The acute effects on perch and long‐term effects on whitefish were studied after liming one side of a lake [initial pH: 4.6–5.5; aluminium (Al): 29–54 μg L?1; Ca2+: 0.02–0.07 mmol L?1] that was divided into two parts with a plastic curtain. Some Al precipitation was observed on the gill surface of perch 1 day after liming; the concentration of plasma sodium of perch females in the limed side was also higher than in the acidic side. Nearly 6 months after liming, at the spawning time of whitefish, the plasma chloride concentration of whitefish in the limed side was higher and blood glucose concentration lower than in the acidic side. Four of five whitefish males from the limed side, but only one of five males and none of the five females from the acidic side, were ready to spawn. The growth of whitefish in the limed side was also more rapid. These changes illustrate that liming had no acute harmful effects on perch, and allowed whitefish to recover from acidity‐related physiological stress.  相似文献

The genetic relationships among New and Old World coregonid fishes were studied by electrophoresis. The genetic composition of 60 populations, representing perhaps nine commonly recognized species of Coregonus and Stenodus from Europe and North America was determined for 37 genetic loci. Six distinct genetic groups were evident. The first contained only populations of the inconnu, Stenodus leucichthys (Güldenstadt). The Nei genetic distance between Stenodus and Coregonus was 0.305, a relatively small value as compared to other salmonid inter-generic comparisons. The second genetic grouping contained the Arctic cisco, C. autumnalis (Pallas), the N. American lake cisco, C. artedii Lesueur, and the Irish pollan, C. autumnalis pollan Thompson. These three taxa appear to be conspecific on the basis of genetic distances. The third genetic grouping contained the European whitefish, C. lavaretus (L.), and the N. American lake whitefish, C. clupeaformis (Mitchill). European and lake whitefish may be conspecific. Lake whitefish from northwestern N. America were more closely related to European whitefish (genetic distance 0.038) than to lake whitefish from central N. America (genetic distance 0.098). The fourth group contained the broad whitefish, C. nasus (Pallas), which is perhaps more closely related to the European/lake whitefish groups than other coregonids. The fifth genetic grouping contained only the Asian endemic, C. peled (Gmelin), and the sixth contained the least cisco, C. sardinella Valenciennes, and vendace, C. albula (L.), which also appear to be conspecific. The widespread genetic groupings obtained for ciscoes indicate that they do not constitute a single closely related group within the genus Coregonus.  相似文献

Contamination of the culture water by high densities of phantom midge larvae, Chaoborus sp. resulted in size diminution, retarded reproduction and decrease in the clutch size of D. pulex, and size diminution of D. magna. Furthermore, D. pulex suffered heavy mortality. The results are discussed in the context of the hypothesis that energy is expended in the formation of defensive cyclomorphic spines. Background

Peroxisome proliferator-activated receptor gamma (PPARγ) agonists are clinically used to counteract hyperglycemia. However, so far experienced unwanted side effects, such as weight gain, promote the search for new PPARγ activators.

Methods

We used a combination of in silico, in vitro, cell-based and in vivo models to identify and validate natural products as promising leads for partial novel PPARγ agonists.

Results

The natural product honokiol from the traditional Chinese herbal drug Magnolia bark was in silico predicted to bind into the PPARγ ligand binding pocket as dimer. Honokiol indeed directly bound to purified PPARγ ligand-binding domain (LBD) and acted as partial agonist in a PPARγ-mediated luciferase reporter assay. Honokiol was then directly compared to the clinically used full agonist pioglitazone with regard to stimulation of glucose uptake in adipocytes as well as adipogenic differentiation in 3T3-L1 pre-adipocytes and mouse embryonic fibroblasts. While honokiol stimulated basal glucose uptake to a similar extent as pioglitazone, it did not induce adipogenesis in contrast to pioglitazone. In diabetic KKAy mice oral application of honokiol prevented hyperglycemia and suppressed weight gain.

Conclusion

We identified honokiol as a partial non-adipogenic PPARγ agonist in vitro which prevented hyperglycemia and weight gain in vivo.

