Scotopic vision is the result of a cascade of light-dependent biochemical events in rod outer segments (ROS) involving mainly a cGMP-modulation of sodium current. This modification of ionic currents induces changes of membrane potential which generates electroretinographic (ERG) waves. As (i) ERG disturbances are commonly recorded in hypoxic and inflammatory retinal diseases (ii) leukotrienes (LTs), a very potent mediators of inflammation, disturb ionic exchanges in several artificial or natural membrane systems, we undertook the investigation of the effects of LTs on ERG record in mammalian isolated retina. LTB4, LTC4 and LTD4, all induced a dose-dependent marked reduction of the b wave amplitude of ERG. This effect is correlated with a significant decrease in the survival time of the retina. The analysis of the modification of ERG indicates that LTs exhibit a real toxic effect since b wave is mainly affected while P III wave is unchanged. Comparatively with other nervous cells, this phenomenon may be attributed to an increase in Na+ permeability of ROS. It is suggested that LTs may be involved in the development of inflammatory or ischemic retinal diseases.  相似文献

Questions: What was the tree species composition of forests prior to European settlement at the northern hardwood range limit in eastern Québec, Canada? What role did human activities play in the changes in forest composition in this region? Location: Northern range limit of northern hardwoods in the Lower St. Lawrence region of eastern Québec, Canada. Methods: We used early land survey records (1846–1949) of public lands to reconstruct pre‐settlement forest composition. The data consist of ranked tree species enumerations at points or for segments along surveyed lines, with enumerations of forest cover types and notes concerning disturbances. An original procedure was developed to weigh and combine these differing data types (line versus point observations; taxa versus cover enumerations). Change to present‐day forest composition was evaluated by comparing survey records with forest decadal surveys conducted by the government of Québec over the last 30 years (1980–2009). Results: Pre‐settlement dominance of conifers was strong and uniform across the study area, whereas dominance of maple and birches was patchy. Cedar and spruce were less likely to dominate with increasing altitude, whereas maple displayed the reverse trend. Frequency of disturbances, especially logging and fire, increased greatly after 1900. Comparison of survey records and modern plots showed general increases for maple (mentioned frequency increased by 39%), poplar (36%) and paper birch (31%). Considering only taxa ranked first by surveyors, cedar displayed the largest decrease (19%), whereas poplar (15%) and maple (9%) increased significantly. Conclusions: These changes in forest composition can be principally attributed to clear‐cutting and colonization fire disturbances throughout the 20th century, and mostly reflected the propensity of taxa to expand (maples/aspen) or decline (cedar/spruce) with increased disturbance frequency. Québec's land survey archives provide an additional data source to reconstruct and validate our knowledge of North America's pre‐settlement temperate and sub‐boreal forests.  相似文献

We investigated the efficacy of Ocimum basilicum (OB) essential oils for treating depression related behavioral, biochemical and histopathological changes caused by exposure to chronic unpredictable mild stress (CUMS) in mice and to explore the mechanism underlying the pathology. Male albino mice were divided into four groups: controls; CUMS; CUMS plus fluoxetine, the antidepressant administered for pharmacological validation of OB; and CUMS plus OB. Behavioral tests included the forced swim test (FST), elevated plus-maze (EPM) and the open ?eld test (OFT); these tests were performed at the end of the experiment. We assessed serum corticosterone level, protein, gene and immunoexpression of brain-derived neurotropic factor (BDNF) and glucocorticoid receptors (GRs) as well as immunoexpression of glial fibrillary acidic protein (GFAP), Ki67, caspase-3 in the hippocampus. CUMS caused depression in the mice as evidenced by prolonged immobility in the FST, prolonged time spent in the open arms during the EPM test and reduction of open field activity in the OFT. OB ameliorated the CUMS induced depressive status. OB significantly reduced the corticosterone level and up-regulated protein and gene expressions of BDNF and GR. OB reduced CUMS induced hippocampal neuron atrophy and apoptosis, and increased the number of the astrocytes and new nerve cells. OB significantly increased GFAP-positive cells as well as BDNF and GR immunoexpression in the hippocampus.  相似文献

In the floriculture region of Tenancingo in the State of Mexico, the application of stabilized organic matter, such as vermicompost and leachates, contributes to improve the quality of the soil and plant nutrition. However, it is important to know the chemical composition of a vermicompost and the mineralization process. This is because the amount and speed of nutrient release which will be available to the crop will depend on that knowledge. The objective of the study was to evaluate the effect of the application of a vermicompost and leachates on various quantitative variables of Solidago x hybrid, and the mineralization of organic carbon under aerobic incubations. Vermicompost (74 and 36 g/kg soil), leachates (5 and 10 L/kg soil), 0.33 g/kg soil of chemical fertilizer Ca (NO3)2, and not treated soil (control) were applied under greenhouse conditions to evaluate their effects on plant growth variables. Mixtures of 100 g of soil with vermicompost and leachates were made in the laboratory which were incubated during 9 weeks to obtain the potentially mineralizable organic carbon (Corg PM) and the rate of mineralization (k) after adjusting an exponential model. In the greenhouse experiment there were no statistical differences after applying vermicompost and leachates on the quantitative variables (number of stems per plant, diameter of the panicle, fresh weight, plant length and stem diameter) with respect to the control (p>0.05). The effect among the applied doses was evident only for variables such as fresh weight, panicle length and stem diameter with respect to the control. In incubated soils, k values ranged between 0.209-0.325 C mg/kg soil/week. Only with the application of leachates in high doses two pools of organic matter were shown: one soluble labile (102.9 mg/kg soil) and the other hydrolysable (819 mg/kg soil). The soluble, labile fraction favored nutrient availability immediately after its application to the soil. However, a single pool of hydrolysable organic C (987-1074 mg/kg soil) was found when vermicompost was applied. It was associated with a release of organic matter during crop development, and to a possible stimulation of microbial activity. High values of electrical conductivity in vermicompost and leachates (8.2-11.7 mS/m) suggest a moderate application of both products.  相似文献

