2-(Trimethylammonium) ethyl (R)-3-methoxy-3-oxo-2-stearamidopropyl phosphate [(R)-TEMOSPho], a derivative of an organic chemical identified from a natural product library, promotes highly efficient megakaryopoiesis. Here, we show that (R)-TEMOSPho blocks osteoclast maturation from progenitor cells of hematopoietic origin, as well as blocking the resorptive function of mature osteoclasts. The inhibitory effect of (R)-TEMOSPho on osteoclasts was due to a disruption of the actin cytoskeleton, resulting from impaired downstream signaling of c-Fms, a receptor for macrophage-colony stimulating factor linked to c-Cbl, phosphoinositol-3-kinase (PI3K), Vav3, and Rac1. In addition, (R)-TEMOSPho blocked inflammation-induced bone destruction by reducing the numbers of osteoclasts produced in mice. Thus, (R)-TEMOSPho may represent a promising new class of antiresorptive drugs for the treatment of bone loss associated with increased osteoclast maturation and activity. 相似文献

The immature fruits of domesticated Luffa acutangula (L.) Roxb. are a common vegetable in Asia and India. To learn more about
traditional cultivars, accessions were collected from southern Yunnan Province of China, northern Laos, and southeastern Nepal,
and assessed for various parameters of genetic diversity. The size and shape of the fruits and seeds varied considerably.
A form that we found cultivated only in Nepal bore clusters of small fruits that are produced by hermaphrodite flowers. Plants
produced male flowers first, and the first node to bear flowers varied from the second to the twenty-seventh. Twenty-nine
allozyme loci were assayed. Within L. acutangula one allozyme locus was polymorphic. Luffa acutangula andL. aegyptiaca are fixed for different alleles at nine loci, indicating that they are completely reproductively isolated from each other. 相似文献

We investigated the effect of daidzein feeding and estradiol treatment on food intake in cholecystokinin-1 receptor (CCK1R) deficiency, leptin receptor (ObRb) deficiency rats and their wild-type rats. These rats underwent an ovariectomy or a sham operation. For the 5 week experiment, each rat was divided in three groups: control, daidzein (150 mg/kg diet), and estradiol (4.2 μg/rat/day) groups. In both CCK1R+ and CCK1R? rats, daidzein feeding and estradiol treatment significantly decreased food intake. Daidzein feeding significantly reduced food intake in ovariectomized ObRb? rats, although not in ObRb+ rats. Estradiol treatment significantly lowered food intake in ovariectomized ObRb+ and ObRb? rats. In the ovariectomized rats, estradiol treatment significantly increases uterine weight, while daidzein feeding did not change it, suggesting that daidzein might have no or weak estrogenic effect in our experiment. These results suggest that CCK1R and ObRb signalings were not essential for the daidzein- and estradiol-induced anorectic action. 相似文献

A new fused tetracyclic heterocyclic compound, (4bR,10bR)-4b-hydroxy-10b,12-dihydrodibenzo[c,h][2,6]naphthyridine-5,11(4bH,6H)-dione (1), and a known compound, butyl 2-[(benzoyloxy)methyl]benzoate, spatozoate 2, were isolated from the broth culture of Serratia sp. PAMC 25557. The structure of 1 was determined by analyzing spectroscopic data. Compound 1 did not exhibit antimicrobial activity against Escherichia coli, Staphylococcus aureus, or Candida albicans. In addition, up to 100 μg/ml compound 1 did not show any toxicity against Artemia salina larvae. However, compound 1 showed DPPH free radical scavenging activity (IC50 = 16.7 ± 0.34 μg/ml). This was the first report of spatozoate isolation from bacterial sources. 相似文献

