High level of divergence of male-reproductive-tract proteins, between Drosophila melanogaster and its sibling species, D. simulans 总被引:1,自引:0,他引:1
We compared male-reproductive-tract polypeptides of Drosophila melanogaster
and D. simulans by using two-dimensional gel electrophoresis. Approximately
64% of male-reproductive-tract polypeptides were identical between two
randomly chosen isofemale lines from these two species, compared with 83%
identity for third-instar imaginal wing-disc polypeptides. Qualitatively
similar differences were found between reproductive tracts and imaginal
discs when D. sechellia was compared with D. melanogaster and with D.
simulans. When genic polymorphism was taken into account, approximately 10%
of male- reproductive-tract polypeptides were apparently fixed for
different alleles between D. melanogaster and D. simulans; this proportion
is the same as that found for soluble enzymes by one-dimensional gel
electrophoresis. Strikingly, approximately 20% of male-reproductive- tract
polypeptides of either D. melanogaster or D. simulans had no detectable
homologue in the other species. We propose that proteins of the Drosophila
male reproductive tract may have diverged more extensively between species
than have other types of proteins and that much of this divergence may
involve large changes in levels of polypeptide expression.

A double blind study was undertaken to determine the effectiveness or oral prostaglandin E2 as a means of improving the pelvic score prior to induction of labour. 48 patients who were greater than 37 were gestation and who had Bishop scores of less than 6 entered the study. Ten tablets were given on an hourly regime. Of 25 patients in the prostaglandin group, 17 were considered successes (68.0%), whereas of 23 patients who received a placebo, 9 were successes (39.1%). No adverse effects were recorded. Prostaglandin E2 is therefore considered a safe and effective method for priming the unfavourable cervix prior to induction of labour. 相似文献

Cytochrome P-448 (mol wt 55,000 Daltons) from rabbit liver was purified to a specific content of 16.6 nmol/mg. Mice were immunised with this preparation, their spleens removed and dissociated lymphocytes hybridised with myeloma cells. Four monoclonal antibodies against cytochrome P-448 were raised and partially characterised. All four antibodies interacted with cytochrome P-448 in intact microsomal fractions and selectively immunoadsorbed cytochrome P-448 from solubilised microsomal preparations. One of the antibodies inhibited benzo[a] pyrene hydroxylase activity in a reconstituted system, one had no effect on activity and two increased activity. The bioavailability of iron from a new commercial source containing ferric gluconate stabilized with glycine sold under the
trade name Bioferrico™ was studied in this work by means of the prophylactic-preventive test in rats. NaFeEDTA was also studied
by the same methodology for comparative purposes and ferrous sulfate was used as the reference standard. The test was conducted
for 4 wk with male weaned rats, which were randomized into four groups of at least eight animals each. A control group received
a basal diet of low-iron content, whereas the other groups received the same diet with iron added at a dose of 20 mg/kg as
FeSO4·7H2O, NaFeEDTA, and Bioferrico, respectively. Individual hemoglobin concentrations (HbC) and weights were determined at the beginning
and at the end of the study and food intake was daily registered. The iron bioavailability (BioFe) of each source was calculated
as the ratio between the amount of iron incorporated into hemoglobin during the treatment (HbFe) and the total iron intake
per animal (ToFeIn). A relative biological value (RBV) was obtained for each iron source under study as the ratio between
the BioFe of the tested compound and that of the reference standard. The RBVs were 98% and 86% for Bioferrico and NaFeEDTA,
respectively. Bioferrico showed a high bioavailability and behaved inertly in relation to the sensorial properties of the
fortified food when it was added to flour. These qualities emphasize Bioferrico as a promising source for iron fortification. 相似文献

