Immunization of C57BL/6 mice with BCGcw stimulated a population of "suppressor cells" which had a decreased capacity to induce the graft-versus-host response. The graft-versus-host response was quantitated using the Simonsen splenomegaly assay. F1 mice (C57BL/6 X CBA) were inoculated intraperitoneally with 1 X 10(8) parental (C57BL/6) or (CBA) spleen cells. The F1 mice were sacrificed 13 days later and the resulting splenomegaly was 3-4 times the normal amount. F1 mice which were injected with parental BCGcw-primed C57BL/6 spleen cells had a 50% inhibition of splenomegaly, whereas BCGcw-primed CBA spleen cells (a strain which does not develop suppressor cells) did not show this inhibition. In vitro results also confirmed that only C57BL/6 mice and not CBA mice developed suppressor cells after BCGcw immunization. A second study showed that X-irradiated (1000 R) BCGcw-primed "suppressor cells" could inhibit splenomegaly caused by the inoculation of normal parental C57BL/6 cells into F1 mice. The mechanism by which BCGcw-primed "suppressor cells" caused this inhibition of splenomegaly was delineated and found to be dependent upon the secretion of prostaglandin (PGE-1). Indomethacin and aspirin, potent inhibitors of prostaglandin synthesis, blocked the activity of C57BL/6 BCGcw "suppressor cells" and splenomegaly resulted. Systemic administration of the prostaglandin (15S)-15-methyl PGE-1 reduced splenomegaly approximately 50% in F1 mice which were injected with C57BL/6 or CBA cells. These results indicated that immunization with BCGcw stimulated a population of "suppressor cells" which could cause a decrease in graft-versus-host response and that the secretion of prostaglandin was responsible for this inhibition.  相似文献

A study was initiated to determine the number, chromosomal location, and magnitude of effect of QTL (quantitative trait loci or locus depending on context) controlling protein and starch concentration in the maize (Zea mays L.) kernel. Restriction fragment length polymorphism (RFLP) analysis was performed on 100 F3 families derived from a cross of two strains, Illinois High Protein (IHP), X Illinois Low Protein (ILP), which had been divergently selected for protein concentration for 76 generations as part of the Illinois Long Term Selection Experiment. These families were analyzed for kernel protein and starch in replicated field trials during 1990 and 1991. A series of 90 genomic and cDNA clones distributed throughout the maize genome were chosen for their ability to detect RFLP between IHP and ILP. These clones were hybridized with DNA extracted from the 100 F3 families, revealing 100 polymorphic loci. Single factor analysis of variance revealed significant QTL associations of many loci with both protein and starch concentration (P < 0.05 level). Twenty-two loci distributed on 10 chromosome arms were significantly associated with protein concentration, 19 loci on 9 chromosome arms were significantly associated with starch concentration. Sixteen of these loci were significant for both protein and starch concentration. Clusters of 3 or more significant loci were detected on chromosome arms 3L, 5S, and 7L for protein concentration, suggesting the presence of QTL with large effects at these locations. A QTL with large additive effects on protein and starch concentration was detected on chromosome arm 3L. RFLP alleles at this QTL were found to be linked with RFLP alleles at the Shrunken-2 (Sh2) locus, a structural gene encoding the major subunit of the starch synthetic enzyme ADP-glucose pyrophosphorylase. A multiple linear regression model consisting of 6 significant RFLP loci on different chromosomes explained over 64 % of the total variation for kernel protein concentration. Similar results were detected for starch concentration. Thus, several chromosomal regions with large effects may be responsible for a significant portion of the changes in kernel protein and starch concentration in the Illinois Long Term Selection Experiment.  相似文献

Reaction rates were measured for the low pH-induced Cu ligand modification of azurins from Pseudomonas aeruginosa and Alcaligenes faecalis. Loss of the intense absorption band at 625 nm obeyed a rate law:
where Az is the concentration of azurin in its native oxidized form possessing the 625 nm band. For Pseudomonas aeruginosa at 25°C, n = 1 and k = 4.0 × 10-2 sec-1 M-1 in citrate buffer but 1.2 × 10-2 sec-1 M-1 in phosphate buffer. For Alcaligenes faecalis, n = 3 and k = 1.5 × 104 sec-1 M-3 in citrate and 2.6 × 103 sec-1 M-3 in phosphate. In equilibrium experiments on Alcaligenes faecalis azurin in citrate buffer, the pH-dependent change to the low pH form exhibited an apparent transition pK of 3.1. The form of the rate law implies a mechanistic scheme that contains fast equilibrium protonation steps prior to a rate limiting ligand rearrangement.  相似文献

