Isoamylase production in batchwise and fed-batch cultures of Pseudomonas amyloderomosa (strain WU7211-2) was investigated. By feeding maltose in a mode motivated by its structure gene (iam), the final isoamylase activity of 5100 U/ml was achieved, as compared to 4100 U/ml in batch cultivation and 3800 U/ml in shaken flasks. The enhancement may be due to the fact that the production of isoamylase can be induced effectively by the maltose only if the glucose concentration is maintained below the inhibitory level. Also, cultivations based on 500-ml shaken flasks, performed with different amount of proteimax HE90, showed that higher amounts of proteimax HE90 resulted in an increased value of pH that had an adverse effect on isoamylase yield.The authors wish to thank the Food Industry Research and Development Institute (FIRDI), Taiwan, R.O.C for the financial support (Cooperative Agreement 94M904-2B). To be effective, signals must propagate through the environment and be detected by receivers. As a result, signal form evolves in response to both the constraints imposed by the transmission environment and receiver perceptual abilities. Little work has examined the extent to which signals may act as selective forces on receiver sensory systems to improve the efficacy of communication. If receivers benefit from accurate signal assessment, selection could favour sensory organs that improve discrimination of established signals. Here, we provide evidence that visual resolution coevolves with visual signals in Polistes wasps. Multiple Polistes species have variable facial patterns that function as social signals, whereas other species lack visual signals. Analysis of 19 Polistes species shows that maximum eye facet size is positively associated with both eye size and presence of visual signals. Relatively larger facets within the eye''s acute zone improve resolution of small images, such as wasp facial signals. Therefore, sensory systems may evolve to optimize signal assessment. Sensory adaptations to facilitate signal detection may represent an overlooked area of the evolution of animal communication.  相似文献

A bioassay was developed to screen extracellular phytase-producing microorganisms. Washed cells of Corynebacterium glutamicum, which cannot use sodium phytate as source of phosphate, were mixed with phytate-minimal agar as indicator strain. By this method, we could easily obtain phytase-producing strains from soil samples and 71 % of the isolates had phytase activities above 0.01 U/ml when they were grown in modified phytase screening medium. © Rapid Science Ltd. 1998  相似文献

Apoplastic Ca2+ concentration controls membrane permeability, cell wall stabilization and cell integrity; however, little is known about its role in thermotolerance in plants. Here, we report that the acquired thermotolerance of etiolated rice seedlings (Oryza sativa) was abolished by an exogenously supplied Ca2+ chelator, EGTA, related to increased cellular content leakage during heat shock (HS) treatment. Thermotolerance was restored by the addition of Ca2+ during EGTA incubation. Pectin methylesterase (EC 3.1.1.11), a cell-wall remodeling enzyme, was activated in response to HS and its elevated activity was related to the recovery of the HS-released Ca2+ concentration. EGTA interfered with the capability of HS to increase oscillation of [Ca2+]cyt content. We assume that heat-activated PME activity is involved in cell-wall localized Ca2+. The removal of apoplastic Ca2+ might participate in HS signaling to induce HS protein expression and cell-wall remodeling to retain plasma membrane integrity, prevent cellular content leakage and confer thermoprotection.Key words: Ca2+, cell wall, EGTA, HSP, HSR, pectin methylesterase, thermotoleranceBiological organisms have developed a remarkable number of strategies to adapt environmental changes. Induction of heat shock protein (HSP) expression is one of the best characterized responses to elevated temperature and plays an important role in the acquisition of thermotolerance.1 Besides HSPs, Ca2+ has a role in the plant adaptation to stress,2 but little evidence of apoplastic Ca2+ homeostasis contributes to the adaptation to heat stress, which is crucial to elucidate its mechanisms. Our previous study showed that the recovery of HS-released Ca2+ in Ca2+-pectate reconstitution through pectin methylesterase (PME) activity is required for cell-wall remodeling during HS in soybean, which retains plasma membrane integrity and co-ordinates with HSPs to confer thermotolerance.3 Here, we demonstrated that administering the extracellular Ca2+ chelator EGTA to rice plants inhibited the development of thermotolerance, which could be overcome by the addition of Ca2+. We also investigated HS inducing apoplastic Ca2+ mobilization to increase cytosolic Ca2+ level and that, combined with Ca2+-pectate networks, may be a universal response for the acquisition of thermotolerance in planta.  相似文献

Genetic modified baculovirus (GMBV) are among the most promising alternatives to chemical insecticides. One of the deterrents to the GMBV development is the lack of simple and cost-effective methods for monitoring their efficacy and ecology in fields. Here, we demonstrate the DsRed gene from coral can serve as a convenient GMBV tracer. Insect larvae, including Trichoplusia ni, Spodoptera exigua, and Spodoptera litura, infected the GMBV containing the DsRed gene can emit red fluorescence under sun light without any prosthetic apparatus.  相似文献