General significance

This observed activity profile suggests honokiol as promising new pharmaceutical lead or dietary supplement to combat metabolic disease, and provides a molecular explanation for the use of Magnolia in traditional medicine.  相似文献

Several plant species defend themselves indirectly from herbivores by producing herbivore-induced volatile compounds that attract the natural enemies of herbivores. Here we tested the effects of elevated atmospheric CO(2) (720 micromol mol(-1)) concentration on this indirect defense, physiological properties, and constitutive and induced emissions of white cabbage (Brassica oleracea ssp. capitata, cvs Lennox and Rinda). We monitored the orientation behavior of the generalist predator Podisus maculiventris (Heteroptera: Pentatomidae) and the specialist parasitoid Cotesia plutellae (Hymenoptera: Braconidae) to plants damaged by Plutella xylostella (Lepidoptera: Plutellidae) in the Y-tube olfactometer. Elevated CO(2) levels did not affect stomatal densities but reduced specific leaf area and increased leaf thickness in cv Lennox. In addition to enhanced constitutive monoterpene emission, P. xylostella-damaged cabbages emitted homoterpene (E)-4,8-dimethyl-1,3,7-nonatriene, sesquiterpene (E,E)-alpha-farnesene, and (Z)-3-hexenyl acetate. Growth at elevated CO(2) had no significant effect on the emissions expressed per leaf area, while minor reduction in the emission of homoterpene (E)-4,8-dimethyl-1,3,7-nonatriene and (E,E)-alpha-farnesene was observed at elevated CO(2) in one of two experiments. The generalist predator P. maculiventris discriminated only between the odors of intact and P. xylostella-damaged cv Rinda plants grown at ambient CO(2) concentration, preferring the odor of the damaged plants. The specialist parasitoid C. plutellae preferred the odor of damaged plants of both cultivars grown at ambient CO(2) but did not detect damaged cv Lennox plants grown at elevated CO(2). The results suggest that elevated atmospheric CO(2) concentration could weaken the plant response induced by insect herbivore feeding and thereby lead to a disturbance of signaling to the third trophic level.  相似文献

Responses to Potato virus A (PVA, genus Potyvirus) segregate to three phenotypic groups in a diploid cross between Solanum tuberosum subsp. andigena and a highly interspecific potato hybrid. The aim of this study was to compare gene expression between the progeny genotypes which react with hypersensitive response (HR) to PVA, allow PVA accumulation in inoculated leaves but restrict PVA infection to the inoculated leaf by blocking systemic movement [non-necrotic resistance (nnr)], or are susceptible (S) and systemically infected with PVA. Expression levels of ca 10 000 genes were compared using probes arranged in a microarray format, and real-time RT-PCR was applied for quantitative comparison of the expression of selected defense-related genes (DRGs). Results showed that a few DRGs were autoactivated in HR genotypes at an early stage of plant growth in the absence of PVA infection, which was not observed in the two other phenotypic groups (nnr and S). More detailed studies on the DRGs encoding a beta-1,3-glucanase, a chitinase and a basic PR-1b protein showed that autoactivation of the genes was not evident in vitro and up to 2 weeks of growth in soil in a controlled growth cabinet but was apparent 2 weeks later. Hence, autoinduction of these DRGs in the HR genotypes could be associated with growth stage, environmental factors or both. Furthermore, a number of other DRGs were induced in the inoculated leaves of HR genotypes as a response to infection with PVA, which was not observed in nnr and S genotypes. These results provide some novel information about factors underpinning the higher levels of virus resistance realised in potato genotypes carrying virus-specific R genes and suggest that part of the resistance is attributable to additional ‘minor’ genes functioning simultaneously, hence adding to the overall responsiveness and level of resistance against infection. These results also imply that some genotypes might be more responsive to chemical induction of pathogen and pest resistance, which could be considered in screening of progenies in plant-breeding programs.  相似文献

The formation of four Cu(II)-xylitol complexes was observed in aqueous alkaline solutions (11.0< or =pH< or =14.0, I=1.0, 20 degrees C) by means of direct current polarography and VIS spectrophotometry. Mononuclear hydroxy complexes, CuXyl(OH)- (log beta=17.7 +/- 0.5), CuXyl(OH)2(2-) (log beta=20.2 +/- 0.3) and CuXyl2(OH)2(4-) (log beta=22.4 +/- 0.3), are formed at high ligand-to-metal ratios (L:M> or =10), whereas dinuclear complex Cu2Xyl (log beta=29.2 +/- 0.3) is the predominant species at low ligand-to-metal ratio (L:M=0.5). Diffusion coefficients and molar absorptivities of the complex species were determined. pH variable 13C NMR suggested that pKa values of xylitol are rather similar and equal to 13.8 +/- 0.2, 13.9 +/- 0.1 and 13.9 +/- 0.2 for OH-groups adjacent to (C-1,C-5), (C-3) and (C-2,C-4) carbon atoms, respectively.  相似文献