We have previously reported that altered culture conditions (a broth media with shaking) could induce a strain of Helicobacter pylori to assume a long spiral morphology resembling that described for Helicobacter heilmannii. In order to evaluate if snorkeling had significant effects on coral community structure, three different coral reefs (Madrizquí, Pelona de Rabusquí and Crasquí) located at Archipelago Los Roques National Park, Venezuela, were surveyed. For each site, the coral community structure of two different areas, one subjected to intense snorkeling use (FB) and other not frequently used (PFB), were compared. Community structure was determined with 1 m2-quadrants and 20 m-long transects. These communities were described in terms of species richness, diversity (Shannon-Wiener) and evenness indexes, live and dead coral cover and cover of other organisms (sponges, octocorals and algae). Comparisons within sites were performed with a Kruskall-Wallis test. A total of 24 species of scleractinian corals were found. Live coral cover ranged from 29.9% +/- 26.43 (Crasquí) to 34.55% +/- 6.43 (Madrizquí), while dead coral cover ranged from 32.51% +/- 2.86 (Madrizquí) to 60.78% +/- 21.3 (Pelona de Rabusquí). The PFB areas showed higher live coral cover compared to FB areas; however, significant differences were only found in Crasquí and Pelona de Rabusquí (p < 0.05). Species richness, diversity and evenness were variable and no trends were observed between FB and PFB areas. The frequency of both damaged and diseased colonies were low (< 1%), most damages observed were natural (parrotfish predation). Damages caused by divers such as fin impacts, were not found at the reefs studied. These results suggest that, currently, diving pressure is not as high to cause massive loses of live coral cover in these reefs. However, the lack of strict controls for these activities might produce long-term changes in the structure of these coral communities. 相似文献

Low intensity resistance exercise (RE) with blood flow restriction (BFR) has gained attention in the literature due to the beneficial effects on functional and morphological variables, similar to those observed during traditional RE without BFR, while the effects of BFR on post-exercise hypotension remain unclear. The aim of the present study was to compare the blood pressure (BP) response of trained normotensive individuals to RE with and without BFR. In this cross-over randomized trial, eight male subjects (23.8 ± 4 years, 74 ± 3 kg, 174 ± 4 cm) completed two exercise protocols: traditional RE (3 x 10 repetitions at 70% one-repetition maximum [1-RM]) and low intensity RE (3 x 15 repetitions at 20% 1-RM) with BFR. Blood pressure measurements were performed after 15 min of seated rest (0), immediately after and 10 min, 20 min, 30 min, 40 min, 50 min and 60 min after the experimental sessions. Similar hypotensive effects for systolic BP (SBP) were observed for both protocols (P < 0.05) after exercise, with no differences between groups (P > 0.05) and no statistically significant difference for diastolic BP (P > 0.05). These results suggest that in normotensive trained individuals, both traditional RE and RE with BFR induce hypotension for SBP, which is important to prevent cardiovascular disturbances. 相似文献

In triosephosphate isomerase, Cys126 is a conserved residue located close to the catalytic glutamate, Glu165. Although it has been mentioned that Cys126 and other nearby residues are required to maintain the active site geometry optimal for catalysis, no evidence supporting this idea has been reported to date. In this work, we studied the catalytic and stability properties of mutants C126A and C126S of Saccharomyces cerevisiae TIM (wtTIM). None of these amino acid replacements induced significant changes in the folding of wtTIM, as indicated by spectroscopic studies. C126S and C126A have K(M) and k(cat) values that are concomitantly reduced by only 4-fold and 1.5-fold, respectively, compared to those of wtTIM; in either case, however, the catalytic efficiency (k(cat)/K(M)) of the enzyme is barely affected. The affinity of mutated TIMs for the competitive inhibitor 2-phosphoglycolate augmented also slightly. In contrast, greater susceptibility to thermal denaturation resulted from mutation of Cys126, especially when it was changed to Ser. By using values of the rate constants for unfolding and refolding, we estimated that, at 25 degrees C, C126A and C126S are less stable than wtTIM by about 5.0 and 9.0 kcal mol(-)(1), respectively. Moreover, either of these mutations slows down the folding rate by a factor of 10 and decreases the recovery of the active enzyme after thermal unfolding. Thus, Cys126 is required for proper stability and efficient folding of TIM rather than for enzymatic catalysis. 相似文献
