Mycotoxins are metabolites of molds frequently found on and in agricultural commodities, food and feeds. Owing to their demonstrated acute, sub-acute and, in some cases, chronic toxicity, an effort has been made, worldwide, to control human and animal exposure to these toxic chemicals. This effort depends upon the availability of validated analytical methods for their detection and quantitation. This paper outlines the methodology available, and the procedures used to validate, i.e. evaluate, these methods based on the use of interlaboratory collaborative studies and the application of the HORRAT.  相似文献

Recent changes in Brazilian legislation for commercial disinfectants have been published due to the recent epidemic of nosocomial infections caused by rapidly growing mycobacteria (RGM) in many states of Brazil over the last 8 years. One of these documents requires that all the manufacturers provide evidence of efficacy of sterilizing and disinfectant products, used for semi critical medical devices, against the Mycobacterium bovis BCG Moreau and Mycobacterium abscessus subsp. bolletii INCQS 00594 strains by using the Confirmative in vitro Test for Determining Tuberculocidal Activity of Disinfectants recommended by the Association of Official Analytical Chemists. These changes have caused additional costs and increased problems for importation of enrichment products at national laboratories where disinfectant efficacy assay service is performed. Middlebrook ADC Enrichment (ADC) is provided by a unique manufacturer and used in the official protocol. The aim of the present study was to evaluate an alternative in house low-cost enrichment composed of fetal bovine serum and glucose (FBSG) with ADC for performance of disinfectant efficacy assay against mycobacteria. After obtaining the growth curves for M. abscessus ATCC 19977, M. abscessus subsp. bolletii INCQS 00594, Mycobacterium chelonae ATCC 35752, and Mycobacterium fortuitum ATCC 6841 by using ADC enrichment and FBSG in Kirchners and 7H9 culture media. Through statistical analysis via the Kruskal-Wallis test on the evaluation of microorganism growth rate, it was observed that there was no inhibition of RGM growth by any of the enrichments used. These results suggest that low-cost enrichment FBSG may be used as a potential substitute of ADC for composition of media for mycobacterial growth, including in disinfectant tests.  相似文献

Zinc is an essential nutrient that plays an important role in several biological processes of living organisms. When bound to an organic substrate, Zn is more efficiently absorbed by organisms, has a high biological activity and a low toxicity. Due to its ability to incorporate metals, yeast biomass has been used frequently as a delivery vehicle for many mineral supplements. This study describes the screening of strains of yeast for production of biomass enriched with Zn by submerged fermentation. Five strains of yeasts, belonging to the genera Saccharomyces, Kluyveromyces and Pichia, were evaluated. The highest Zn concentration was 6820 mg/kg of dry weight biomass, using Pichia guilliermondii Wickerham LPB 063 after 120 h of cultivation in a medium with 0.5 g/L ZnSO4. Process conditions were optimized using statistical experimental design methodology. Four parameters were identified in the 28−4 fractional factorial design as having a significant effect on Zn accumulation: ZnSO4 and Fe2(SO4)3 concentrations, time of addition of the ZnSO4 solution and concentration of soybean molasses. In the 32 experimental design, the influence of ZnSO4 and Fe2(SO4)3 concentrations were studied more closely. The highest Zn concentration (75,090 mg/kg dry weight) in the biomass was reached using the conditions: ZnSO4, 10.0 g/L; Fe2(SO4)3, 0.1 g/L in Erlenmeyer flasks. A batch liquid fermentation was carried out in a 2 L bioreactor for production of P. guilliermondii Wickerham LPB 063 containing organically bound Zn. The concentration of organically bound Zn after 144 h of fermentation was of 96,030 mg/kg, with a biomass production of 30 g/L. The maximum specific growth rate obtained (μmax) was 0.0077/h, while the maximum productivity of biomass was at 0.1511 g/L/h.  相似文献

Dinoflagellates are microalgae that are associated with the production of many marine toxins. These toxins poison fish, other wildlife and humans. Dinoflagellate-associated human poisonings include paralytic shellfish poisoning, diarrhetic shellfish poisoning, neurotoxic shellfish poisoning, and ciguatera fish poisoning. Dinoflagellate toxins and bioactives are of increasing interest because of their commercial impact, influence on safety of seafood, and potential medical and other applications. This review discusses biotechnological methods of identifying toxic dinoflagellates and detecting their toxins. Potential applications of the toxins are discussed. A lack of sufficient quantities of toxins for investigational purposes remains a significant limitation. Producing quantities of dinoflagellate bioactives requires an ability to mass culture them. Considerations relating to bioreactor culture of generally fragile and slow-growing dinoflagellates are discussed. Production and processing of dinoflagellates to extract bioactives, require attention to biosafety considerations as outlined in this review.  相似文献
