Global amphibian declines have fueled an increased interest in amphibian assisted reproductive technologies. Within the genus Rhaebo, half of the species are experiencing decreasing population trends; however, insufficient information is available on many of these species’ reproductive biology. Using the smooth-sided toad, Rhaebo guttatus, we present effective methods for collecting and cryopreserving an example of Rhaebo sperm. Specifically, our findings show that administering 10 IU/g body weight of hCG (human chorionic gonadotropin) yields the most motile and concentrated sperm and that cryopreserving spermic urine in a solution of 5% DMFA (N,N-Dimethylformamide) and 10% trehalose returns sperm with a 33 ± 3% average post-thaw motility. These findings may represent an important step forward in developing techniques that can be safely applied to other, more vulnerable species within the Rhaebo genus.  相似文献

We present a comprehensive survey of genetic variation across the range of the narrowly distributed endemic Yosemite toad Bufo canorus, a declining amphibian restricted to the Sierra Nevada of California. Based on 322 bp of mitochondrial cytochrome b sequence data, we found limited support for the monophyly of B. canorus and its closely related congener B. exsul to the exclusion of the widespread western toad B. boreas. However, B. exsul was always phylogenetically nested within B. canorus, suggesting that the latter may not be monophyletic. SSCP (single-strand conformation polymorphism) analysis of 372 individual B. canorus from 28 localities in Yosemite and Kings Canyon National Parks revealed no shared haplotypes among these two regions and lead us to interpret these two parks as distinct management units for B. canorus. Within Yosemite, we found significant genetic substructure both at the level of major drainages and among breeding ponds. Kings Canyon samples show a different pattern, with substantial variation among breeding sites, but no substructure among drainages. Across the range of B. canorus as well as among Yosemite ponds, we found an isolation-by-distance pattern suggestive of a stepping stone model of migration. However, in Kings Canyon we found no hint of such a pattern, suggesting that movement patterns of toads may be quite different in these nearby parklands. Our data imply that management for B. canorus should focus at the individual pond level, and effective management may necessitate reintroductions if local extirpations occur. A brief review of other pond-breeding anurans suggests that highly structured populations are often the case, and thus that our results for B. canorus may be general for other species of frogs and toads.  相似文献

Variation in the content and pattern of brain gangliosides from three anurans and two urodeles was analysed by thin-layer chromatography. The pattern for all three anurans was basically similar, though quantitative differences among species were detected. Ganglioside patterns from the two salamanders were distinctively different from that of the anurans and from one another, and more closely resembled the patterns seen in other tetrapods. Ganglioside variations in amphibians appear to reflect phylogenetic relationships better than life history or ecological factors.  相似文献

Aim The species‐rich family of true toads (Anura: Bufonidae) has been the focus of several earlier studies investigating the biogeography of geographically widespread taxa. Herein, we employ newly developed Bayesian divergence estimate methods to investigate the biogeographical history of this group. Resulting age estimates are used to test several key temporal hypotheses including that the origin of the bufonid clade pre‐dates Gondwanan vicariance (~105 million years ago, Ma). Area cladograms are also invoked to investigate the geographical origin of the family. Location Worldwide, except the Australia–New Guinea plate, Madagascar and the Antarctic. Methods A phylogenetic hypothesis of the relationships among true toads was derived from analysis of 2521 bp of DNA data including fragments from three mitochondrial (12S, tRNAval, 16S) and two nuclear (RAG‐1, CXCR‐4) genes. Analysis of multiple, unlinked loci with a Bayesian method for estimating divergence times allowed us to address the timing and biogeographical history of Bufonidae. Resulting divergence estimates permitted the investigation of alternative vicariance/dispersal scenarios that have been proposed for true toads. Results Our area cladogram resulting from phylogenetic analysis of DNA data supports a South American origin for Bufonidae. Divergence estimates indicate that the family originated earlier than had been suggested previously (78–99 Ma). The age of the enigmatic Caribbean clade was dated to the late Palaeocene–early Eocene. A return of bufonids to the New World in the Eocene was followed by rapid diversification and secondary expansion into South America by the early Oligocene (Rupelian). Main conclusions The South American origin of Bufonidae in the Upper Cretaceous was followed by relatively rapid expansion and radiation around the globe, ending with a return to the Americas via a Eurasian/North American land bridge in the Eocene. Though the exact route of this dispersal (Beringia or North Atlantic) remains unclear, an argument is made for the less frequently invoked North Atlantic connection. The origin of the enigmatic Caribbean lineage was found to be consistent with colonization following the bolide impact at the K/T boundary. These findings provide the first, firm foundation for understanding true toad divergence times and their truly remarkable and global radiation.  相似文献

