Spore-producing organisms have small dispersal units enabling them to become widespread across continents. However, barriers to gene flow and cryptic speciation may exist. The common, haploid peatmoss Sphagnum magellanicum occurs in both the Northern and Southern hemisphere, and is commonly used as a model in studies of peatland ecology and peatmoss physiology. Even though it will likely act as a rich source in functional genomics studies in years to come, surprisingly little is known about levels of genetic variability and structuring in this species. Here, we assess for the first time how genetic variation in S. magellanicum is spatially structured across its full distribution range (Northern Hemisphere and South America). The morphologically similar species S. alaskense was included for comparison. In total, 195 plants were genotyped at 15 microsatellite loci. Sequences from two plastid loci (trnG and trnL) were obtained from 30 samples. Our results show that S. alaskense and almost all plants of S. magellanicum in the northern Pacific area are diploids and share the same gene pool. Haploid plants occur in South America, Europe, eastern North America, western North America, and southern Asia, and five genetically differentiated groups with different distribution ranges were found. Our results indicate that S. magellanicum consists of several distinct genetic groups, seemingly with little or no gene flow among them. Noteworthy, the geographical separation of diploids and haploids is strikingly similar to patterns found within other haploid Sphagnum species spanning the Northern Hemisphere. Our results confirm a genetic division between the Beringian and the Atlantic that seems to be a general pattern in Sphagnum taxa. The pattern of strong genetic population structuring throughout the distribution range of morphologically similar plants need to be considered in future functional genomic studies of S. magellanicum.  相似文献

Summary The ultrastructure of glycerol extracted thrombocyte models was studied before and after incubation with ATP and under the influence of Salyrgan as inhibitor of contraction. The contractile system of the thrombocytes—thrombosthenin—consists of a spatial network of 50 Å wide thrombosthenin A and 100–120 Å wide thrombosthenin M filaments. At rest, the contractile system is arranged in a marginal zone. The ATP-induced contraction led to a concentric condensation of the network which resulted in a central cluster of cell organelles and at a later stage, disruption of the plasma membrane and release of the thrombocyte granules. All these changes which also occur during the formation of the hemostatic plug in vivo are therefore attributed to the reaction of the contractile protein.  相似文献

The Branchiura are fish ectoparasites which include the well-known and very commonly found genus Argulus. But the Branchiura also contains the monotypic genus Dipteropeltis, about which only very little is known. In this investigation of two female paratype specimens we describe for the first time the external morphology of Dipteropeltis based on Scanning Electron Microscopy. We show that Dipteropeltis has a pre-oral spine, unique suction disc support structures, an Argulus-like mouth cone and second maxillae with a combination of features seen in both Argulus and Chonopeltis. These data are compared with data from the three remaining branchiuran genera Dolops, Chonopeltis and Argulus and discussed in a phylogenetic context based on a recently proposed phylogeny of the group.  相似文献

Isolated rat liver microsomes were subjected to enzymatic or non-enzymatic lipid peroxidation in vitro. NADPH-dependent cytochrome c reductase activity was released from the microsomes into the media during peroxidation. This activity could be recovered from the media by DEAE-cellulose chromatography. The recovered enzyme retained high activity for the reduction of cytochrome c and a lower level of activity for the reduction of cytochrome P-450. The active fractions were capable of enzymatically supporting the peroxidation of isolated mitochondria in the presence of organically complexed Fe+3 and NADPH, and in this respect the specific activity was found to be about ten times higher than in microsomes.  相似文献

Summary The spectral sensitivity (wavelength region 330 to 650 nm) of single photoreceptor cells in the compound eye of the butterfly Heliconius numata (Lepidoptera) was studied by intracellular recording. The configuration and amplitude of the receptor potential was similar to that reported for other insect photoreceptor cells. Maximal sensitivity was found in one of the following wavelength regions: 390 to 410 nm, 450 to 470 nm and 530 to 550 nm. Side maxima of variable amplitude were found. The results suggest three photoreceptor types with different spectral sensitivity maxima.  相似文献

Aim Spatial evolutionary and ecological vicariance analysis (SEEVA) is a simple analytical method that evaluates environmental or ecological divergence associated with evolutionary splits. It integrates evolutionary hypotheses, phylogenetic data, and spatial, temporal, environmental and geographical information to elucidate patterns. Using a phylogeny of Prepusa Mart. and Senaea Taub. (Angiospermae: Gentianaceae), SEEVA is used to describe the radiation and ecological patterns of this basal gentian group across south‐eastern Brazil. Location Latin America, global. Methods Environmental data for 151 geolocated botanical collections, associated with specimens from seven species, were compiled with Arc GIS, and were matched with geolocated base layers of eight climatological variables, as well as one each of geological, soil type, elevational and vegetation variables. Sister groups were defined on the basis of the six nested nodes that defined the phylogenetic tree of these two genera. A (0, 1)‐scaled divergence index (D) was defined and tested for each of 12 environmental and for each of the six phylogenetic nodes, by means of contingency analyses. We contrast divergence indices of nested clades, allopatric and sympatric sister clades. Results The level of ecological divergence between sister clades/species, defined in terms of D measures, was substantial for five of six nodes, with 21 of 72 environmental comparisons having D > 0.75. Soil types and geological age of bedrock were strongly divergent only for basal nodes in the phylogeny, by contrast with temperature and precipitation, which exhibited strong divergence at all nodes. There has been strong divergence and progressive occupation of wetter and colder habitats throughout the history of Prepusa. Nodes separating allopatric sister clades exhibited larger niche divergence than did those separating sympatric sister clades. Main conclusions SEEVA provides a multi‐source, direct analysis method for correlating field collections, phylogenetic hypotheses, species distributions and georeferenced environmental data. Using SEEVA, it was possible to quantify and test the divergence between sister lineages, illustrating both niche conservatism and ecological specialization. SEEVA permits elucidation of historical and ecological vicariance for evolutionary lineages, and is amenable to wide application, taxonomically, geographically and ecologically.  相似文献
