Detergent permeabilized Euplotes eurystomus (a fresh water hypotrichous ciliate) was reacted with monoclonal and polyclonal antibodies specific for either detyrosinated or tyrosinated alpha-tubulin (Glu- or Tyr-tubulin). The isolated cytoskeleton-nuclear complex was examined by Western immunoblotting and by immunofluorescent and electron microscopic methods. Both Glu- and Tyr-tubulins were detected by immunoblot analysis. Immunofluorescent microscopy indicated that the alpha-tubulin isotypes are concentrated in different regions of permeabilized cells: Glu-tubulin is located primarily in cirri, membranelles, and surrounding the macro- and micronuclei. Tyr-tubulin is principally at the bases of cirri and membranelles. This differential distribution of alpha-tubulin isotypes is discussed in terms of current concepts concerning the correlation of tubulin post-translational modifications to microtubule stability. Confocal immunofluorescent imaging was of critical importance in clearly differentiating the Glu-tubulin isotype surrounding the macro- and micronuclei from a brilliantly fluorescent environment originating from cytoskeletal structures. In conjunction with conventional and stereo-electron microscopy, confocal optical microscopy provided convincing evidence for a "basket" of microtubules surrounding both nuclei.  相似文献

为研究纤毛虫在不同生理条件下结构的分化及其调节机理,本文应用透射电镜术显示,营养期包囊游仆虫背、腹面皮层表膜下含3种方式排列组成的纵微管层以及深部微管;口区皮层内含高电子密度的杆状小体;口围带小腹基部含电子致密带和小腹托架,棘毛基体基部及基体下微管束形成围棘纤维篮;背纤毛基体下方也含微管结构;大核染色质附着在核膜上,核膜其他区域有规则排列的核孔。  相似文献

包囊游仆虫休眠包囊中,各类纤毛器的纤毛基体上方的大部分纤毛杆退化,或仅保留毛基体,有时部分额腹棘毛的毛基体也瓦解消失。残留纤毛的纤毛杆周围微管和中央微管仍具有“9 2”结构特征,也有少数纤毛杆出现2套“9 2”微管共处于一层纤毛膜内的现象。毛基体中周围三联体微管的中央形成微管形结构聚合体,基体附属结构仅存在基体间连接及纤毛器托架的残余物;非纤毛区皮层表膜下未见微管层。纤毛区皮层含纤毛器腔周围微管层(相当于表膜下微管层)、纤毛器深部及附近的微管束和分散的微管群。并且,纤毛区皮层囊泡内含有呈不同形态的纤毛杆结构;大核核孔明显变大,核孔数目减少,核孔内膜附着染色质。  相似文献

Electrical responses upon mechanostimulation at the posterior cell end were investigated in the marine hypotrichous ciliate Euplotes vannus. A new mechanostimulator was developed to mimic stimuli that are identical with those involved in cell-cell collisions. The receptor potential hyperpolarized by 18–35 mV within 12–25 msec, reached a peak value of -62 mV with a delay of 4–9 msec after membrane deformation, and was deactivated after 50–70 msec. Cirri were stimulated to beat accelerated backward. The corresponding receptor current exerted a similar time course with a peak of 2.4 nA. The shift of the reversal potential by 57.6 mV at a tenfold increase of [K+] 0 identifies potassium ions as current carriers within the development of the receptor potential. An intracellular K concentration of 355 mmol/liter was calculated for cells in a medium that was composed similar to sea-water. The mechanically activated potassium current was totally inhibited by extracellular TEA and intracellular Cs+, and partially inhibited by extracellular 4-AP. The total inhibition of the current by injected EGTA points to a Ca dependence of the posterior mechanosensitivity. It was confirmed by the increase of the peak current amplitude with rising [Ca2+] 0 . Sodium presumably repolarizes the receptor potential because the repolarization was delayed and after-depolarizations were eliminated in media without sodium. 在伍氏游仆虫(Euplotes woodruffi)接合后体发育过程中,已呈退化状态的老大核后碎块,在细胞第二次形态发生时,逐渐恢复其正常形态结构。T形新大核原基向后延伸而与恢复正常形态的老大核后碎块紧密靠拢。此时在光镜下观察,很容易误认为二者已融合为一。但在接合后体分裂之前,老大核后碎块再次瓦解,T形大核原基缩短成棒状而与老大核后碎块分开,此时二者界限分明。细胞分裂后,残存的老大核后碎块停留于后子虫中,最后被吸收。几个关键时期大核原基和老大核后碎块DNA含量的测定,也证明新老大核不融合。本文还讨论了老大核后碎块在有性过程中的功能。  相似文献

包囊游仆虫(Euplotes encysticus)形成包囊时,各类纤毛器中的纤毛杆被部分地或全部地吸收,毛基体被保留下来。休眠包囊中,背纤毛器的定位无明显变化,但原腹面纤毛器中的口围带和波动膜、额腹棘毛和横棘毛,以及左、右尾棘毛都按序陷入在细胞质内深处,并相互汇聚在一起。脱包囊时,纤毛结构在原毛基体上再分化,新纤毛器按口围带、横棘毛、额腹棘毛和左、右尾棘毛的顺序从细胞内显露出来。  相似文献

The taxonomy of the Iberian Leptodirini species of the section Anillochlamys Jeannel, 1909 has been revised. The proposed classification is based on the study of the genital structures of both sexes, in particular the internal sac of the aedeagus.