We determined and compared the efficiency of thermoregulation of Rhinella arenarum in the Monte desert (Argentina) in two seasons, dry and wet. In the field, we measured body temperatures, micro-habitat temperatures and operative temperatures; while in the laboratory; we measured the selected body temperatures. Our results show a change in the thermoregulatory strategy of R. arenarum that is related to environmental constraints on their thermal niche. R. arenarum has the ability to be plastic and combine two strategies: (i) a moderate thermoregulator in the wet season, where thermal resources are available such that body temperature is maintained within the set points and physiologicial and behavioral processes are optimized; and (ii) a thermoconformer in the dry season where the thermal environment is more homogeneous and there is greater time invested in searching for food.  相似文献

Here I report on 12 microsatellite loci designed for populations of the Great Plains toad (Bufo cognatus) in the deserts of the southwestern USA. Diversity at these loci measured for 134 individuals from four breeding aggregations was relatively high with seven to 34 alleles per locus (mean = 17.8). Observed heterozygosities ranged from 0.444 to 0.949 and expected heterozygosities ranged from 0.597 to 0.951. These markers will be useful for studies of population genetic structure, parentage and relatedness in this explosively breeding amphibian.  相似文献

Members of the Family Bufonidae, true toads, are famous for their endogenously synthesized cardioactive steroids that serve as defensive toxins. Evolution of resistance to these toxins is not understood. We sequenced a key region of the toxin''s binding site in the Na+/K+ ATPase for relevant taxa representing Hyloidea (including bufonids), Ranoidea and Archaeobatrachia and tested for positive selection in a phylogenetic context. Bufonidae were distinct from other Hyloidea at 4–6 of 12 sites and, with one exception, had a homologous amino acid sequence. Melanophryniscus stelzneri had a distinct sequence, consistent with other independent evidence for a differentiated toxin. Tests within Bufonidae detected positive selection within the binding region, providing, to our knowledge, the first evidence of this type for positive selection within Amphibia. There was no evidence for positive selection on Bufonidae or M. stelzneri lineages. Sequence change in Leptodactylus ocellatus, a leptodactylid predator of Bufonidae, provides a molecular basis for predator resistance possibly associated with gene duplication.  相似文献

The composition of indolealkylamines of Bufo rubescens cutaneous secretions was compared to those from six other Brazilian bufonids. Skin, parotoid and tibial gland secretions were obtained for analysis by thin-layer chromatography. A triple-quadrupole mass spectrometer was used to confirm the indolealkylamines standards (serotonin, 5-HT; bufotenin, BTN; dehydrobufotenin, DHB and bufotenidin, BTD). We observed clear variation in the composition of indolealkylamines of the cutaneous secretions studied and also between those found in the skin and parotoid gland secretions of the same species. We discuss the utility of indolealkylamines to the phylogeny of this group of toads.  相似文献

Relationships among 14 species of North American Bufo were studied using the quantitative immunological technique of micro-complement fixation. Separate antisera to serum albumins of five North American Bufo and one Central American Bufo were used as probes of phylogenetic relationships. A cladogram for these species constructed with the albumin data was compatible with phylogenetic interpretations based on morphological analyses and hybridization studies. Using albumin as a molecular dating device radiations within the B. americanus, B. cognatus and B. boreas species groups were each estimated to be Pleistocene events. Separation of North and Central American lineages is estimated to have occurred as early as the upper Oligocene.  相似文献

The treatment advocated requires coding of every multistate character in additive binary form and accepts, but does not require, assignment of direction to the transformations. For each binary character is computed the ratio of the number of incompatibilities observed to the number of incompatibilities expected on the null hypothesis of the random distribution of its states. At this stage the 'noisy' characters are identified by their high ratios. These ratios are used as the basis for selecting a set of core characters common to several large compatible sets (cliques), plus the several characters particular to each large clique, followed by those characters which add further resolution or corroboration with low levels of homoplasy. The 'labelling' procedure of Guise, Peacock & Cleaves (1982) is adapted to identify 'noisy' taxa and individual character scores which are likely to be homoplastic. The procedure facilitates recognition of the robust portions of a dendrogram, alternative further resolutions, and remaining areas of uncertainty.
For the purposes of illustration, data for African toads of the Nectophrynoides lineage are analysed and the resulting dendrogram is plotted on the map of Africa. Some observations are also made on data for lizards of the family Pygopodidae and on birds of the family Alcidae.  相似文献