Herein, we evaluated the concordance of population inferences and conclusions resulting from the analysis of short mitochondrial fragments (i.e., partial or complete D-Loop nucleotide sequences) versus complete mitogenome sequences for 53 bobwhites representing six ecoregions across TX and OK (USA). Median joining (MJ) haplotype networks demonstrated that analyses performed using small mitochondrial fragments were insufficient for estimating the true (i.e., complete) mitogenome haplotype structure, corresponding levels of divergence, and maternal population history of our samples. Notably, discordant demographic inferences were observed when mismatch distributions of partial (i.e., partial D-Loop) versus complete mitogenome sequences were compared, with the reduction in mitochondrial genomic information content observed to encourage spurious inferences in our samples. A probabilistic approach to variant prediction for the complete bobwhite mitogenomes revealed 344 segregating sites corresponding to 347 total mutations, including 49 putative nonsynonymous single nucleotide variants (SNVs) distributed across 12 protein coding genes. Evidence of gross heteroplasmy was observed for 13 bobwhites, with 10 of the 13 heteroplasmies involving one moderate to high frequency SNV. We report that phytosphingosine, a sphingolipid found in many organisms and implicated in cellular signaling, promotes megakaryocytic differentiation of myeloid leukemia cells. Specifically, phytosphingosine induced several hallmark changes associated with megakaryopoiesis from K562 and HEL cells including cell cycle arrest, cell size increase and polyploidization. We also confirmed that cell type specific markers of megakaryocytes, CD41a and CD42b are induced by phytosphingosine. Phospholipids with highly similar structures were unable to induce similar changes, indicating that the activity of phytosphingosine is highly specific. Although phytosphingosine is known to activate p38 mitogen-activated protein kinase (MAPK)-mediated apoptosis, the signaling mechanisms involved in megakaryopoiesis appear to be distinct. Background
Mainland Tanzania scaled up multiple malaria control interventions between 1999 and 2010. We evaluated whether, and to what extent, reductions in all-cause under-five child mortality (U5CM) tracked with malaria control intensification during this period.Methods
Four nationally representative household surveys permitted trend analysis for malaria intervention coverage, severe anemia (hemoglobin <8 g/dL) prevalence (SAP) among children 6–59 months, and U5CM rates stratified by background characteristics, age, and malaria endemicity. Prevalence of contextual factors (e.g., vaccination, nutrition) likely to influence U5CM were also assessed. Population attributable risk percentage (PAR%) estimates for malaria interventions and contextual factors that changed over time were used to estimate magnitude of impact on U5CM.Results
Household ownership of insecticide-treated nets (ITNs) rose from near zero in 1999 to 64% (95% CI, 61.7–65.2) in 2010. Intermittent preventive treatment of malaria in pregnancy reached 26% (95% CI, 23.6–28.0) by 2010. Sulfadoxine-pyrimethamine replaced chloroquine in 2002 and artemisinin-based combination therapy was introduced in 2007. SAP among children 6–59 months declined 50% between 2005 (11.1%; 95% CI, 10.0–12.3%) and 2010 (5.5%; 95% CI, 4.7–6.4%) and U5CM declined by 45% between baseline (1995–9) and endpoint (2005–9), from 148 to 81 deaths/1000 live births, respectively. Mortality declined 55% among children 1–23 months of age in higher malaria endemicity areas. A large reduction in U5CM was attributable to ITNs (PAR% = 11) with other malaria interventions adding further gains. Multiple contextual factors also contributed to survival gains.Conclusion
Marked declines in U5CM occurred in Tanzania between 1999 and 2010 with high impact from ITNs and ACTs. High-risk children (1–24 months of age in high malaria endemicity) experienced the greatest declines in mortality and SAP. Malaria control should remain a policy priority to sustain and further accelerate progress in child survival. 相似文献8.
Aim The study explores fern species richness patterns along a central Himalayan elevational gradient (100–4800 m a.s.l.) and evaluates factors influencing the spatial increase and decrease of fern richness. Location The Himalayas stretch from west to east by 20°, i.e. 75–95° east, and Nepal is located from 80 to 88° east in this range. Methods We used published data of the distribution of ferns and fern allies to interpolate species elevational ranges. Defining species presence between upper and lower elevation limit is the basis for richness estimates. The richness pattern was regressed against the total number of rainy days, and gradients that are linearly related to elevation, such as length of the growing season, potential evapotranspiration (PET, energy), and a moisture index (MI = PET/mean annual rainfall). The regressions were performed by generalized linear models. Results A unimodal relationship between species richness and elevation was observed, with maximum species richness at 2000 m. Fern richness has a unimodal response along the energy gradients, and a linear response with moisture gradients. Main conclusions The study confirms the importance of moisture on fern distributions as the peak coincides spatially with climatic factors that enhance moisture levels; the maximum number of rainy days and the cloud zone. Energy‐related variables probably control species richness directly at higher elevations but at the lower end the effect is more probably related to moisture. 相似文献

Corridor design is a centripetal conservation tool to facilitate movement between fragmented patches. Increases in anthropogenic activity have caused degradation in forest connectivity, influencing animal movement to a small degree. Laljhadi-Mohana wildlife corridor (LMWC), a corridor between Shuklaphanta National Park (Nepal) and Dudhwa National Park (India) created to be used by Panthera tigris and Elephas maximus in western Nepal, is under pressure of anthropogenic change. Using current knowledge, we analyzed land cover changes (LCC) of LMWC between 2002 and 2012. We used ERDAS IMAGINE 9.2 and Arc GIS 9.2 to process satellite images, and occupancy survey to assess status of corridor. We classified land cover into dense forest, sparse forest, cultivation, water bodies, grassland, expose surfaces, and sand bank as structural attributes of the corridor. Our analysis found dense forest area was reduced by 18.35% in a decade while cultivation and sparse forest increased by 10.15% and 8.89%, respectively. Illegal forest encroachment, resource extraction, grazing pressure, invasive species, and flood were major drivers of forest change. The null occupancy model estimated the highest detection probability of Elephas maximus (0.48 ± 0.08) and the lowest of Axis axis (0.20 ± 0.08). Incorporating site covariates improved occupancy estimates of Sus scrofa (0.82), Axis axis (0.76), Elephas maximus (0.76), Boselaphus tragocamelus (0.66), and Panthera pardus (0.55). Distance to cultivation was the most influential covariate, supported by the expansion of cultivated land in the corridor. LMWC is a functional wildlife corridor despite a decline in forest cover. This decline influenced the number and detection rates of large mammals, instigating crop raiding and conflict. Mitigation measures on LCC drivers, particularly forest encroachment, can improve the functional status of LMWC and raise detection rates of large mammals in future studies. 相似文献

Collection and Use of Wild Edible Fungi in Nepal. The significance of the contribution of wild edible mushrooms to rural livelihoods is acknowledged, but remains largely
unexplored. This study investigates the collection of wild edible Nepalese fungi, which species are used, and what are the
specific characteristics of the collectors and the collection. Data were collected using 282 structured questionnaires, interviews,
and forest walks in 17 districts, environmental household accounts from 413 households in two communities, and a review of
literature. A total of 228 species of wild fungi are confirmed to be used for food, and collection is most widespread among
the high-mountain Tibeto-Nepalese ethnic groups, which collect an average of 18.1 kilograms (kg) fresh mushrooms per year
per household, with some households collecting as much as 160 kg. We found no relation between wealth status and subsistence
collection, although the commercial collectors are predominantly the poor earning up to six months of unskilled labor salary
per season. Given the limited local trade and occurrence of globally marketed species, the scope for increasing contributions
to rural livelihoods from collection of mushrooms, and risks in relation to this are briefly discussed. 相似文献
