreed with the experimental data f both uptake and Pi release were considered in the model. The zlariable P*(the concentration of Pi at which the net uptake is equal to μ·QP is a function of uptake and release of Pi but could not explain the chemostat results. S. luetkemuellerii was the winner in many experiments even if its P*was higher thou that of M. aeruginosa. Thus, in the present case Pc (the concentration at which the net uptake is zero) was a better predictor of the ability to compete for Pi under steady state as well as transient conditions in the Pi supply.  相似文献

Abstract A new sulfated, cyclic depsipeptide, called cyanopeptolin S, from Microcystis sp. was isolated from a water bloom in the Auensee/Leipzig (Germany). The depsipeptide had a relative molecular mass of 925 and contained l-arginine, l-threonine, l-isoleucine, N-methyl-l-phenylalanine, a l-glutamic acid-δ-aldehyde ring system and a sulfated d-configurated glyceric acid as a side chain. The structure was elucidated by means of two-dimensional 1H and 13C nuclear magnetic resonance spectroscopy, fast atom bombardment mass spectroscopy, Fourier transformed infrared spectroscopy and combined gas-liquid chromatography/mass spectrometry. Cyanopeptolin S inhibited trypsin with an IC50≤ 0.2 μg ml−1.  相似文献

The growth interactions amongst the blue-green algal species Anabaena oscillarioides, Microcystis aeruginosa and the green alga, Chlorella sp. were studied both in mixed cultures and in filter cultures separated by a membrane filter in the two arms of an interaction U-tube. The role of nutrients especially phosphate upon the interaction has also been studied. Anabaena and Microcystis both inhibited the growth of Chlorella while Microcystis also inhibited the growth of Anabaena. The inhibitory effect of Microcystis was found to be dependent on high concentrations of the initial algal inocula and independent of the initial concentration of nutrients such as inorganic phosphate, indicating that the nature of the inhibition is probably due to the production of inhibitory extracellular products by Microcystis. On the other hand, the inhibitory effect of Anabaena on Chlorella is the consequence of nutrient competition with Anabaena competing more effectively for the available phosphate.  相似文献

Light-energy absorption by Microcystis aeruginosa with and without gas vacuoles was observed, respectively by using an integrating sphere photometer. As far as the concentration of cell suspension of the order of 1067 cells/ml in this work was concerned, the performance of gas vacuoles to shield incident light was most unlikely. Referring to a correlation secured by the integrating sphere photometer between light absorption and cell concentration of the suspension, a turbidostat culture of the blue-green alga demonstrated that the growth efficiency, Y kJ defined as g cells harvested per kJ of light energy absorbed by the cells was nearly 0.004. In stratified lakes, dominance of the phytoplankton by cyanobacteria is largely the result of their buoyancy and depth regulation. Bloom-forming cyanobacteria regulate the gas vesicle and storage polymer contents of their cells in response to interactive environmental factors, especially light and nutrients. While research on the roles of nitrogen and phosphorus in cyanobacterial buoyancy regulation has reached a consensus, evaluations of the roles of carbon have remained open to dispute. We investigated the various effects of changes in carbon availability on cyanobacterial buoyancy with continuous cultures of Microcystis aeruginosa Kuetz. emend. Elenkin (1924), a notorious bloom-former. Although CO2 limitation of photosynthesis can promote buoyancy in the short term by preventing the collapse of turgor-sensitive gas vesicles and/or by limiting polysaccharide accumulation, we found that sustained carbon limitation restricts buoyancy regulation by limiting gas vesicle as well as polysaccharide synthesis. These results provide an explanation for the positive effects of bicarbonate enrichment on cyanobacterial nitrogen uptake and bloom formation in lake experiments and may help to explain the pattern of cyanobacterial dominance in phosphorus-enriched, low-carbon lakes.  相似文献