A DNA fragment carrying the thrP region of Escherichia coli has been identified and characterized. Starting from a secondary site lysogen of bacteriophage λ, where the position of the prophage is either within thrP or between thrP and thrP structural genes (Gardner et al., 1974; Gardner &; Smith, 1975) it has been possible to isolate transducing phages which carry bacterial DNA from either side of the prophage. By Int-Xis mediated site-specific recombination we have generated recombinant transducing phages which carry an intact thrP region. A phage carrying a regulatory mutation, which maps in the thrP region, has also been constructed.Mapping with several restriction endonucleases has allowed us to construct a map of the cleavage sites of the thrP region. A 1700 base-pair HaeIII fragment carrying the secondary attachment site (attΔOΔ′) was isolated and its position was localized within a 180 base-pair TaqI-HhaI restriction fragment. The location of attΔOΔ′ in the HaeIII fragment suggests that the entire thrP region is carried by this fragment.  相似文献

We present a simple, efficient system for preparative polyacrylamide gel electrophoresis (PAGE) in sodium dodecyl sulfate. The instrument features a novel sample collection assembly which is fitted directly onto the bottom of the gel cylinder. Buffer pumped uniformly upward through a porous plastic disk from the lower tray sweeps the emerging sample into a collection system with high efficiency. Measures which prevent the destruction of tryptophan residues and the blockage of α-amino groups observed in electrophoresed polypeptides are described.  相似文献

An accurate understanding of harvest trends is required for effective wildlife management. Trapper harvest data represent valuable long-term data for evaluating patterns and trends for wildlife species at broad spatiotemporal scales. Inferring accurate trends from harvest data, however, first requires identifying and controlling for confounding factors that vary independent of abundance. We investigated trends in 43 years of trapper harvest data (1976–2018) from Illinois, USA, for red fox (Vulpes vulpes), gray fox (Urocyon cinereoargenteus), and coyote (Canis latrans) while controlling for factors that may affect trapper effort, including number of effective (i.e., successful) trappers, pelt price, gasoline price, winter unemployment, and winter weather conditions. Annual trapper harvest for red and gray foxes declined and was affected by gasoline price and winter unemployment, whereas annual trapper harvest for coyotes increased and was not strongly affected by other covariates. After adjusting for pelt price, harvest of red foxes was relatively stable, but harvest of gray foxes declined and harvest of coyotes increased. Effects of covariates on harvest per successful trapper varied by species; nevertheless, we detected an increasing trend for coyotes and decreasing trends for gray foxes and red foxes. Concordance across indices for gray foxes and coyotes was consistent with hypothesized declines for gray foxes and increases for coyotes in the midwestern United States. Trends for red foxes varied depending on how we accounted for potential confounding factors and it is unclear if these trends suggest population declines or distribution shifts to urban areas with reduced trapping susceptibility. Our results highlight the importance of understanding sources of variation in harvest data and that their effects can vary across species. © 2020 The Wildlife Society.  相似文献

This paper presents the results of an investigation to determine perceptions, awareness, and knowledge of the unwanted horse population in Illinois from the viewpoint of horse owners, non-horse owners, and equine industry stakeholders. A questionnaire included items that pertained to knowledge of current legislation, equine background, current methods of controlling the unwanted horse population, and methods that respondents believe would reduce the unwanted horse population in Illinois. Results indicated that 58% of horse owners viewed horses as companion animals. Respondents perceived financial hardship to be the major reason why horses become unwanted. Current methods of managing unwanted horse populations were found to be ineffective. Reducing the costs of euthanasia and carcass disposal, allowing processing facilities to reopen in Illinois, and increasing the availability of gelding programs emerged as the most effective ways to manage the unwanted horse population. Results of this survey may lead to greater awareness of the unwanted horse population in Illinois. Furthermore, these results may lead to discussions about future legislation in the State designed to support and manage unwanted horses.  相似文献