The objective of this study was to apply transthoracic Doppler echocardiography (TTDE) in mice to study coronary flow reserve (CFR), an index of coronary microvascular function, in mild and severe forms of experimental viral myocarditis. Regarding methodology, BALB/c mice were infected with cardiotropic coxsackieviruses causing either a mild (Nancy strain) or a severe (Woodruff strain) myocarditis. Left ventricular dimensions, fractional shortening, and CFR (ratio of left coronary artery flow velocity during maximal adenosine-induced vasodilatation to rest) were measured by TTDE before infection and again 1 or 2 wk after infection. As a result, the resting flow velocity did not change after infection. In contrast, CFR reduced significantly 1 wk after infection with either virus variant [from 2.5 (SD 0.3) to 1.4 (SD 0.1) in severe and from 2.4 (SD 0.4) to 2.1 (SD 0.3) in mild myocarditis], being significantly lower in the severe than mild myocarditis. CFR remained low in severe myocarditis 2 wk after infection. Fractional shortening decreased to the same levels 1 wk after infection with either virus variant [from 0.54 (SD 0.02) to 0.43 (SD 0.03) in severe and from 0.51 (SD 0.03) to 0.44 (SD 0.02) in mild myocarditis, P < 0.05]. However, 2 wk after infection, mice with severe myocarditis had enlarged left ventricles and lower fractional shortening [0.31 (SD 0.03)] than mice with mild myocarditis [0.47 (SD 0.02), P < 0.01]. In conclusion, CFR measured with TTDE is reduced in coxsackievirus myocarditis in mice. Low CFR is associated with progressive heart failure, indicating that dysfunction of coronary microcirculation is a determinant of poor outcome in viral myocarditis.  相似文献

Dieback of the terminal shoot and consequently bushy growth induced by boron deficiency have been reported widely throughout the world in several tree species. Recently, similar growth damage was documented in half of the young spruce stands in eastern Finland. To clarify the role of B deficiency, the light microscopic structure of emerging buds and of developing and previous-year needles of mature Norway spruce (Picea abies L. Karst.) from damaged (D stand), partly damaged (PD stand) and healthy (H stand) stands were analysed. The samples, on which needle nutrient concentrations were also determined, were taken seven times between early spring (April) and early winter (November). Cell death characterized by precipitation of the cell content, possibly due to the release of tannins after membrane rupture, was seen in the apex of emerging buds, and this led to fatal damage in about half of the buds in the trees from the D stand, where the needle B concentration was well below the deficiency level of 4–5 mg kg−1. Furthermore, an increase in living cells that accumulated tannins in the vacuoles, which is a common stress and/or defense reaction, was found in the primordial shoots of buds and in the differentiating needles in the PD and D stands. The increase in the areas of the central cylinder and of the xylem found in the needles indicate structural plasticity during needle differentiation to drought. The time frame for bud emergence from late May up to mid-September means that an adequate B supply is necessary throughout the summer in order to avoid fatal bud damage and thus bushy growth of the trees.  相似文献

We discovered, using transfer functions, that climatic changes in the Atlantic control the abundance of Temora longicornis , a dominant pelagic copepod of the Baltic Sea. The seawater salinity was increasing and copepod numbers were high from 1960s up to 1970s. Then the freshwater runoff started to increase, which resulted in decreasing salinities and abundance of the copepod. At the end of 1990s, runoffs remained at a high level, and the decrease of surface salinities and Temora leveled off. Due to time lags between variables studied, we also make predictions of changes expectable in early 2000s. The total freshwater runoff to the Baltic Sea followed the North Atlantic Oscillation with an immediate lag. Salinity followed the runoff non-linearly with a lag of 4–9 months. Temora longicornis followed the salinity with a lag of 1–3 months. Predicted abundance of T. longicornis will remain low implicating poor feeding conditions for planktivores. Our study points out the importance of physical factors in control of pelagic environments compared to ecological interactions, such as top-down and bottom-up.  相似文献