The mushrooms of diverse Lingzhi species have been traditionally consumed as luxurious functional food supplements in Chinese society. FIP-gts, a fungal immunomodulatory protein found in Song-Shan Lingzhi (Ganodera tsugae) has been proposed to possess therapeutic effects on cancer and autoimmune diseases. To produce active FIP-gts for evaluation of oral administration, a recombinant FIP-gts (rFIP-gts) fused with a 6His-tag at its C-terminus was expressed in Sf21 insect cells by the baculovirus expression system. High yield (about 70%) and purity (about 90%) of rFIP-gts was obtained by one-step nickel-affinity chromatography. The correctness of the harvested rFIP-gts was verified by Western blot and MALDI-MS analyses. Optimal expression of rFIP-gts was observed when the Sf21 cells were infected with multiplicity of infection of 10 for 72 h, and the yield was up to 47.2 microg/3 x 10(6) infected cells. The immunomodulatory activity of the purified rFIP-gts was detected as the induction of interleukin 2 released from murine splenocytes. Compared with the rFIP-gts produced in Escherichia coli cells, the rFIP-gts produced in Sf21 cells possessed evidently higher specific immunomodulatory activity.  相似文献

The baculovirus–insect expression system is a popular tool for recombinant protein production. The standard method for infecting insect larvae with recombinant baculovirus for protein production involves either feeding occlusion bodies or injecting budded virus into the cuticle. In this study, we showed that the recombinant Autographa californica multiple nucleopolyhedrovirus (AcMNPV) at titers >108 pfu/mL efficiently infected Trichoplusia ni (T. ni) larvae through aerosol inoculation of budded virus at a pressure of 5.5 × 104 Pa. The dipping T. ni larvae in virus‐containing solution efficiently infected them. These results indicate that surface contamination, either by aerosol or dipping, lead to infection via spiracles. The aerosol infection route for AcMNPV was restricted to T. ni and Plutella xylostella larvae, whereas Spodoptera litura and Helicoverpa armigera larvae were resistant to this inoculation process. The yields of the reporter proteins DsRed and EGFP from T. ni larvae following aerosol infection were nearly identical to those following oral feeding or injection. It has been shown that ultrasound (US) stimulation accelerates fracture healing in the animal models and non‐operatively clinical uses. Nitric oxide (NO) is a crucial early mediator in mechanically induced bone formation. Here we found that US‐mediated inducible nitric oxide synthase (iNOS) expression was attenuated by Ras inhibitor (manumycin A), Raf‐1 inhibitor (GW5074), MEK inhibitor (PD98059), NF‐κB inhibitor (PDTC), and IκB protease inhibitor (TPCK). US‐induced Ras activation was inhibited by manumycin A. Raf‐1 phosphorylation at Ser338 by US was inhibited by manumycin A and GW5074. US‐induced MEK and ERK activation was inhibited by manumycin A, GW5074, and PD98059. Stimulation of preosteoblasts with US activated IκB kinase α/β (IKK α/β), IκBαphosphorylation, p65 phosphorylation at Ser276, p65, and p50 translocation from the cytosol to the nucleus, and κB‐luciferase activity. US‐mediated an increase of IKK α/β, IκBα, and p65 phosphorylation, κB‐luciferase activity and p65 and p50 binding to the NF‐κB element was inhibited by manumycin A, GW5074, and PD98059. Our results suggest that US increased iNOS expression in preosteoblasts via the Ras/Raf‐1/MEK/ERK/IKKαβ and NF‐κB signaling pathways. J. Cell. Chondrosarcoma is a malignant primary bone tumor that responds poorly to both chemotherapy and radiation therapy. (-)-Epigallocatechin-3-gallate (EGCG), the major polyphenol in green tea, has been shown to inhibit tumorigenesis and cancer cell growth in animal models. The aim of this study was to elucidate the mechanism of EGCG-induced apoptosis of human chondrosarcoma cells. EGCG induced cell apoptosis in human chondrosarcoma cell lines but not primary chondrocytes. EGCG induced upregulation of Bax and Bak, downregulation of Bcl-2 and Bcl-XL, and dysfunction of mitochondria in chondrosarcoma. We also found that the accumulation of reactive oxygen species (ROS) is a critical mediator in EGCG-induced cell death. EGCG induced apoptosis signal-regulating kinase 1 (ASK1) dephosphorylation and its dissociation from 14-3-3. Treatment of chondrosarcoma cells with EGCG induced p38 and c-jun-NH2-kinase (JNK) phosphorylation. Transfection with ASK1 siRNA or p38 and JNK mutant antagonized the EGCG-induced cell apoptosis. Therefore, EGCG triggered ROS and activated the ASK1-p38/JNK pathway, resulting chondrosarcoma cell death. Importantly, animal studies revealed a dramatic reduction in tumor volume after 24 days of treatment. Thus, EGCG may be a novel anti-cancer agent for the treatment of chondrosarcoma.  相似文献
