WNT signaling was discovered in tumor models and has been recognized as a regulator of cancer development and progression for over 3 decades. Recent work has highlighted a critical role for WNT signaling in the metabolic homeostasis of mammals, where its misregulation has been heavily implicated in diabetes. While the majority of WNT metabolism research has focused on nontransformed tissues, the role of WNT in cancer metabolism remains underinvestigated. Cancer is also a metabolic disease where oncogenic signaling pathways regulate energy production and macromolecular synthesis to fuel rapidly proliferating tumors. This review highlights the emerging evidence for WNT signaling in the reprogramming of cancer cell metabolism and examines the role of these signaling pathways as mediators of tumor bioenergetics.  相似文献

A fast, simple, and cost-effective HPLC method for the quantitation of the antiviral drug ganciclovir is described. The serum samples are extracted with perchloric acid and neutralized with potassium phosphate buffer, and urine samples are diluted with distilled water. A reversed-phase column with isocratic elution by 15 mM potassium phosphate buffer (pH 2.5) containing 0.25% acetonitrile is used to separate ganciclovir; quantitation is by UV absorbance at 254 nm. Total turnaround time is 22 min; more than 3000 samples can be run on a single column without loss of peak quality. The limit of quantitation is 0.05 μg/ml. Recoveries varied from 91 to 10% with coefficients of variation ranging from 0.387 to 7.95%.  相似文献

A radioimmunoassay for erythropoietin has been developed using 125I labeled pure human erythropoietin and an anti-erythropoietin antiserum produced in a rabbit immunized with human erythropoietin. Two techniques are presented for labeling erythropoietin, both resulting in an immunologically reactive labeled reactant. One method involves the use of lactoperoxidase and the other a reagent known as IODO-GEN. The second International Reference Preparation of human erythropoietin is used as a standard and a double antibody scheme is used for the separation of the free and antibody bound labeled hormone. The radioimmunoassay is sensitive to an absolute amount of erythropoietin equivalent to 0.4 milliunits. Bioassays for erythropoietin require approximately 100 times this amount. The use of pure erythropoietin as the labeled reactant has removed certain discrepancies seen in previous attempts to develop radioimmunoassays for this hormone, e.g., sera from patients without kidneys do not give the high values previously seen. Sera from anemic individuals not only give rise to high radioimmunoassay values but also show a parallel relationship with the erythropoietin standard when halving dilutions are analyzed. Desialated erythropoietin is also reactive with the same parallelism. Bleeding of a normal individual increases the serum erythropoietin level and transfusion decreases it. Erythropoietin from a variety of laboratory animals is also reactive in the radioimmunoassay, with very high values being observed in hypoxic animals.  相似文献

ABSTRACT

Free-roaming cats present a multidisciplinary challenge for wildlife conservation and feline welfare. Veterinary professionals are critical stake-holders in the issue of free-roaming cats, making it imperative that their attitudes and perspectives are well understood. In this study, we conducted an electronic survey of veterinarians practicing across British Columbia to evaluate their perceptions of 1) whether cat overpopulation is a concern, 2) the impacts of cats on wildlife, and 3) alternative management strategies for dealing with cat overpopulation. Of the 389 respondents, 56% agreed that cat overpopulation was a local problem, and 91% agreed that veterinarians play a role in reducing cat overpopulation. Veterinarians in rural communities had a greater involvement in cat overpopulation and perceived it as more problematic than their urban colleagues. The majority (70%) of veterinarians reported that they performed early-age sterilization, supported the principle of low-cost spay-neuter services (67%), and 65% of them were involved in these services. Just over half (54%) of respondents reported that they provide permanent identification to cats, typically offering both microchips and tattoos. Veterinarians were less cohesive on their perceptions of the impact of free-roaming cats on wildlife: 53% agreed that after habitat loss cats are one of the most significant, direct causes of bird and wildlife mortality, while 30% were neutral and 17% disagreed. Fifty-six percent of veterinarians felt that keeping cats indoors would be one of the most effective strategies to reduce cat impacts on wildlife, but they also felt that public education programs and low-cost spay-neuter programs would be effective and more popular with cat owners. Most veterinarians (79%) recommended against unsupervised outdoor access, citing safety and disease risks. Rodent control was the most common reason for not recommending an indoor lifestyle. Overall, our survey results suggest that veterinarians are committed participants in the prevention of cat overpopulation. Conservationists need to focus on increased engagement with veterinarians regarding the impact of cats on wildlife.  相似文献