Semi-aquatic mammals are dependent upon streams and riparian areas, which are a product of the landscapes they drain. Both local stream morphology and surrounding land use are likely to have important influences on current occupancy of semi-aquatic mammals and potentially affect future geographic distributions. We identified aspects of the riparian system and stream structure at multiple scales that relate to the presence of river otter (Lontra canadensis) and mink (Neovison vison) to better understand how changing landscapes affect occupancy dynamics of these semi-aquatic mammals and to facilitate future monitoring and management. We estimated multi-season occupancy using 103 sites sampled over 6 seasonal sampling periods in southern Illinois, USA (44,526 km2) during 2012–2014. We hypothesized river otter and mink occupancy were related to multiple aspects of landscape and local habitat attributes including land cover, water availability, human disturbance, and stream characteristics. Occupancy of river otter was predicted by large stream size, less developed area near the stream site, and proximity to areas with reintroduced or remnant populations of river otter. Mink were more likely to occupy sites with small streams and decreased water availability near the site. However, top models for both species had low weights and high uncertainty for multiple variables. Habitat-based models may not be the best predictors of occupancy for these carnivores because they are more likely to respond to prey diversity or availability, but landscape changes that decrease natural water availability and increase human disturbance to the stream at the local scale are likely to negatively affect river otter. © 2019 The Authors. ABSTRACT Populations of feral domestic cats have increased throughout the United States, affecting wildlife and warranting attention from a variety of management agencies. This contentious issue requires a greater understanding of public attitudes and preferences for population control. We used data from a 2004 mail survey of Illinois, USA, homeowners’ attitudes towards wildlife and conservation to investigate support for the trap-neuter–release (TNR) of feral cats and to examine factors (demographic, experience, and wildlife values orientations) that may influence preference for TNR as a management option. Age, gender, and wildlife rights values orientations were significant predictors of preference for TNR, while negative experiences with feral cats were correlated with preferences for management options other than TNR. Investigations of public perceptions of feral cat management can help wildlife managers understand the growing public debate regarding feral cat management and provide appropriate input and educational materials regarding cat management and wildlife.  相似文献

Abstract: Establishment and spread of infectious diseases are controlled by the frequency of contacts among hosts. Although managers can estimate transmission coefficients from the relationship between disease prevalence and age or time, they may wish to quantify or compare contact rates before a disease is established or while it is at very low prevalence. Our objectives were to quantify direct and indirect contacts rates among white-tailed deer (Odocoileus virginianus) and to compare these measures of contact rate with simpler measures of joint space use. We deployed Global Positioning System (GPS) collars on 23 deer near Carbondale, Illinois, USA, from 2002 to 2005. We used location data from the GPS collars to measure pairwise rates of direct and indirect contact, based on a range of proximity criteria and time lags, as well as volume of intersection (VI) of kernel utilization distributions. We analyzed contact rates at a given distance criterion and time lag using mixed-model logistic regression. Direct contact rates increased with increasing VI and were higher in autumn—spring than in summer. After accounting for VI, the estimated odds of direct contact during autumn—spring periods were 5.0–22.1-fold greater (depending on the proximity criterion) for pairs of deer in the same social group than for between-group pairs, but for direct contacts during summer the within:between-group odds ratio did not differ significantly from 1. Indirect contact rates also increased with VI, but the effects of both season and pair-type were much smaller than for direct contacts and differed little as the time lag increased from 1–30 days. These results indicate that simple measures of joint space use are insufficient indices of direct contact because group membership can substantially increase contacts at a given level of joint space use. With indirect transmission, however, group membership had a much smaller influence after accounting for VI. Relationships between contact rates and season, VI, and pair-type were generally robust to changes in the proximity criterion defining a contact, and patterns of indirect contacts were affected little by the choice of time lag from 1–30 days. The use of GPS collars provides a framework for testing hypotheses about the form of contact networks among large mammals and comparing potential direct and indirect contact rates across gradients of ecological factors, such as population density or landscape configuration.  相似文献
