The highly fluorescent dye Lucifer Yellow CH (LYCH), now in common use in microinjection studies, has been shown to enter the vacuole of a range of plant-cell protoplasts from the external medium. Uptake was quantified by lysing the protoplasts following incubation and determining the amount of LYCH incorporated by spectrofluorimetry. Uptake was biphasic with respect to both time and substrate concentration, enhanced at low pH and inhibited by low temperature and metabolic inhibitors. The kinetics of uptake showed several similarities with those reported for the fluid-phase endocytosis of LYCH in animal cells and yeast cells. A calculated membrane permeability coefficient for LYCH, based on the observed rates of uptake, was too high to be consistent with simple diffusion of the undissociated form of the molecule and inconsistent with the membrane-impermeant properties of the dye. 为检验小麦原胚基端特定位点上的外连丝型胞间连丝和开放孔道在摄取外源物质上的作用,以不透膜的阳离子铁蛋白(cationized ferritin)和萤黄(lucifer yellow CH )为示踪物,对其吸入与传布动态进行了荧光与电子显微镜观察。结果表明,这两种物质确可以以非跨膜运输的方式沿着原胚基端的特定通道进入原胚细胞。  相似文献

Arctic-boreal landscapes are experiencing profound warming, along with changes in ecosystem moisture status and disturbance from fire. This region is of global importance in terms of carbon feedbacks to climate, yet the sign (sink or source) and magnitude of the Arctic-boreal carbon budget within recent years remains highly uncertain. Here, we provide new estimates of recent (2003–2015) vegetation gross primary productivity (GPP), ecosystem respiration (Reco), net ecosystem CO2 exchange (NEE; Reco − GPP), and terrestrial methane (CH4) emissions for the Arctic-boreal zone using a satellite data-driven process-model for northern ecosystems (TCFM-Arctic), calibrated and evaluated using measurements from >60 tower eddy covariance (EC) sites. We used TCFM-Arctic to obtain daily 1-km2 flux estimates and annual carbon budgets for the pan-Arctic-boreal region. Across the domain, the model indicated an overall average NEE sink of −850 Tg CO2-C year−1. Eurasian boreal zones, especially those in Siberia, contributed to a majority of the net sink. In contrast, the tundra biome was relatively carbon neutral (ranging from small sink to source). Regional CH4 emissions from tundra and boreal wetlands (not accounting for aquatic CH4) were estimated at 35 Tg CH4-C year−1. Accounting for additional emissions from open water aquatic bodies and from fire, using available estimates from the literature, reduced the total regional NEE sink by 21% and shifted many far northern tundra landscapes, and some boreal forests, to a net carbon source. This assessment, based on in situ observations and models, improves our understanding of the high-latitude carbon status and also indicates a continued need for integrated site-to-regional assessments to monitor the vulnerability of these ecosystems to climate change.  相似文献

The induction of “petite” mutants by guanidine hydrochloride (GuHCl) is inhibited in several conditions. Anaerobiosis inhibited the induction either with or without cell multiplication. Both nalidixic acid (NA) and cycloheximide (CH) inhibited the induction of mutants. On the other hand, chloramphenicol (CAP) produced a dual effect: at low concentration it stimulated, at high concentration it inhibited, the induction. The effect of these different inhibitors on the transformation of + mother cells into by GuHCl is discussed.  相似文献

