Light-stimulated GTP binding to a membrane protein in rod outer segments.

Bovine as well as frog rod outer segments contain a membrane-bound protein which binds the GTP analog GppNp in the light (Kd=0.3μM). The amount of GppNp bound is 2.5–3.5 mmole per mol rhodopsin. The binding protein (M.Wt. ? 54,000) can be extracted from rod membranes with detergent and purified on an Agarose column. The chromatographic profile indicates that the binding protein is distinct from rhodopsin, GTPase or cyclic nucleotide phosphodiesterase.     

Advances in alternative DNA delivery techniques

This review describes recent advances in alternative DNA-delivery techniques with particular emphasis on silicon carbide fibers, intact tissue electroporation, electrophoresis and microinjection. The advantages/disadvantages of each method along with a historical overview and theory of practice are discussed.     

Ionophore-mediated cation translocation in artificial systems. I. A23187-mediated calcium translocation.

The inophore A23187 stimulates the translocation of calcium from an aqueous Hepes buffer into an organic immiscible phase. At saturating calcium concentrations, 2 molecules of ionophore seem to complex each atom of calcium. Consistent with such a stoichiometric behaviour, the apparent ratio of calcium-ionophore association to dissociation rate constants increases as the concentration of ionophore is raised. As a result, at low calcium concentrations, the amount of translocated calcium increases as a power function of A23187 concentration. When allowance is made for such a phenomenon, the relation between calcium translocation and concentration is characterized by usual substrate-receptor binding kinetics.
