Abstract Reference strains of ' Pseudomonas diazotrophicus ' produce a range of polar lipids atypical of authentic Pseudomonas species. In addition to the phospholipids common in Gram-negative bacteria (phosphatidylglycerol, diphosphatidylglycerol, and phosphatidylethanolamine), N -methylated derivatives of phosphatidylethanolamine (including phosphatidylcholine) and an ornithine amide lipid are also present. The preponderant ester-bound fatty acid (up to 80% of the total) is cis -vaccenic acid ( cis -octadec-11-enoic acid), while 3-hydroxyoctadecanoic acid is the major amide-bound fatty acid in the ornithine lipid. Possible implications of the data for classification of the organism are discussed.  相似文献

Human cytomegalovirus (HCMV) US2, US3, US6 and US11 act in concert to prevent immune recognition of virally infected cells by CD8+ T-lymphocytes through downregulation of MHC class I molecules (MHC-I). Here we show that US2 function goes far beyond MHC-I degradation. A systematic proteomic study using Plasma Membrane Profiling revealed US2 was unique in downregulating additional cellular targets, including: five distinct integrin α-chains, CD112, the interleukin-12 receptor, PTPRJ and thrombomodulin. US2 recruited the cellular E3 ligase TRC8 to direct the proteasomal degradation of all its targets, reminiscent of its degradation of MHC-I. Whereas integrin α-chains were selectively degraded, their integrin β1 binding partner accumulated in the ER. Consequently integrin signaling, cell adhesion and migration were strongly suppressed. US2 was necessary and sufficient for degradation of the majority of its substrates, but remarkably, the HCMV NK cell evasion function UL141 requisitioned US2 to enhance downregulation of the NK cell ligand CD112. UL141 retained CD112 in the ER from where US2 promoted its TRC8-dependent retrotranslocation and degradation. These findings redefine US2 as a multifunctional degradation hub which, through recruitment of the cellular E3 ligase TRC8, modulates diverse immune pathways involved in antigen presentation, NK cell activation, migration and coagulation; and highlight US2’s impact on HCMV pathogenesis.  相似文献

The fluorescent and photo-affinity derivatives of enkephalin, Tyr-D-Ala-Gly-Phe-Leu-Lys-Nε-Rhodamine (II) and Tyr-D-Ala-Gly-Phe-Leu-Lys-Nε-nitro-azidophenyl (III), were prepared by conventional methods followed by chemical modification. The two peptides inhibit the binding of 125I-labeled enkephalin to brain membrane preparations, with apparent IC50 values of 5.9 nM and 5.5 nM for peptides II and III, respectively. The iodinated derivative of peptide III binds specifically to brain membrane preparations with an apparent Kd of about 2.1 × 10?9M.  相似文献

Overall association and dissociation rate constants were measured at 20 degrees C for O2, CO, and alkyl isocyanide binding to position 45 (CD3) mutants of pig and sperm whale myoglobins and to sperm whale myoglobin reconstituted with protoheme IX dimethyl ester. In pig myoglobin, Lys45(CD3) was replaced with Arg, His, Ser, and Glu; in sperm whale myoglobin, Arg45(CD3) was replaced with Ser and Gly. Intramolecular rebinding of NO, O2, and methyl isocyanide to Arg45, Ser45, Glu45, and Lys45(native) pig myoglobins was measured following 35-ps and 17-ns excitation pulses. The shorter, picosecond laser flash was used to examine ligand recombination from photochemically produced contact pairs, and the longer, nanosecond flash was used to measure the rebinding of ligands farther removed from the iron atom. Mutations at position 45 or esterification of the heme did not change significantly (less than or equal to 2-fold) the overall association rate constants for NO, CO, and O2 binding at room temperature. These data demonstrate unequivocally that Lys(Arg)45 makes little contribution to the outer kinetic barrier for the entry of diatomic gases into the distal pocket of myoglobin, a result that contradicts a variety of previous structural and theoretical interpretations. However, the rates of geminate recombination of NO and O2 and the affinity of myoglobin for O2 were dependent upon the basicity of residue 45. The series of substitutions Arg45, Lys45, Ser45, and Glu45 in pig myoglobin led to a 3-fold decrease in the initial rate for the intramolecular, picosecond rebinding of NO and 4-fold decrease in the geminate rate constant for the nanosecond rebinding of O2. XY/XYY sex-chromosome mosaicism was demonstrated in both bone marrow and germ cells of a wild adult common shrew. Secondary sexual characteristics were those of a normal male, but the testes were small, and the sperm count was only about 3% of normal. Most of the seminiferous tubule cross-sections examined revealed serious spermatogenic impairment and a reduced diameter. A range of sex-chromosome pairing configurations was observed in XYY primary spermatocytes, including configurations involving the X and both Y chromosomes in a linear or radial array. The presence of metaphase II (MII) spreads with an XY sex-chromosome complement indicated that XYY primary spermatocytes could contribute products to MII. Following Burgoyne (1979) and Burgoyne and Biddle (1980), a number of models of spermatocyte loss were tested. The data indicated that there was an association between the sex-chromosome complement of primary spermatocytes and their contribution to MII. The best fit to the observed MII frequency data was provided by a model which assumed that all XYY primary spermatocytes with a univalent Y chromosome and a high proportion of XYY primary spermatocytes with an unpaired X chromosome failed to contribute products to MII.  相似文献