Research examining the electromyographic (EMG) burst structure of rapid discrete limb movements has led to discordant findings concerning agonist burst duration. Some research has shown that duration varies as a function of movement speed while other research has shown burst constancy. Unfortunately, much of this research may be confounded by not carefully controlling movement termination accuracy and movement time (MT). Due to these potential problems, the present study was conducted to determine the effects of strict spatiotemporal constraints on EMG characteristics of a rapid elbow flexion-extension response under two movement extent conditions across five different MTs. Results revealed that a decreased MT was accompanied by a decreased agonist (biceps) burst duration and increased agonist burst amplitude. The burst duration and amplitude both increased as the movement extent increased with MT held constant. None of three current theoretical perspectives of rapid movement control (the impulse-timing model, the speed-control system hypothesis, or the speed-sensitive strategy) could fully account for these results. Instead, a control strategy was exhibited in which moving faster was accomplished by relative scaling of burst area via concomitant expansion of burst amplitude and compression of burst duration.  相似文献

A physical map of the 952kbp chromosome of Borrelia burgdorferi Sh-2-82 has been constructed. Eighty-three intervals on the chromosome, defined by the cleavage sites of 15 restriction enzymes, are delineated. The intervals vary in size from 96kbp to a few hundred bp, with an average size of 11.5 kbp. A striking feature of the map is its linearity; no other bacterial groups are known to have linear chromosomes. The two ends of the chromosome do not hybridize with one another, indicating that there are no large common terminal regions. The chromosome of this strain was found to be stable in culture; passage 6, 165 and 320 cultures have identical chromosomal restriction maps. We have positioned all previously known Borrelia burgdorferi chromosomal genes and several newly identified ones on this map. These include the gyrA/gyrB/dnaA/dnaN gene cluster, the rRNA gene cluster, fla, flgE, groEL (hsp60), recA, the rho/hip cluster, the dnaK (hsp70)/dnaJ/grpE cluster, the pheT/pheS cluster, and the genes which encode the potent immunogen proteins p22A, p39 and p83. Our electrophoretic analysis detects five linear and at least two circular plasmids in B. burgdorferi Sh-2-82. We have constructed a physical map of the 53 kbp linear plasmid and located the operon that encodes the two major outer surface proteins ospA and ospB on this plasmid. Because of the absence of functional genetic tools for this organism, these maps will serve as a basis for future mapping, cloning and sequencing studies of B. burgdorferi.  相似文献

Since nasopharyngeal carriage of pneumococcus precedes invasive pneumococcal disease, characteristics of carriage isolates could be incorrectly assumed to reflect those of invasive isolates. While most pneumococci express a capsular polysaccharide, nontypeable pneumococci are sometimes isolated. Carriage nontypeables tend to encode novel surface proteins in place of a capsular polysaccharide synthetic locus, the cps locus. In contrast, capsular polysaccharide is believed to be indispensable for invasive pneumococcal disease, and nontypeables from population-based invasive pneumococcal disease surveillance have not been extensively characterized. We received 14,328 invasive pneumococcal isolates through the Active Bacterial Core surveillance program during 2006–2009. Isolates that were nontypeable by Quellung serotyping were characterized by PCR serotyping, sequence analyses of the cps locus, and multilocus sequence typing. Eighty-eight isolates were Quellung-nontypeable (0.61%). Of these, 79 (89.8%) contained cps loci. Twenty-two nontypeables exhibited serotype 8 cps loci with defects, primarily within wchA. Six of the remaining nine isolates contained previously-described aliB homologs in place of cps loci. Multilocus sequence typing revealed that most nontypeables that lacked capsular biosynthetic genes were related to established non-encapsulated lineages. Thus, invasive pneumococcal disease caused by nontypeable pneumococcus remains rare in the United States, and while carriage nontypeables lacking cps loci are frequently isolated, such nontypeable are extremely rare in invasive pneumococcal disease. Most invasive nontypeable pneumococci possess defective cps locus genes, with an over-representation of defective serotype 8 cps variants.  相似文献
