Effects of Extracellular Potassium on Acid Release and Motility Initiation in Toxoplasma gondii

The internal pH (pH_i) of Toxoplasma gondii was estimated by measuring the accumulation of the weak base 9-aminoacridine in buffers with various ionic compositions. The pH_i of the metabolizing parasite increased when the extracellular K⁺ was elevated in alkaline medium or when the external pH (pH_c) was substantially increased in medium employing high external K⁺ (90 mM). The parasite in mouse peritoneal fluid, or in potassium sulfate buffer (pH 8.2), where the pH_i was demonstrated to be increased to 7.9, became motile when acidic buffer was substituted for the original suspension medium. This acid-induced independent movement subsided within 5 min but was repeatedly induced if the pH_c was serially lowered to 6.0. Basic buffers, on the other hand, abolished motility when applied to the moving parasites. Nigericin, which is known to collapse pH gradients across the membrane, also abolished motility.     

Restriction Enzyme Analysis of Mitochondrial DNA of Acanthamoeba Strains in Japan

ABSTRACT. Eight isolates, identified as either Acanthamoeba castellanii or A. polyphaga from human eye infections, contact lens containers, and soil in Japan, were characterized by restriction fragment length polymorphisms (RFLP) of mitochondrial DNA (mtDNA). Mitochondrial DNA was digested with either Bgl II, Eco R I, Hind III, Hpa I, Sca I or Xba I, electrophoresed in agarose gels, and stained with ethidium bromide. Four distinct RFLP phenotypes that refer to the collection of six fragment size patterns obtained for a single strain with six enzymes, were discovered among the eight strains used in this study. Three strains morphologically classified as A. polyphaga share a single RFLP phenotype with the Ma strain of A. castellanii. The interspecific sequence differences of 7.06–12.74% in DNA nucleotide were estimated from the proportion of DNA fragments shared by each pair of mtDNA.     

ROLE OF GLUCOCORTICOIDS IN TERMINAL DIFFERENTIATION AND TISSUE-SPECIFIC FUNCTION (A REVIEW)

Glucocorticoids (GCs) are required by many kinds of organs, tissues and cells for initiation or maintenance of their specific functions in vivo and in vitro. It is noticeable that most of these GC actions can be induced at much lower levels of dosage or concentration than the well-known actions of the steroids in gluconeogenesis, immune suppression and anti-inflammation. Such "differentiation-stimulating" actions of GC are regarded as main physiological roles of the steroid.     

Peroxidase isozymes and their indoleacetate oxidase activity in the Japanese morning glory, Pharbitis nil

Distributions of peroxidase isozymes in various organs of theJapanese morning glory (Pharbitis nil CHOISY) are described.Most peroxidase isozymes had indoleacetate oxidase activity. (Received December 29, 1969; )     

Anti-oxidation activity of ethanol extracts from natural thalli of lichens

Screening test on anti-oxidation activity using 1,1-diphenyl-2-picrylhydrazyl(DPPH) was performed for 99 ethanol extracts of 85 species of natural thalli of lichens in order to find novel anti-oxidation compounds.The 17 extracts of natural thalli showed high anti-oxidation activity.Among them,the activities of extracts from Hypogymnia vittata,Peltigera aphthosa,Nephromopsis ornata,Pseudevernia furfuracea,Cladonia vulcani and Peltigera elizabethae were higher.Extracts of Peltigera spp.showed higher activity than those of other genera.The ethanol extract of P.aphthosa had been separated into ethyl acetate-soluble and water-soluble fractions.Two anti-oxidative spots were found only in the water-soluble fractions by thin-layer chromatography.The compound in the lower spot had the same Rf value,UV spectrum,and color as authentic solorinine that was previously found as a unique quaternary ammonium compound from Peltigera spp.We now report that the hydrophilic lichen substance,solorinine showed a nearly same anti-oxidation activity(EC50=120?mol/Lol/L) as standard antioxidant Trolox(EC50=150?mol/L).     

Isolation of the Antheridiogen of Anemia phyllitidis

The antheridiogen (antheridium-inducing substance) of the fern species Anemia phyllitidis has been obtained in pure form based on the isolation procedure described below. Pure antheridiogen is active to a dilution of 10 μg/l in antheridium formation and 0.3 μg/l in dark-germination. Its molecular formula is C₁₉H₂₂O₅.     

Neuroendocrine Regulation of the Development of Seasonal Forms of the Asian Comma Butterfly, Polygonia c-aureum L

In the butterfly, Polygonia c-aureum , development of seasonal forms controlled by the photoperiod and temperature was shown to involve a neuroendocrine system of the brain-corpus cardiacum-corpus allatum complex.
For analysis of the neuroendocrine system concerned, the innervation of the complex was investigated first by cobalt chloride perfusion staining and then by severance of axons, ablation of the candidate cells, injection of a homogenate of these cells and transplantation of corpora cardiaca using pupae programmed to be either summer-form or autumn-form adults.
The results suggested that medial nerve cells produce what is called material producing the summer form.
The seasonal forms of the Asian comma butterfly, Polygonia c-aureum L., summer and autumn forms (Fig. la, b), are determined by the photoperiod and the temperature during the larval period (1–3). Previous studies have given the following results on the physiological mechanism involved in the effect of environmental factors in inducing these seasonal forms. First, the mechanism involves neurosecretory cells located somewhere in the brain (2). Second, the nervous connections between the brain and the corpus cardiacum (NCC I+II (4)) and between the right and left brain lobes are indispensable for the effect (2, 5–7).
The present study consisted of two series of experiments. One was designed to demonstrate morphologically the axonal connection of the corpus cardiacum with the corpus allatum in this butterfly, like that shown in several other insects (8–13). The other series was designed to locate the neurosecretory cells producing material related to the seasonal form and to see if this material is also present in the corpus cardiacum.