Extracellular signal-regulated kinase 3 (ERK3) is an atypical member of the mitogen-activated protein kinase (MAPK) family whose function is largely unknown. Given the central role of MAPKs in T cell development, we hypothesized that ERK3 may regulate thymocyte development. Here we have shown that ERK3 deficiency leads to a 50% reduction in CD4+ CD8+ (DP) thymocyte number. Analysis of hematopoietic chimeras revealed that the reduction in DP thymocytes is intrinsic to hematopoietic cells. We found that early thymic progenitors seed the Erk3−/− thymus and can properly differentiate and proliferate to generate DP thymocytes. However, ERK3 deficiency results in a decrease in the DP thymocyte half-life, associated with a higher level of apoptosis. As a consequence, ERK3-deficient DP thymocytes are impaired in their ability to make successful secondary T cell receptor alpha (TCRα) gene rearrangement. Introduction of an already rearranged TCR transgene restores thymic cell number. We further show that knock-in of a catalytically inactive allele of Erk3 fails to rescue the loss of DP thymocytes. Our results uncover a unique role for ERK3, dependent on its kinase activity, during T cell development and show that this atypical MAPK is essential to sustain DP survival during RAG-mediated rearrangements.  相似文献

We investigated the atmospheric concentrations and deposition fluxes of major ions to the Turkey Lakes Watershed (TLW) between 1980 and 1996. During that time, daily SO4 2− concentrations in precipitation decreased markedly, while NO3 , NH4 +, and H+ concentrations remained roughly constant. It appears that precipitation acidity did not decrease in spite of declining SO4 2− concentrations due to a concurrent and counterbalancing decrease in the concentrations of Ca2+, Mg2+, and K+ in precipitation. The reasons for the decline in base cations are unknown, but this decline is probably related to decreasing emissions of soil-derived particles from agricultural, industrial, and road sources. A similar situation was seen during the same period in other parts of Canada, the eastern United States, and Europe. Wet, dry, and total (wet + dry) deposition fluxes of sulphur (S) and nitrogen (N) were estimated annually for the years 1980–96. The 17-year mean annual total (wet + dry) deposition of S to the watershed was estimated at 38.5 mmol m−2 y−1 (range 24.3–50.3). Total S deposition decreased by 35% from the early 1980s (1982–84) to the mid-1990s (1994–96), a decline consistent with the 23% decline in annual SO2 emissions in eastern North America during the same period. In contrast, the annual total (wet + dry) deposition of oxidized N ranged from 39.8 to 60.4 mmol m−2 y−1, with a 15-year mean of 50.1 mmol m−2 y−1 and a net increase of 10% between the early 1980s (1983–85) and the mid-1990s (1994–96). This is in keeping with a 10% increase in NOx emissions in eastern North America during the same period. For both S and N (oxidized), wet deposition dominated over dry deposition as the major mechanism for atmospheric input to the watershed. Annually, wet deposition accounted for approximately two-thirds of the total atmospheric deposition of both S and N. Dry S deposition was due more to gaseous SO2 deposition (two-thirds of dry S deposition) than to particulate SO4 2− deposition (one-third of dry S deposition). Dry deposition of oxidized N, however, was dominated (95%) by gaseous HNO3 deposition, with minimal input from particulate NO3 deposition. Compared to several selected watershed/forest sites in Canada, the United States, and Europe, the estimated total deposition of S and N at the TLW was relatively high during the measurement period. Gene flow has tremendous importance for local adaptation, by influencing the fate of de novo mutations, maintaining standing genetic variation and driving adaptive introgression. Furthermore, structural variation as chromosomal rearrangements may facilitate adaptation despite high gene flow. However, our understanding of the evolutionary mechanisms impending or favouring local adaptation in the presence of gene flow is still limited to a restricted number of study systems. In this study, we examined how demographic history, shared ancestral polymorphism, and gene flow among glacial lineages contribute to local adaptation to sea conditions in a marine fish, the capelin (Mallotus villosus). We first assembled a 490‐Mbp draft genome of M. villosus to map our RAD sequence reads. Then, we used a large data set of genome‐wide single nucleotide polymorphisms (25,904 filtered SNPs) genotyped in 1,310 individuals collected from 31 spawning sites in the northwest Atlantic. We reconstructed the history of divergence among three glacial lineages and showed that they probably diverged from 3.8 to 1.8 million years ago and experienced secondary contacts. Within each lineage, our analyses provided evidence for large Ne and high gene flow among spawning sites. Within the Northwest Atlantic lineage, we detected a polymorphic chromosomal rearrangement leading to the occurrence of three haplogroups. Genotype–environment associations revealed molecular signatures of local adaptation to environmental conditions prevailing at spawning sites. Our study also suggests that both shared polymorphisms among lineages, resulting from standing genetic variation or introgression, and chromosomal rearrangements may contribute to local adaptation in the presence of high gene flow.  相似文献