We present a nearest neighbor lattice model of the effects of modifiers on two-state enzyme catalysis of the reaction s ? p-We do not in general make the assumptions of the classical approach to cooperative catalysis that yield (1) adsorption isotherms of the same form as those for the corresponding equilibrium system and (2) a rate of the catalyzed reaction proportional to the number of occupied catalytic sites. Closed form results are obtained for two approximations, the Bragg-Williams and the quasi-chemical. The latter requires (l),but is exact for several simple cases, including the concerted model, under this condition. Under (1) it is found that an interaction between modifier and catalytic sites, whether attractive or repulsive, increases the magnitudes of the slopes of the adsorption isotherms but that interactions between identical sites (catalytic or modifier) increase these magnitudes if attractive and decrease them if repulsive. Thus, the former interaction allows for phase transitions if sufficiently attractive or repulsive, but the latter only if sufficiently attractive. Herein also lies the explanation for why the concerted model displays only “positive cooperativity”. It is further seen that it is not possible to classify a modifier as an activator or inhibitor of the catalyzed reaction solely on the basis of the sign of the interaction energy between catalytic and modifier sites. For agiven energy, the rate of the reaction may increase or decrease in response to the modifier, or it may respond biphasically. Similarly, the rate may respond biphasically to the activities of s or p, lead- ing to instabilities. The cysteine and glycine-rich protein 1 and 2 genes (CSRP1 and CSRP2) are an effective growth factor in promoting skeletal muscle growth in vitro and vivo. However, in cattle, the information on the CSRP1 and CSRP2 genes is very limited. The aim of this study was to examine the association of the CSRP1 and CSRP2 variants with growth and carcass traits in cattle breeds. Three single nucleotide variants (SNVs) were identified within the bovine CSRP1 gene, whereas CSRP2 gene has not detected any SNVs, using DNA pooled sequencing, PCR-RFLP, and forced PCR-RFLP methods. These SNVs include g. 801T>C (Intron 2), g. 46T>C (Exon 3) and g. 99C>G (Intron 3). Besides, we also investigated haplotype frequencies and linkage disequilibrium (LD) coefficients for three SNVs in all study populations. LD and haplotype structure of CSRP1 were different between breeds. The result of haplotype analysis demonstrated eight haplotype present in QC (Qinchuan) and one haplotype in CH (Chinese Holstein). Only haplotype 1 (TTC), shared by all two populations, comprised 10.74% and 100.00%, of all haplotypes observed in QC and CH, respectively. Haplotype 5 (CTC) had the highest haplotype frequencies in QC (30.98%) and haplotype 1 had the highest haplotype frequencies in CH (100.00%). The statistical analyses indicated that one single SNV and 19 combined haplotypes were significantly or highly significantly associated with growth and carcass traits in the QC cattle population (P < 0.05 or P < 0.01). Quantitative real-time PCR (qRT-PCR) analyses showed that the bovine CSRP1 and CSRP2 genes were widely expressed in many tissues. The results of this study suggest that the CSRP1 gene possibly is a strong candidate gene that affects growth and carcass traits in the Chinese beef cattle breeding.  相似文献

以亚热带常见树种米槠、木荷、浙江桂、罗浮栲、杉木和柑橘为对象,利用控制试验研究了温度对树木叶片甲烷(CH4)排放的影响.结果表明:当温度在10℃时,供试的6种树木中,仅木荷、柑橘和罗浮栲的叶片排放CH4;温度高于20℃时,所有树木叶片均可排放CH4.温度高于30℃时,叶片排放CH4的平均排放速率(1.010ngCH4·g-1DM·h-1)是10~30℃时平均排放速率(0.255ngCH4·g-1DM·h-1)的3.96倍.增温对柑橘和杉木CH4排放速率的影响显著高于其他4种树木.培养时间对叶片排放CH4速率有显著影响,温度胁迫对树木排放CH4的影响受植物活性的控制.在低温或高温条件下,树木干叶均不能排放CH4.高温胁迫对树木叶片排放CH4有重要影响,全球变暖可能增加植物的CH4排放.  相似文献

以中国科学院辽宁沈阳农田生态系统国家野外科学观测研究站连续两年的试验平台为依托,以潮棕壤为供试土壤,开展了稳定性氮肥配合秸秆还田对水稻产量及N2O和CH4排放的影响研究,设置对照(CK)、尿素(U)、尿素+脲酶抑制剂+硝化抑制剂(U+I)、秸秆还田(S)、秸秆还田+尿素(S+U)、秸秆还田+尿素+脲酶抑制剂+硝化抑制剂(S+U+I)6个处理.结果表明: 与CK相比,尿素显著提高了水稻产量、N2O和CH4累积排放及全球增温潜势.硝化抑制剂和脲酶抑制剂与尿素配施可显著减缓N2O的累积排放.秸秆还田显著增加了N2O和CH4累积排放、全球增温潜势和温室气体排放强度.S+U+I处理水稻产量最高,但温室气体排放强度也显著高于其他处理;U+I处理产量略低于S+U+I,但温室气体排放强度最小.秸秆单独还田处理作物产量与对照相比无显著差异.在东北潮棕壤发育的水田中,S+U+I和U+I是相对较优的施肥模式.  相似文献
